本發明係提供一種硫化氫緩釋敷料及其製造方法。本發明之硫化氫緩釋敷料包含一水膠體、一介面活性劑以及一硫氫化鈉。
Description
本發明係有關於一種硫化氫緩釋敷料及其製造方法,尤其是關於一種可以緩慢釋放硫化氫氣體以促進慢性傷口癒合的硫化氫緩釋敷料及其製造方法。
硫化氫(H2S)是繼一氧化氮以及一氧化碳後,第三個被發現的內生性氣相傳遞物質。在動物實驗中已被證明了硫化氫氣體具有舒張血管、抑制血管平滑肌細胞增殖、誘導血管平滑肌細胞凋亡、促進微血管內皮細胞增殖、抗發炎反應以及抗氧化作用等功效。在現有技術中已知細胞會以緩慢的速率製造及低濃度的硫化氫氣體,然而外部之硫化氫供體(donor)並無法真實模擬硫化氫在人體中的生成速率。
硫氫化鈉(NaHS)因溶於水後可以產生硫化氫氣體,故可做為硫化氫之供體。然而,因硫氫化鈉中的硫氫根離子(HS-)與水中的氫離子(H+)結合放出硫化氫的反應過程非常迅速,故會在短時間內釋放出高濃度的硫化氫。高濃度的硫化氫對細胞有毒性,會引發細胞凋亡或發炎等不良反應。低濃度
之硫化氫雖然有助於傷口癒合的功效,但因高濃度之硫化氫有毒性,因此限制了硫化氫在醫療上的應用。
因此,仍需要一種可以緩慢釋放硫化氫且不會產生細胞毒性以促進傷口癒合的敷料。
本發明提供一種新穎的硫化氫緩釋敷料及其製造方法。本發明之硫化氫緩釋敷料可緩慢的釋放硫化氫而不會產生細胞毒性,故可有效促進慢性傷口的癒合。
本發明之一態樣為提供一種硫化氫緩釋敷料,其包含一水膠體、一介面活性劑以及一硫氫化鈉。
在本發明之一實施例中,前述水膠體可為60至140重量份,前述介面活性劑可為0.2至2重量份,且前述硫氫化鈉可為0.1至0.5重量份。
在本發明之一實施例中,前述介面活性劑可為聚山梨醇酯80、聚山梨醇酯20、聚山梨醇酯60或聚山梨醇酯40。
在本發明之一實施例中,前述水膠體包含一彈性體、一親水性高分子、一增黏劑以及一延展劑。
在本發明之一實施例中,前述彈性體可為10至30重量份,前述親水性高分子可為20至60重量份,前述增黏劑可為20至60重量份,且前述延展劑可為2至20重量份。
在本發明之一實施例中,前述彈性體可以是苯乙烯-異戊二烯-苯乙烯(SIS)共聚物、苯乙烯-丁二烯-苯乙烯(SBS)共聚物、苯乙烯-(乙烯-丁烯)-苯乙烯(SEBS)共聚物、苯
乙烯-(乙烯-丙烯)-苯乙烯(SEPS)或其組合。
在本發明之一實施例中,前述親水性高分子可以是羧甲基纖維素鈉、羥乙基纖維素、海藻酸鈉、明膠、果膠、羧甲基殼聚醣、瓜爾豆膠、刺槐豆膠、膠原蛋白、刺梧桐樹膠或其組合。
在本發明之一實施例中,前述增黏劑係選自由松香樹脂、萜烯樹脂、C5石油樹脂、C9石油樹脂、高純度二環戊二烯(H-DCPD)或其組合。
在本發明之一實施例中,前述延展劑可以是礦物油、液體石蠟、蓖麻油、鄰苯二甲酸二丁酯、羊毛脂、環烷油或其組合。
在本發明之另一實施例中,前述水膠體可更包含0.2至2重量份之抗氧化劑。
在本發明之另一實施例中,前述抗氧化劑可為受阻酚、硫增效劑(thiosynergists)、二級芳香胺(secondary aromatic amines)或亞磷酸酯(phosphites)。
