Claims (22)
- 인체 과립구 콜로니 자극인자(hG-CSF) 폴리펩타이드의 아미노산 서열의 N 말단부의 1번 내지 17번 아미노산중 하나 이상의 아미노산이 Cys, Ser, Arg, Ala, Thr, Pro, Tyr, Asn, Gly, Lys, Glu, Val 및 Ile을 포함하는 그룹중에서 선택되는 하나 이상의 아미노산에 의해 독립적으로 치환되는, 인체 과립구 콜로니 자극 인자 폴리펩타이드의 아미노산 서열로부터 유도된 아미노산 서열을 갖는 폴리펩타이드.
- 제1항에 있어서, N 말단부의 1번 내지 6번 아미노산중 하나 이상의 아미노산 및 17번 위치의 아미노산이 hG-CSF 폴리펩타이드의 상응하는 부위에서의 아미노산과 다른 폴리펩타이드.
- hG-CSF 폴리펩타이드의 N 말단부의 1번 내지 17번 아미노산중 하나 이상의 아미노산이 Cys, Ser, Arg, Ala, Thr, Pro, Tyr, Asn, Gly, Lys, Glu, Val 및 Ile을 포함하는 그룹중에서 선택되는 하나 이상의 아미노산에 의해 독립적으로 치환되는, hG-CSF 폴리펩타이드의 아미노산 서열로부터 유도된 아미노산 서열을 갖는 폴리펩타이드를 코드화하는 DNA.
- 플라스미드 DNA와, 그중의 삽입물로서, hG-CSF 폴리펩타이드의 N 말단부의 1번 내지 17번 아미노산중 하나 이상의 아미노산이 Cys, Ser, Arg, Ala, Thr, Pro, Tyr, Asn, Gly, Lys, Glu, Val 및 Ile을 포함하는 그룹중에서 선택되는 하나 이상의 아미노산에 의해 독립적으로 치환되는, hG-CSF 폴리펩타이드의 아미노산 서열로부터 유도된 아미노산 서열을 갖는 폴리펩타이드를 코드화하는 DNA 단편을 포함하는 재조합 플라스미드.
- 제4항에 있어서, 플라스미드 DNA가 트립토판 프로모터-함유 플라스미드 DNA이고, DNA 단편이 트립토판 프로모터의 하부 스트림 위치에서 플라스미드 DNA에 삽입된 재조합 플라스미드.
- 제4항 또는 제5항에 있어서, DNA 단편이, 염기 서열의 1번 내지 51번 위치까지의 염기중의 하나 이상의 염기가 다른 염기에 의해 치환된 서열을 갖는 재조합 플라스미드.
- 제4항 또는 제5항에 있어서, pCfTL38, pCfTL41, pCfTL23, pCfTL35, pCfBB101, pCfBC42B1, pCfBC45, pCfBC52, pCfBC59, pCfBC76, pCfBC77, pCfBC93, pCfBC95, pCfBC97, pCfBD28, pCfBD56, pCfBD82, pCfTM14, pCfTM17, pCfTM113, pCfTAArg4S, pCfTAArg4, pCfBD28A17 및 pCfBD28T17으로 이루어진 그룹중에서 선택되는 재조합 플라스미드.

- 제8항에 있어서, 재조합 플라스미드가 pCfTL38, pCfTL41, pCfTL23, pCfTL35, pCfBB101, pCfBC42B1, pCfBC45, pCfBC52, pCfBC59, pCfBC76, pCfBC77, pCfBC93, pCfBC95, pCfBC97, pCfBD28, pCfBD56, pCfBD82, pCfTM14, pCfTM17, pCfTM113, pCfTAArg4s, pCfTAArg4, pCfBD28A17 및 pCfBD28T17으로 이루어진 그룹중에서 선택되는 미생물.
- (a) 플라스미드 DNA와, 그중의 삽입물로서, hG-CSF 폴리펩타이드의 N 말단부의 1번 내지 17번 아미노산중 하나 이상의 아미노산이 Cys, Ser, Arg, Ala, Thr, Pro, Tyr, Asn, Gly, Lys, Glu, Val 및 Ile을 포함하는 그룹중에서 선택되는 하나 이상의 아미노산에 의해 독립적으로 치환된 아미노산 서열을 갖는 폴리펩타이드를 코드화하는 DNA 단편을 포함하는 재조합 플라스미드를 함유하는 에스케리키아 콜라이 종에 속하는 미생물을 배지에서 배양함으로써 폴리펩타이드를 배양액중에 형성 및 축적시키는 단계, 및 (b) 배양액으로부터 상기 폴레펩타이드를 회수하는 단계들을 포함하는, hG-CSF 폴리펩타이드의 아미노산 서열로부터 상기와 같이 유도된 아미노산 서열을 갖는 폴리펩타이드를 제조하는 방법.
