Description

에트골린의 N-메틸 유도체 제조방법
본 발명은 에르골린의 N-메틸 유도체의 제조방법에 관한 것이다.
이 화합물들은 말초 및 뇌혈관 확장제(니세르골린) 또는 편두통 예방제(메틸세르기드, 메테르골린)과 같은 약리학적 활성을 갖는 화합물 제조에 있어서 유용한 중간체이다.
일반적으로, 알카리금속 수산화물 또는 수소화물, 아미드 또는 알코올레이트 또는 유기마그네슘 또는 유기리튬 유도체와 같은 유기금속 유도체인 염기 존재하에서 할로겐화메틸 또는 황산디메틸로인돌고리계를 메틸화하는 것이 공지되어 있다(M.C. Reinecke, J.Org. Chem., 37 (20) 3066 (1972) 참조). 그러나 이 기술은 인돌고리계가 질소-포함하거나 또는 히드록실 치환제를 가질때 만족스러운 결과를 얻을 수 없다.

독일연방공화국 특허출원 제2,618,033호 및 제3,007,196호에는 150℃ 부근의 온도에서 요오드화메틸 및 탄산메틸을 사용하거나 또는 180-250℃의 온도에서 탄산메틸의 사용에 의한 아닐린의 메틸화가 개시되어 있다.
프랑스공화국 특허출원 제2,478,639호에는 수산화나트륨 존재하에서 탄산메틸을 사용하고, 혼합물을 테트라히드로푸란의 환류온도까지 6시간동안 가열함에 의한 한 특정 인돌 유도체, 1-(2-(3-인돌일)에틸-2-피롤리돈의 메틸화가 개시되어 있다. 그러나 에르골린 유도체의 메틸화에 이 방법의 적용은 만족스러운 결과를 유도하지 못했다.
본 발명은 하기 구조식(II)의 에르골린 유도체를 임의적으로 하기 구조식(III)의 화합물 또는 크라운 에테르 같은 금속 이온봉쇄제의 존재하, 및 4차 암모늄염 예를들면 염화 테트라부틸 암모늄같은 상전이 촉매 존재하의 비극성, 중성 용매에서 수소화알카리 금속 또는 알코올레이트 예를들면 수소화나트륨 또는 칼륨 또는 알코올레이트 같은 염기성 시약 존재하에서 탄산메틸로 메틸화하는 것으로 구성되는 하기 구조식(I)의 에르골린의 N-메틸 유도체의 제조방법을 제공한다.