本發明之另一態樣為提供一種硫化氫緩釋敷料之製造方法,其步驟包含:(a)加熱攪拌一水膠體材料;(b)將介面活性劑以及硫氫化鈉加入前述水膠體材料中;以及(c)將含有前述介面活性劑以及前述硫氫化鈉之水膠體材料注入成型模具中熱壓成型以形成一硫化氫緩釋敷料。
在本發明之製造方法之一實施例中,前述水膠體材料可為60至140重量份,前述介面活性劑可為0.2至2重量份,且前述硫氫化鈉可為0.1至0.5重量份。
在本發明之製造方法之一實施例中,在前述步驟(a)中之加熱溫度可介於100℃至200℃之間。
在本發明之製造方法之一實施例中,在前述步驟(a)中之加熱時間係介於1小時至1.5小時之間。
在本發明之製造方法之一實施例中,前述步驟(b)中可更包含進行一加熱處理,且加熱溫度可介於100℃至150℃之間。
在本發明之製造方法之一實施例中,前述步驟(b)中之加熱時間可介於15分鐘至45分鐘之間。

在本發明之硫化氫緩釋敷料之一實施例中,水膠體可為60至140重量份,介面活性劑可為0.2至2重量份,且硫氫化鈉可為0.1至0.5重量份。當硫氫化鈉之添加量過多時可能使所釋放出之硫化氫氣體濃度過快而造成細胞毒性。當硫氫化鈉之添加量過少時則可能因所釋放出之硫化氫氣體濃度過低而無法有效促進傷口癒合。
適合之水膠體可為習知的可做為敷料的水膠體(Hydrocolloid)組合物。水膠體具有吸收傷口滲液的能力。吸收滲液後,水膠體中的親水性材料可形成類似凝膠的半固體物質,附著於傷口基部,提供並維持有利於創面癒合的濕性環境。此外,因水膠體具有黏性,其可形成密閉(occlusive)創面,以促進微血管的增生和肉芽組織的形成,從而加速傷口癒合。因水膠體敷料可提供密閉環境,有利於巨噬細胞清除壞死組織,故水膠體亦具有清創功能。在本發明之一實施例中,水膠體可包含一彈性體、一親水性高分子、一增黏劑以及一延展劑。
彈性體的作用主要是成型和提供黏性及柔軟性等。因彈性體為疏水結構,藉由彈性體包覆硫氫化鈉,可避免硫氫化鈉直接與水接觸,故可減緩並調控硫化氫的釋放速率。適合之彈性體可以例如是苯乙烯-異戊二烯-苯乙烯(SIS)共聚物、苯乙烯-丁二烯-苯乙烯(SBS)共聚物、苯乙烯-(乙烯-丁烯)-
苯乙烯(SEBS)共聚物、苯乙烯-(乙烯-丙烯)-苯乙烯(SEPS)共聚物或其組合,但不限於此。
親水性高分子具有吸收液體的能力,且因具有交聯結構而在吸收滲液後不會溶解而是發生溶脹,故可用以提供吸水性。可使用的親水性高分子可包括天然、半合成或合成的親水性高分子。天然的親水性高分子可例如包括多糖類高分子如果膠、阿拉伯膠、瓜爾膠、瓊脂、澱粉、黃原膠、葡聚糖等;和蛋白質或多肽類高分子如明膠、白蛋白、酪蛋白等。半合成的親水性高分子可例如包括羧甲基纖維素、羧甲基纖維素鈉、甲基纖維素、乙基纖維素、羥乙基纖維素、羥丙基纖維素、甲基羥丙基纖維素、海藻酸鈉、羧甲基澱粉等。合成的親水性高分子可例如包括丙烯酸類聚合物(如聚丙烯酸和聚丙烯醯胺)、聚乙烯醇、聚乙烯基吡咯烷酮、聚乙二醇、聚乙烯基甲基醚等。在本發明之一較佳實施例中,親水性高分子可以是羧甲基纖維素鈉、羥乙基纖維素、海藻酸鈉、明膠、果膠、羧甲基殼聚醣、瓜爾豆膠、刺槐豆膠、膠原蛋白以及刺梧桐樹膠之至少之一或其組合,但不限於此。當親水性高分子之添加量過多時可能使水膠體之吸水速率過高因而使硫化氫氣體之釋放速度過快而造成細胞毒性。當親水性高分子之添加量過少時則可能因水膠體之吸水速率過慢而使硫化氫氣體之釋放速度過低而無法有效促進傷口癒合。
