(4)支持体の少なくとも片面にドネペジルとアクリル系粘着剤とを含む粘着剤層を有するドネペジル含有貼付製剤が透湿防止袋中に封入され、且つ、該粘着剤層の含水率が1000ppm以上にコントロールされた、包装体。
(5)前記粘着剤層の含水率が、8000ppm以下にコントロールされた、上記(4)に記載の包装体。
(6)前記粘着剤層の含水率が、7000ppm以下にコントロールされた、上記(4)に記載の包装体。
(7)前記透湿防止袋中に、前記貼付製剤および乾燥剤が封入された、上記(4)~(6)のいずれか1項に記載の包装体。
(8)支持体の少なくとも片面にドネペジルとアクリル系粘着剤とを含む粘着剤層を有するドネペジル含有貼付製剤が透湿防止袋中に封入され、該粘着剤層の含水率が1000ppm以上である、包装体。
(9)支持体の少なくとも片面にドネペジルとアクリル系粘着剤とを含む粘着剤層を有するドネペジル含有貼付製剤と乾燥剤とが透湿防止袋中に封入され、該粘着剤層の含水率が1000ppm以上である、包装体。
(10)前記粘着剤層の含水率が、8000ppm以下である、上記(8)または(9)に記載の包装体。
(11)前記粘着剤層の含水率が、7000ppm以下である、上記(8)または(9)に記載の包装体
(13)支持体の少なくとも片面にドネペジルとアクリル系粘着剤とを含む粘着剤層を有するドネペジル含有貼付製剤を、該粘着剤層の含水率が8000ppm以下となるようにコントロールして、該粘着剤層からのドネペジルの放出性の劣化を抑制する方法。
(14)支持体の少なくとも片面にドネペジルとアクリル系粘着剤とを含む粘着剤層を有するドネペジル含有貼付製剤を、該粘着剤層の含水率が7000ppm以下となるようにコントロールして、該粘着剤層からのドネペジルの経時的な放出性の劣化を抑制する方法。
本実施形態のドネペジル含有貼付製剤は、支持体と、支持体の少なくとも片面に形成された粘着剤層とを有する。
支持体は、粘着剤層を安定して支持可能なものであれば、その素材や形状は特に限定されないが、粘着剤層中のドネペジルを透過させてその配合割合を低下させないもの(すなわち、ドネペジルに対して不透過性を有する材料)が好ましい。
粘着剤層は、少なくとも、ドネペジルおよびアクリル系粘着剤を含むものである。この粘着剤層は、上述した支持体の少なくとも片面に設けられていればよく、例えば、支持体の両面に設けられていても構わない。
ドネペジル含有貼付製剤は、粘着剤層の粘着面を使用直前まで保護する観点から、粘着剤層上に剥離ライナーを有することが好ましい。すなわち、上記のドネペジル含有貼付製剤は、支持体の少なくとも片面に粘着剤層が積層され、且つ、その粘着剤層の粘着面(支持体に積層した面と反対の面)上に剥離ライナーが積層された構成であることが好ましい。
ドネペジル含有貼付製剤の製造方法は、特に限定されないが、例えば、少なくともドネペジルとアクリル系粘着剤とを含む調合物を調製し、この調合物を支持体上に塗布して成膜する方法が簡便である。
本実施形態のドネペジル含有貼付製剤は、抗アルツハイマー型認知症薬として用いることができる。また、その他の用途としては、抗脳血管性認知症、片頭痛予防などが挙げられるが、これらに特に限定されない。
本実施形態の包装体は、上記のドネペジル含有貼付製剤を透湿防止袋中に封入することにより粘着剤層の含水率が約1000ppm以上,好ましくは約1000~8000ppm,より好ましくは約1000~7000ppmにコントロールされたものである。
<ドネペジルの放出率>
日本薬局方一般試験法 溶出試験法第2法(パドル法)に基づき、下記の条件下で、ドネペジルの放出率(%;試験片に含まれるドネペジル総質量に対する、溶出したドネペジルの質量割合)を測定した。
試験液 :pH4.0 酢酸・酢酸ナトリウム緩衝液
(0.05mol/L)
(酢酸ナトリウム三水和物54.44gを蒸留水3000m
Lに溶解させ,さらに酢酸を加えてpH3.95~4.0
4とした後、蒸留水で全量を8000mLとした。)
試験液量 :900mL
温度 :32℃
パドル回転数:50rpm
溶出時間 :4時間
HPLC条件:カラム Inertsil(登録商標) ODS-2
(GLサイエンス社製)
(4.6mmI.D.×15cm、5μm)
移動相 :10mM 1-デカンスルホン酸ナトリウム水溶液/アセ
