Abstract
Se describe un método para seleccionar células huésped recombinantes con niveles elevados de expresion de una proteína deseada. Este método utiliza células huésped eucarioticas que albergan un constructo o plásmido recombinante de ADN que comprende un gen seleccionable (de manera preferible un gen amplificable) y un gen producto que se proporciona en la direccion 3' respecto al gen seleccionable. El gen seleccionable se coloca dentro de un intron definido por un sitio donador de empalme y sitio aceptor de empalme, y el gen seleccionable y el gen producto están bajo control transcripcional de una sola region reguladora transcripcional. El sitio donador de empalme generalmente es un sitio donador de empalme eficiente y por lo tanto regula la expresion del gen de producto mediante la utilizacion de la region reguladora transcripcional. Las células transfectadas se cultivan de manera que expresan el gen que codifica para el producto en un medio selectivo que comprende un agente amplificador durante tiempo suficiente para permitir que se lleve a cabo la amplificacion, después de la cual el producto deseado se recupera o se identifican las células que tienen copias multiples del gen producto.