增黏劑可用以進一步調節水膠體的黏性。適合之增黏劑可以是松香樹脂、萜烯樹脂、C5石油樹脂、C9石油樹脂以及高純度二環戊二烯(H-DCPD)之至少之一或其組合。
延展劑可提供潤濕作用及/或黏度控制。適合之延展劑可以是礦物油、液體石蠟、蓖麻油、鄰苯二甲酸二丁酯、羊毛脂以及環烷油之至少之一或其組合。
在本發明之一實施例中,水膠體之彈性體可為10至30重量份,親水性高分子可為20至60重量份,增黏劑可為20至60重量份,且延展劑可為2至20重量份。
在本發明之另一實施例中,水膠體可選擇性地更包含0.2至2重量份之抗氧化劑,以避免彈性體老化造成特性改變。適合之抗氧化劑可為受阻酚、硫增效劑(thiosynergists)、亞磷酸酯(phosphites)或二級芳香胺(secondary aromatic amines)。
親水性高分子分散於彈性體中,藉由添加介面活性劑,其親水端可使水膠體表面遇水後浸潤,使親水性高分子得以澎潤及吸收滲液,並促使水分子滲透進疏水性的彈性體中。故,藉由調整親水性高分子以及介面活性劑之比例,可使水膠體具有不同的吸水速率,並進而使包覆於水膠體中的硫氫化鈉隨吸水速率而有不同的硫化氫釋放速率。在本發明之一實施例中,介面活性劑可為聚山梨醇酯80、聚山梨醇酯20、聚山梨醇酯60或聚山梨醇酯40。當介面活性劑之添加量過多時可能會使水膠體之吸液速度過快而使硫化氫之釋放濃度過高。當介面活性劑之添加量過少時則可能會使水膠體之吸液速度過慢而影響硫化氫之釋放速度。
本發明之另一態樣為提供一種硫化氫緩釋敷料之製造方法,其步驟可包含但不限於下述步驟。
首先,在步驟(a)中,加熱攪拌一水膠體材料。在本發明之製造方法之一實施例中,水膠體材料可為60至140重量份,水膠體材料可例如前述包含一彈性體、一親水性高分子、一增黏劑以及一延展劑。在本發明之製造方法之一較佳實施例中,在步驟(a)中之加熱溫度可介於100℃至200℃之間,加熱時間可介於1小時至1.5小時之間。
接著,在步驟(b)中,將介面活性劑以及硫氫化鈉加入水膠體材料中。介面活性劑之添加量可為0.2至2重量份,且硫氫化鈉之添加量可為0.1至0.5重量份。在本發明之製造方法之一較佳實施例中,在步驟(b)中可更包含進行一加熱處理,加熱溫度可介於100℃至150℃之間,加熱時間可介於15分鐘至45分鐘之間。
最後,在步驟(c)中,將含有介面活性劑以及硫氫化鈉之水膠體材料注入成型模具中熱壓成型以形成一硫化氫緩釋敷料。熱壓溫度可例如介於50℃至100℃之間。
下述實施例係用來進一步說明本發明,但本發明並不受其限制。
實施例
實施例1
取15.4克的苯乙烯-異戊二烯-苯乙烯(SIS)共聚物(Kraton D1161,購自Kraton,美國)、36.55克的C9改質樹脂(Wingtack 86,購自CRAY VALLEY,美國)、6.4克的礦物油(Kaydol white mineral oil購自Sonneborn,美國)以及1克的受阻酚四(3,5-二叔丁基-4-羥基)苯丙酸季戊四醇酯(Chinox
1010,購自雙鍵化工,台灣)於180℃在氮氣環境下攪拌60分鐘,接著降溫至120℃後,加入40克的羧甲基纖維素鈉、0.5克的介面活性劑Tween 80以及0.15克的硫氫化鈉,攪拌10分鐘後倒入模具中在90℃下熱壓成型,形成一硫化氫緩釋敷料。
