Claims (1)
- 【特許請求の範囲】 1.転写開始部位、転写終了部位、選択可能な遺伝子、選択可能な遺伝子の3' に設けられた生成物遺伝子、選択可能な遺伝子および生成物遺伝子の両方の転写 を制御する転写制御鎖を含んでなり、選択可能な遺伝子はイントロンのスプライ スドナー部位5'を有するイントロンの内部に位置し、そのスプライスドナー部 位は転写制御領域を用いて生成物遺伝子の発現を制御するDNA構築物。 2.スプライスドナー部位が有効なスプライスドナー配列を含有する請求項1に 記載のDNA構築物。 3.スプライスドナー部位がコンセンサススプライスドナー配列を含有する請求 項2に記載のDNA構築物。 4.スプライスドナー部位が配列GACGTAAGTを含有する請求項2に記載 のDNA構築物。 5.選択可能な遺伝子が増幅可能な遺伝子である請求項1に記載のDNA構築物 。 6.増幅可能な遺伝子がDHFRである請求項5に記載のDNA構築物。 7.転写制御領域がプロモーターおよびエンハンサーを含有する請求項1に記載 のDNA構築物。 8.請求項1に記載のDNA構築物を含有するベクター。 9.DNA構築物の選択可能な遺伝子が増幅可能な遺伝子である請求項8に記載 のベクター。 10.真核宿主中で複製できる請求項8に記載のベクター。 11.請求項10に記載のベクターを含有する真核宿主細胞。 12.請求項5のDNA構築物を含有する真核宿主細胞。 13.ベクターを宿主細胞にエレクトロポレーションにより導入する請求項11 に記載の宿主細胞。 14.宿主細胞の染色体に組込まれた請求項1に記載のDNA構築物を含有する 真核宿主細胞。 15.哺乳動物細胞である請求項14に記載の宿主細胞。 16.請求項11に記載の宿主細胞を培養して生成物遺伝子を発現させ、宿主細 胞培養から生成物を回収することを含んでなる問題のタンパク質の製造方法。 17.培地から生成物を回収することを更に含んでなる請求項16に記載の方法 。 18.選択可能な遺伝子が増幅可能な遺伝子であり、スプライスドナー部位が有 効なスプライスドナー配列を含有する請求項16に記載の方法。 19.請求項12に記載の宿主細胞を培養して、十分な時間増幅剤を含有する選 択培地中で生成物遺伝子を発現させて、増幅を起こさせ、生成物を回収すること を含んでなる問題の生成物の製造方法。 20.真核細胞を請求項5に記載のDNA構築物でトランスホームし、増幅剤を 含む選択培地中で増幅が起こるのに十分な時間増殖させ、生成物遺伝子の多重コ ピーを有する細胞を選択することを含んでなる、生成物遺伝子の多重コピーを有 する真核細胞の製造方法。 21.問題の生成物を選択した細胞から回収することを更に含んでなる請求項2 0に記載の方法。
