Abstract

본 발명은 코리네형 박테리아의 말레이트신타제 유전자 앞에 위치하고 코리네형 박테리아로부터 분리되는 DNA 단편에 관한 것이다. 단백질을 코드화하는 어떤 구조의 유전자도 이러한 DNA 단편뒤에 삽입될 수 있다. 이러한 구조가 코리네형 박테리아로 형질전환된 후 DNA 단편뒤에 삽입된 구조 유전자의 발현이 조절된다. 또한 본 발명은 형질전환된 코리네형 박테리아를 배양함으로써 어떤 단백질을 합성하는 방법에 관한 것이다. 이러한 형태의 박테리아는 복제가능형태로 코리네형 박테리아의 말레이트신타제 유전자로부터 분리된 DNA 단편을 함유하고, 합성할 단백질을 코드화하는 구조 유전자가 삽입된다. 합성할 단백질을 코드화하는 구조 유전자의 발현이 유전자 앞에 위치하는 DNA에 의해 조절되기 때문에, 구조 유전자가 발현되고 적합한 유도제를 배지에 가하자마자 목적하는 단백질이 합성된다.

제1도는 플라스미드 pAB-17의 제한지도를 나타낸다, 제2도는 플라스미드 pEKB1a의 제한지도를 나타낸다, 제3도는 플라스미드 pEKB1a의제한지도를 나타낸다, 제4도는 글루코스를 사용하는 CgC-최소배지에서 배양된 C, 글루타미쿰 WT 및 WT(pEKB1b)-세포의 원료 추출물 및 아세테이트를 사용하는 CgC-최소배지에서 배양된 C, 글루타미쿰 WT(pEKB1b)-세포의 원료 추출물을 이들의 단백질 샘플에서 램리(Laemmli)의 방법에 따라 12%의 폴리아크릴아미드 겔상에 분리시킨 MSY-단백질띠를 나타낸다.

Claims (9)

  1. DNA 단편의 뒤에 삽입하고, 벡터내로 구성되며 코리네형 박테리아로 형질전화된 후, 어떤 단백질을 코드화하는 구조유전자의 발현이 조절되는 코리네형 박테리아의 말레이트산타제-유전자앞에 위치하고 이렇게 분리되는 DNA 단편.
  2. 제1항에 있어서, 코리네 박테리아 글루타미쿰의 말레이트신타제-유전자로부터 유도되는 것을 특징으로 하는 DNA 단편.
  3. 제2항에 있어서, 표 2(여기서 표 2는 본 특허청구의 범위의 구성요소이다)에 따른 뉴클레이티드 1 내지 574의 뉴클레오티드 서열을 갖는 DNA 단편.
  4. 제1항 내지 제3항 중의 어느 한 항에 있어서, 어떤 후속 삽입되는 구조유전자를 갖는 DNA 단편.
  5. 제1항 내지 제4항 중의 어느 한 항에 따른 DNA 단편을 함유하는 벡터.
  6. 제1항 내지 제4항 중의 어느 한 항에 따른 단편을 복제가능한 형태로 함유하는 재조합된 코리네형 세포.
  7. 제6항에 있어서, 제5항에 따른 벡터를 함유하는 재조합된 코리네형 세포.
  8. DNA 단편의 뒤에 합성할 단백질을 코드화하는 구조유전자가 삽입되고 DNA 단편이 합성할 단백질을 코드화하는 구조유전자의 발현을 조절하며, 배지에서 유도제를 가하여 당해 구조유전자가 발현되고 이로써 목적하는 단백질을 합성하는, 복제 가능한 형태로 코리네형 박테리아의 말레이트신타제-유전자로부터 분리된 DNA 단편을 함유하는 형질전환된 코리네형 박테리아의 배양에 의한 어떤 단백질의 합성방법.
  9. 제8항에 있어서, 유도제로서 락테이트, 피루베이트, 바람직하게는 아세테이트가 배양배지에 첨가되는 것을 특징으로 하는 방법.