實施例2
實施例2的步驟及材料同於實施例1,除了使用36.5克的C9改質樹脂取代36.55克的C9改質樹脂以及以0.2克的硫氫化鈉取代0.15克的硫氫化鈉。
實施例3
實施例3的步驟及材料同於實施例1,除了使用15.3克的苯乙烯-異戊二烯-苯乙烯(SIS)共聚物取代15.4克的苯乙烯-異戊二烯-苯乙烯(SIS)共聚物、以36.3克的C9改質樹脂取代36.55克的C9改質樹脂以及以0.5克的硫氫化鈉取代0.15克的硫氫化鈉。
實施例4
實施例4的步驟及材料同於實施例1,除了使用17.93克的苯乙烯-異戊二烯-苯乙烯(SIS)共聚物取代15.4克的苯乙烯-異戊二烯-苯乙烯(SIS)共聚物、以42.59克的C9改質樹脂取代36.55克的C9改質樹脂、以30克的羧甲基纖維素鈉取代40克的羧甲基纖維素鈉以及以0.5克的硫氫化鈉取代0.15克的硫氫化鈉。
比較例1
比較例1的步驟及材料同於實施例1,除了使用15.2克的苯乙烯-異戊二烯-苯乙烯(SIS)共聚物取代15.4克的
苯乙烯-異戊二烯-苯乙烯(SIS)共聚物、以36克的C9改質樹脂取代36.55克的C9改質樹脂以及以1克的硫氫化鈉取代0.15克的硫氫化鈉。
以亞甲基藍法量測實施例1-4以及比較例1之硫化氫緩釋敷料之硫化氫釋放速度。
首先,配製已知濃度之標準品,製作檢量線。檢量線製作步驟如下,先配置含有0.04mg/mL硫氫化鈉之500μL磷酸緩衝溶液(pH=7.4),並序列稀釋為各濃度之標準品,並準備不含有硫氫化鈉之磷酸緩衝溶液。分別取各標準品100μL和100μL的1.0wt%醋酸鋅水溶液混合後,再加入20μL的含有濃度為20mM之N,N-二甲基-1,4-苯二胺硫酸鹽(N,N-dimethyl-p-phenylenediamine sulfate)以及濃度為7.2N之氯化氫水溶液,與20μL的含有濃度為30mM之氯化鐵(FeCl3)以及濃度為1.2N之氯化氫水溶液後,反應10分鐘。因氯化鐵作為氧化劑,可催化N,N-二甲基-1,4-苯二胺硫酸鹽與硫氫化鈉溶於水後的產物硫化氫反應生成亞甲基藍,因此測量各標準品之亞甲基藍在波長670nm下的吸收強度,可建立檢量線,並可依此檢量線回推硫化氫濃度。接著,將實施例1-4以及比較例1之敷料置入1500μL之磷酸緩衝溶液中,分別靜置2小時、4小時、6小時、8小時、18小時、24小時、42小時以及48小時後,取出100μL浸泡過敷料之磷酸緩衝溶液,再加入100μL濃度為1.0wt%之醋酸鋅水溶液、20μL的含有濃度為20mM之N,N-二甲基-1,4-苯二胺硫酸鹽(N,N-dimethyl-p-phenylenediamine sulfate)以及濃度為
7.2N之氯化氫水溶液,與20μL的含有濃度為30mM之氯化鐵(FeCl3)以及濃度為1.2N之氯化氫水溶液,反應20分鐘。之後測量各溶液之亞甲基藍在波長670nm下的吸收強度,以反推實施例及比較例之各敷料在不同時間下的硫化氫濃度。測試結果如下列表1所示。
