Abstract

본 발명은 사이클로로스포린을 활성성분으로서 함유하는 외용약제 조성물에 관한 것이다. 더욱 구체적으로, 본 발명은 활성성분으로서 사이클로스포린, 200℃ 이상의 고비점을 갖는 양친매성의 비알콜성 용매중에서 선택된 가용화 흡수증강제, 계면활성제 및 오일로 구성된 자발유화 마이크로에멀젼(self-emulsifying microemulsion) 예비농축물(A), 및 겔화제 및 정제수로 구성된 하이드로겔(hydrogel) (B)를 서로 혼합한 후, 알칼리화제로 pH를 조정하여 제조되는 마이크로에멀젼-함유 하이드로겔 제제이다. 본 발명에 따르는 외용제제는 안정성 및 피부침투성이 우수하며, 피부도포감이 양호하고, 사이클로로스로린이 매우 균일하게 분산되어 있어 제제학적으로 우수하며 제조방법이 간편하다는 잇점을 갖는다.

Claims (19)

  1. 사이클로스포린을 함유하는 자발유화 마이크로에멀젼을 하이드로겔에 분산시킨 사이클로스포린-함유 외용약제 조성물.
  2. 제1항에 있어서, 200℃ 이상의 고비점을 갖는 양친매성의 비알콜성 용매를 가용화 흡수증강제로서 함유함을 특징으로 하는 사이클로스포린-함유 외용약제 조성물.
  3. 제2항에 있어서 사이클로스포린이 사이클로스포린 A임을 특징으로 하는 사이클로스포린 함유 외용약제 조성물.
  4. 제2항에 있어서, 가용화 흡수증강제가 디메틸이소소르비드, 프로필렌카보네이트 및 N-알킬-2-피롤리돈(여기에서 알킬은 탄소수 1 내지 6의 알킬이다)으로 구성된 그룹중에서 선택됨을 특징으로 하는 사이클로스포린-함유 외용약제 조성물.
  5. 제4항에 있어서, 선택된 2가지 이상의 가용화 흡수증강제를 혼합하여 사용함을 특징으로 하는 사이클로스포린-함유 외용약제 조성물.
  6. 제4항에 있어서, 가용화 흡수증강제가 디메틸이소소르비드 또는 N-메틸-2-피롤리돈임을 특징으로 하는 사이클로스포린-함유 외용약제 조성물.
  7. 제4항에 있어서, 가용화 흡수증강제가 N-메틸-2-피롤리돈임을 특징으로 하는 사이클로스포린-함유 외용약제 조성물.
  8. 제1항 또는 제2항에 있어서, 추가로 계면활성제, 오일, 겔화제, 정제수 및 알칼리화제 중에서 선택된 1종 이상의 성분을 함유함을 특징으로 하는 사이클로스포린-함유 회용액제 조성물.
  9. 제8항에 있어서, 계면활성제가 천연식물성오일 트리글리세라이드와 폴리알킬렌폴리올이 트랜스에스테르화 반응생성물, 폴리옥시에틸렌글리콜화 천연 또는 수소화 식물성 오일, 폴리옥시에틸렌-소르비탄 지방산 에스테르, 스르비탄 지방산 에스테르 및 폴리옥시에틸렌 알킬 에테르로 구성된 그룹중에서 선택됨을 특징으로 하는 사이클로스포린-함유 외용약제 조성물.
  10. 제9항에 있어서, 계면활성제가 천연식물성 오일 트리글리세라이드와 폴리알킬렌폴리올의 트랜스에스테르화 반응생성물임을 특징으로 하는 사이클로스포린-함유 외용약제 조성물.
  11. 제8항에 있어서, 오일이 수증유(o/w)형 에멀젼에서의 요구 HLB가 10 이하인 중급지방산 트리글리세라이드, 식물성 오일 또는 동물성 오일임을 특징으로 하는 사이클로스포린-함유 외용약제 조성물.
  12. 제11항에 있어서, 오일이 정제어유, 옥수수유 또는 카프릴산/카프릭 트리글리세라이드임을 특징으로 하는 사이클로스포린-함유 외용약제 조성물.
  13. 제8항에 있어서, 오일이 수중유(o/w)형 에멀젼에서의 요구 HLB가 10 이하인 오일과 수증유(o/w)형에멀젼에서의 오구 HLB가 10 이상인 오일의 혼합물임을 특징으로 하는 사이클로스포린-함유 외용약제 조성물.
  14. 제8항에 있어서, 겔화제가 셀룰로즈 유도체, 카복시비닐 폴리머 및 폴리옥시에틸렌-폴리옥시프로필렌 공중합체로 구성된 그룹중레서 선택됨을 특징으로 하는 사이클로스포린-함유 외용약제 조성물.
  15. 제14항에 있어서, 겔화제가 카복시비니폴리머임을 특징으로 하는 사이클로스포린-함유 외용약제 조성물.
  16. 제8항에 있어서, 각 구성성분이 조성물의 총중량을 기준으로하여 사이클로스포린 0.1 내지 10% , 가용화 흡수증강제 0.5 내지 50%, 계면활성제 0.4 내지 40%, 오일 0.4 내지 40%, 겔화제 0.1 내지 5%, 정제수 0.1 내지 90%, 알칼리화제 0.01 내지 3.5%의 비로 함유됨을 특징으로 하는 사이클로스포린-함유 외용제 조성물.
  17. 제16항에 있어서, 각 구성성분이 조성물의 총중량을 기준으로하여 사이클로스포린 0.1 내지 5%, 가용화 흡수증강제 0.5 내지 25%, 계면활성제 0.4 내지 20%, 오일 0.4 내지 20%, 겔화제 0.1 내지 2.5%, 정제수 40 내지 90%, 알칼리 화제 0.05 내지 1.8%의 비로 함유됨을 특징으로 하는 사이클로스포린-함유 외용약제 조성물.
  18. 제2항에 있어서, pH가 3 내지 9임을 특징으로 하는 사이클로스포린-함유 외용약제 조성물.
  19. 사이클로스포린을 가용화 흡수증강제에 용해시키고, 생성된 용액에 계면활성제 및 오일을 혼합시켜 자발유화 마이크로에멀젼 예비농축액을 제조한 후, 이 마이크로에멀젼 예비농축액을 겔화제를 정제수에 균질하게 용해시킨 하이드로겔에 혼합시켜 교반한 후, 알칼리화제를 가하여 마이크로에멀젼을 함유하는 하이드로겔 제제를 제조함을 특징으로 하여 제1항 내지 제18항 중의 어느 한 항에 따르는 사이클로스포린-함유 외용약제를 제조하는 방법
