様々な素材上における液状組成物の殺菌又は静菌作用を調べた。
試験例1と同様の手順で、液状組成物(カプリル酸モノグリセリド 0.15%、フィチン酸 0.40%、エタノール 40.00%、水 残余;単位は質量%;pH2.00)、及びサンプル菌液を調製した。
対象素材として、金属(ステンレス板)、ゴム(ゴム手袋)、プラスチック(番重ふた)を準備した。ステンレス板及び番重ふたは、10cm四方に切断して使用した。ゴム手袋は、10cm四方のプラスチック板に皺のないように伸ばして両面テープで貼り、余った部分はカットして使用した。

菌数の測定は、各素材につき以下の手順で行った。各対象素材を3群に分けた。第一の群では、液状組成物0.5mLをコンラージ棒で対象素材の上に均一に塗布し、次いでサンプル菌液0.1mLを接種し、40秒間コンラージ棒で混合しながら塗り広げた(0h群)。残りの2群では、液状組成物0.5mL又は1.0mLを、コンラージ棒で対象素材の上に均一に塗布した後、安全キャビネット内で18時間風乾し、水分とエタノールを完全に蒸発させた。これにサンプル菌液1.0mLを接種し、40秒間コンラージ棒で混合しながら塗り広げた(18h群)。サンプル初発菌数は105~106cfu/mLに調整した。
サンプル菌液を塗布した対象素材を5分間静置後、無菌パウチに入れ、上から滅菌水10mLを加え、熱シールして上下に5回程度軽く振った。パウチを開封して中の液体を採取し、MRS寒天培地で菌数を測定し、試験例1と同様の基準で殺菌又は静菌特性を評価した。

The target material to which the sample bacterial solution was applied was allowed to stand for 5 minutes, then placed in a sterile pouch, added with 10 mL of sterilized water from above, heat-sealed, and gently shaken up and down about 5 times. The pouch was opened, the liquid inside was collected, the number of bacteria was measured with MRS agar medium, and bactericidal or bacteriostatic properties were evaluated according to the same criteria as in Test Example 1.