- 제 10항에 있어서, 재조합 플라스미드가 pCfTL38, pCfTL41, pCfTL23, pCfTL35, pCfBB101, pCfBC42B1, pCfBC45, pCfBC52, pCfBC59, pCfBC76, pCfBC77, pCfBC93, pCfBC95, pCfBC97, pCfBD28, pCfBD56, pCfBD82, pCfTM14, pCfTM17, pCfTM113, pCfTAArg4s, pCfTAArg4, pCfBD28A17 및 pCfBD28T17으로 이루어진 그룹중에서 선택되는 방법.

- 제14항에 있어서, hG-CSF 폴리펩타이드의 N 말단부의 1번 내지 4번, 1번 내지 5번, 1번 내지 6번, 1번 내지 7번, 또는 1번 내지 11번 아미노산을 포함하는 펩타이드가 결실된 폴리펩타이드를 코드화하는 DNA.
- 플라스미드 DNA와, 그중의 삽입물로서, hG-CSF 폴리펩타이드의 N 말단부의 1번 내지 11번 아미노산중 하나 이상의 아미노산이 결실됨으로써 유도된 폴리펩타이드를 코드화하는 DNA 단편을 포함하는 재조합 플라스미드.
- 제16항에 있어서, 플라스미드 DNA와 그중의 삽입물로서, hG-CSF 폴리펩타이드의 N 말단부의 1번 내지 11번 아미노산을 포함하는 펩타이드가 결실됨으로써 유도된 폴리펩타이드를 코드화하는 DNA 단편을 포함하는 재조합 플라스미드.
- 제17항에 있어서, 플라스미드 DNA가 트립토판 프로모터-함유 플라스미드 DNA이고, DNA 단편이 트립토판 프로모터의 하부 스트림 위치에서 플라스미드 DNA에 삽입된 재조합 플라스미드.
- 제18항에 있어서, pCfTNS7, pCfTNS301, pCfTNS401 및 pCfTNS501로 이루어진 그룹중에서 선택되는 재조합 플라스미드.
- 제12항에 있어서, 아미노산 결실 이외에, hG-CSF 폴리펩타이드의 17번 아미노산인 시스테인(Cys)이 세린(Ser)으로 치환된 폴리펩타이드.
- 플라스미드 DNA와, 그중의 삽입물로서, hG-CSF 폴리펩타이드의 아미노산 서열의 N 말단부 1번 내지 11번 아미노산을 포함하는 펩타이드가 결실됨으로써 유도된 아미노산 서열을 갖는 폴리펩타이드를 코드화하는 DNA 단편을 포함하는 재조합 플라스미드를 함유하는 에스케리키아 콜라이 종에 속하는 미생물을 배지에서 배양함으로써 폴리펩타이드를 배양액중에 형성 및 축적시키는 단계, 및 배양액으로부터 폴리펩타이드를 회수하는 단계들을 포함하여, hG-CSF 폴리펩타이드의 아미노산 서열의 N 말단부의 1번 내지 11번 아미노산을 포함하는 펩타이드가 결실됨으로써 유도된 아미노산 서열을 갖는 폴리펩타이드를 제조하는 방법.
- hG-CSF 폴리펩타이드의 17번 아미노산(시스테인)이 세린으로 치환되고 이의 N 말단부 1번 내지 6번 아미노산중의 하나 이상의 아미노산이 Cys, Ser, Arg, Ala, Thr, Pro, Tyr, Asn, Gly, Lys, Glu, Val 및 Ile을 포함하는 그룹중에서 선택되는 하나 이상의 아미노산에 의해 독립적으로 치환됨으로써 유도된 폴리펩타이드를 수정 배지에서 프로테아제로 처리함으로써 반응 혼합물중에서 상기와 같이 유도된 폴리펩타이드를 형성시키는 단계, 및 반응 혼합물로부터 이러한 폴리펩타이드를 회수하는 단계들을 포함하여, hG-CSF 폴리펩타이드의 N 말단부의 4 내지 7개 아미노산이 결실되고 17번 아미노산(시스테인)이 세린으로 치환된 hG-CSF 폴리펩타이드의 아미노산 서열로부터 유도된 아미노산 서열을 갖는 폴리펩타이드를 제조하는 방법.