상기식에서, R1은 알킬 부분이 탄소수 1-4를 포함하는 카르복시 또는 알콕시카르보닐기이고, R2는 수소원자 또는 메톡시기이고, R3는 수소원자이거나; 또는 R2및 R3는 함께 직접적 결합을 형성하거나; 또는 R1이 히드록시메틸기일때 R2는 수소원자 또는 메톡시기이고, R3는 수소원자이고, n은 1, 2 또는 3이다.
반응은 일반적으로 0-100℃ 바람직하게는 40-60℃의 온도에서 수행한다.
에테르, 에스테르, 니트릴 또는 방향족 탄화수소는 비극성의 중성 용매로 사용될 수 있는 화합물의 예이다. 이 공정은 탄산메틸 자체내에서 수행할 수 있고 따라서 이것은 메틸화제 및 용매로 작용하고 이것은 그 비교적 낮은 비동점(90℃)의 결과 쉽게 제거될 수 있다.
본 발명에 의한 공정은 메틸화 반응을 할로겐화메틸 및 황산디메틸과 같은 다른 메틸화제보다 더욱 쉽게 취급될 수 있는 화합물을 사용하여 온화한 조건하에서 수행할 수 있게 한다. 탄산메틸은 특히 훨씬 독성이 적다.
에르골린 고리계가 3차 아민 또는 일차 알코올기로 치환되어 있을 때, 3차 아민의 4차화 또는 일차 알코올의 에테르화 같은 다른 공지의 메틸화제에 의해 일어나는 부반응이 일반적으로 발생하지 않는다. 그러나 에르골린 고리계가 일차 알코올기로 치환되어 있으면, 최종 생성물을 분리하기 전에 수산화나트륨같은 염기로 반응 생성물을 처리하는 것이 유익하다.
에르골린 고리계가 에스테르기로, 특히 메틸에스테르기로 치환되어 있을 때, 이 기는 본 발명에 따르는 공정이 수행되는 조건에 기인한 비누화 반응을 방지한다.
다음의 실시예들은 본 발명을 실행하는 방법을 보여준다.
[실시예 1]
탄산메틸(25cc)내의 10α-메톡시루밀리세르 게이트 메틸(314mg : 10-3)의 용액을 메틸나트륨염(16×10-3mol) 및 염화 테트라부틸암모늄(1.75×10-3mol) 을 포함하는 반응기에 붓는다.
수분을 차단하여 유지된 반응 혼합물을 20℃ 부근의 온도에서 24시간동안 교반한다.
고성능 액체 크로마토그라피로 반응 혼합물을 분석하면 이것이 N-메틸-10α-메톡시루밀리 세르게이트 메틸(0.8×10-3mol) 을 포함한다는 것을 알 수 있다.
[실시예 2]
염화 테트라부틸암모늄(1.5mmol) 및 메틸나트륨염(24mmol)을 계속적으로 탄산메틸(100cc)내의 10α-메톡시루밀리세르 게이트 메틸(943mg : 3mmol)의 용액에 첨가한다.
수분을 차단하여 유지된 반응 혼합물을 60℃에서 3시간 30분간 교반한다. 반응 혼합물 N-메틸-10α-메톡시루밀리세르 게이트 메틸(2.83mmol)을 포함하고 이것은 수율 94%에 상응한다.
10α-메톡시루밀리세르 게이트 메틸의 전환도는 98.5%이다. 초산(24mmol)을 반응 혼합물에 첨가하고, 이것을 실리카상에서 여과한다. N-메틸-10α-메톡시루밀리세르 게이트 메틸의 특성은 양성자 핵자기 공명 및 고성능 액체 크로마토그라피로 확인하고 그것에 의해 분리한다.
[실시예 3]
10α-메톡시루밀리세르골(268mg; 1mmol)을 탄산메틸 및 테트라히드로푸란(부피비 1 : 1)의 혼합물(30cc)에 현탁시킨다. 메틸 나트륨염(8mmol) 및 염화 테트라부틸암모늄(2mmol)을 첨가한다. 반응 혼합물을 60℃에서 5시간동안 교반한다. 용매를 감압하에서 증발시키고 메탄올/몰(부피비 4 : 1) 혼합물(30cc)를 첨가한다. 수성의 메탄올성 용액을 3시간동안 60℃까지 가열한다.
N-메틸-10α-메톡시루밀리세르골을 10℃ 부근의 온도에서 물을 첨가함으로 침전시킨후 분리한다. N-메틸-10α-메톡시루밀리세르골의 수율은 96%이다. 10α-메톡시루밀리세르골의 전환도는 98%이다.
얻어진 생성물의 구조는 핵자기 공명 및 고성능 액체 크로마토그라피로 확인한다.
[실시예 4]
10α-메톡시루밀리세르 게이트 메틸 1mmol, 탄산메틸 8cc, 염화 테트라부틸암모늄 0.02mmol, 및 메틸 나트륨염 8mmol을 사용하여 실시예 2의 공정을 반복한다.
60℃에서 22시간 가열한 후, N-메틸-10α-메톡시루밀리세르 게이트 메틸(0.72mmol)이 수율 72%로 형성되었다.

Claims (9)

  1. 하기 구조식(II)의 에르골린 유도체를 상전이촉매 존재하 비극성의 중성 용매에서, 염기성 시약의 존재하에서 탄산메틸로 메틸화하는 것으로 구성되는, 하기 구조식(I)의 에르골린의 N-메틸 유도체의 제조방법.

    상기식에서, R1은 알킬부분이 탄소수 1-4를 포함하는 카르복시 또는 아콜시카르보닐기이고, R2는 수소우너자 또는 메톡시기이고, R3는 수소원자이거나; 또는 R2및 R3는 함께 직접적 결합을 형성하거나; 또는 R1이 히드록시메틸기일때 R2는 수소원자 또는 메톡시기이고 R3는 수소원자이다.
  2. 제1항에 있어서, 금속이온봉쇄제 존재하에서 수행하는 방법.
  3. 제1항에 있어서, 용매가 에테르, 에스테르, 니트릴 또는 방향족 탄화수소인 방법.
  4. 제1항에 있어서, 용매가 탄산메틸인 방법.
  5. 제1항에 있어서, 상전이촉매가 4차 암모늄염인 방법.

  7. 제2항에 있어서, 금속이온봉쇄제가 크라운 에테르 또는 하기 구조식(Ⅲ)의 화합물인 방법.