由實施例1至4所製得的硫化氫緩釋敷料,其可在24小時內有效緩慢釋放硫化氫氣體而未超過安全使用濃度,尤其實施例2及實施例3之硫化氫緩釋敷料更可延長釋放時間至48小時。而比較例1之硫化氫緩釋敷料在釋放過程中之硫化氫濃度高於1300μM,雖亦可長時間釋放硫化氫氣體但已超過硫化氫之安全使用濃度。
雖然本發明已以實施例揭露如上,然其並非用以限定本發明,任何熟習此技藝者,在不脫離本發明之精神和範圍內,當可作各種之更動與潤飾,因此本發明之保護範圍當視後附之申請專利範圍所界定者為準。
Claims (15)
- 一種硫化氫緩釋敷料,其包含:一水膠體;一介面活性劑;以及一硫氫化鈉。
- 如申請專利範圍第1項之硫化氫緩釋敷料,其中該水膠體為60至140重量份,該介面活性劑為0.2至2重量份,且該硫氫化鈉為0.1至0.5重量份。
- 如申請專利範圍第1項之硫化氫緩釋敷料,其中該介面活性劑為聚山梨醇酯80、聚山梨醇酯20、聚山梨醇酯60或聚山梨醇酯40。
- 如申請專利範圍第1項之硫化氫緩釋敷料,其中該水膠體包含:一彈性體;一親水性高分子;一增黏劑;以及一延展劑。
- 如申請專利範圍第4項之硫化氫緩釋敷料,其中該彈性體為10至30重量份,該親水性高分子為20至60重量份,該增黏劑為20至60重量份,且該延展 劑為2至20重量份。
- 如申請專利範圍第4項之硫化氫緩釋敷料,其中該彈性體係選自由苯乙烯-異戊二烯-苯乙烯(SIS)共聚物、苯乙烯-丁二烯-苯乙烯(SBS)共聚物、苯乙烯-(乙烯-丁烯)-苯乙烯(SEBS)共聚物以及苯乙烯-(乙烯-丙烯)-苯乙烯(SEPS)共聚物所組成之群組之至少之一或其組合。
- 如申請專利範圍第4項之硫化氫緩釋敷料,其中該親水性高分子係選自由羧甲基纖維素鈉、羥乙基纖維素、海藻酸鈉、明膠、果膠、羧甲基殼聚醣、瓜爾豆膠、刺槐豆膠、膠原蛋白以及刺梧桐樹膠所組成之群組之至少之一或其組合。
- 如申請專利範圍第4項之硫化氫緩釋敷料,其中該增黏劑係選自由松香樹脂、萜烯樹脂、C5石油樹脂、C9石油樹脂以及高純度二環戊二烯(H-DCPD)所組成之群組之至少之一或其組合。
- 如申請專利範圍第4項之硫化氫緩釋敷料,其中該延展劑係選自由礦物油、液體石蠟、蓖麻油、鄰苯二甲酸二丁酯、羊毛脂以及環烷油所組成之群組之至少之一或其組合。
- 如申請專利範圍第1項之硫化氫緩釋敷料,其中該水膠體更包含0.2至2重量份之抗氧化劑。
- 如申請專利範圍第10項之硫化氫緩釋敷料,其中該抗氧化劑為受阻酚、硫增效劑(thiosynergists)、亞磷酸酯(phosphites)或二級芳香胺(secondary aromatic amines)。
- 一種硫化氫緩釋敷料之製造方法,其步驟包含:加熱攪拌一水膠體材料;將一介面活性劑以及硫氫化鈉加入該水膠體材料中;以及將含有該介面活性劑以及該硫氫化鈉之該水膠體材料注入一成型模具中熱壓成型以形成一硫化氫緩釋敷料。
- 如申請專利範圍第12項之製造方法,其中該水膠體材料為60至140重量份,該介面活性劑為0.2至2重量份,且該硫氫化鈉為0.1至0.5重量份。
- 如申請專利範圍第12項之製造方法,其中在該步驟(a)中之加熱溫度係介於100℃至200℃之間且加熱時間係介於1小時至1.5小時之間。
- 如申請專利範圍第12項之製造方法,其中在該步驟(b)中更包含進行一加熱處理,且加熱溫度係介於100℃至150℃之間且加熱時間係介於15分鐘至45分鐘之間。