トニトリル/70%過塩素酸=650/350/1(体積
比)
(1-デカンスルホン酸ナトリウム濃度は移動相全体に対
して10mMとした。)
カラム温度 :35℃
検出波長 :UV(271nm)
試料注入量 :50μL
流速 :1.4mL/min
保持時間 :11.0分
未包装のものについては、製造(調温調湿)直後のドネペジル含有貼付製剤を速やかに温度23±2℃および相対湿度40±5%RHに制御された環境下に置き、7.5cm2に打ち抜き加工して、試験片を作製した。その後、試験片から剥離ライナーを除去して水分気化装置へ投入した。なお、上記の処理は、製造(調温調湿)後から1分以内に完了させた。
まず、不活性ガス雰囲気下、アクリル酸2-エチルへキシル75部、N-ビニル-2-ピロリドン22部、アクリル酸3部およびアゾビスイソブチロニトリル0.2部を、酢酸エチル中60℃にて溶液重合させてアクリル系粘着剤の酢酸エチル溶液(粘着剤固形分:28%)を得た。次に、このアクリル系粘着剤とドネペジルとを下記の配合組成で混合し、酢酸エチルで粘度調整して塗工液を得た。
ドネペジル(フリー塩基) 7.57質量%
アクリル系粘着剤 40.53質量%
ミリスチン酸イソプロピル 50.00質量%
塩化ナトリウム 1.16質量%
水酸化ナトリウム 0.01質量%
メタ重亜硫酸ナトリウム 0.50質量%
アスコルビン酸 0.05質量%
エチルアセトアセテートアルミニウムジイソプロピレート0.18質量%
(合計 100質量%)
上記の塗工原反を、23℃30%RHに制御された環境下に3時間以上放置した後、40cm2に打ち抜いて実施例1のドネペジル含有貼付製剤を製造した。また、23℃75%RHで制御された環境下に放置すること以外は、実施例1と同様にして、比較例1のドネペジル含有貼付製剤を製造した。得られた実施例1および比較例1のドネペジル含有貼付製剤について、製造直後の粘着剤層の含水率およびドネペジルの放出率を測定した。表1に、結果を示す。
実施例1のドネペジル含有貼付製剤を、40℃75%RHの恒温器で一定時間加湿することにより、実施例2~5および比較例2~5のドネペジル含有貼付製剤を製造した。得られた実施例2~5および比較例2~5のドネペジル含有貼付製剤について、製造直後の粘着剤層の含水率および粘着剤層のドネペジルの放出率を測定した。表2に、結果を示す。
PETフィルム(厚み12μm)/アルミニウム箔(厚み7μm)/ポリアクリロニトリル系樹脂フィルム(厚み30μm)の3層積層フィルムを用い、ポリアクリロニトリル系樹脂フィルム層を内側にして、上記の実施例2~5および比較例2~5のドネペジル含有貼付製剤を封入(包装)し、さらに長期保管(40℃、2ヶ月間)して、透湿防止袋中にドネペジル含有貼付製剤が封入された、実施例6~9および比較例6~9の包装体を製造した。その後、得られた実施例6~9および比較例6~9の包装体を開封してドネペジル含有貼付製剤を取り出し、長期保管後の粘着剤層の含水率および粘着剤層のドネペジルの放出率を測定した。表3に、結果を示す。
上記の塗工原反を、実施例1と同様に処理して、粘着剤層の含水率が3195ppmである参考例1のドネペジル貼付製剤を製造した。
次に、実施例6と同じ3層積層フィルムを用い、参考例1のドネペジル貼付製剤と乾燥剤である硫酸マグネシウム5gとを同様に封入(包装)し、さらに長期保管(40℃、1ヶ月間)して、透湿防止袋中にドネペジル含有貼付製剤が封入された、実施例10の包装体を製造した。その後、得られた実施例10の包装体を開封してドネペジル含有貼付製剤を取り出し、長期保管後の粘着剤層の含水率を測定したところ、2580ppmであった。
次いで、乾燥剤を使用しないこと以外は実施例10と同様にして、実施例11の包装体を製造した。実施例10と同様に長期保管後の粘着剤層の含水率を測定したところ、3333ppmであった。
さらに、乾燥剤としてゼオライト5gを使用すること以外は実施例10と同様にして、比較例10の包装体を製造した。実施例10と同様に長期保管後の粘着剤層の含水率を測定したところ、415ppmであった。

- 支持体の少なくとも片面にドネペジルとアクリル系粘着剤とを含む粘着剤層を有し、該粘着剤層の含水率が1000ppm以上である、

請求項1に記載のドネペジル含有貼付製剤。 - 前記粘着剤層の含水率が、7000ppm以下である、
