Claims (11)
- 적절 리딩 플레임내에 분비리더 및 숙주에 의해 인식되는 처리시그날 서열과 결합된 인체인슐린 유사생성인자(IGF)를 암호하는 유전자를 갖고 있는 기능성DNA구조물이 상기 숙주에 적합한 벡터내의 전사 개시부위의 전사 조절콘트롤하에 그 하부쪽으로 연결되어 구조 유전자를 형성하고 있는 상태로 포함되어 있는 숙주세포를 생장시키고, 분비된 인체 IGF를 분리해내는 단계로 구성된 적절한 숙주내에서 인체인슐린-유사생장인자(IGF)를 효율적으로 제조하는 방법.
- 제1항에 있어서, IGF유전자가 상기 숙주에 의해 우선적으로 이용되는 코돈을 가진 합성 유전자인 방법.
- 제1항에 있어서, 숙주가 효모인 방법.
- 제1항에 있어서, 숙주가 효모이고 운반체가 2㎛ 플라스미드의 유도체의 방법.
- IGF를 암호하는 제1DNA 서열을 포함하며 이 DNA서열의 제1염기에 또는 그 하부에 암호 사상체의 5'-말단을 갖는 제1DNA 분체를 제조하고, 효모가 인식할 수 있는 분비 및 처리시그라날(processing signal)을 암호하는 제2DNA 서열을 포함하여 상기 처리시그날의 말단 염기에 또는 그 상부에 암호 사상체의 3'-말단을 가지는 제2DNA 분체를 제조하고, 여기에서 상기 제1 및 제2DNA 분체중의 적어도 하나가 그 말단내부에 염기쌍 ㅅ실을 일으키는 암호서열을 가지고, 제 1 및 제 2DNA 분체를 상기 제1및 제2의 DNA 분체의 염기쌍 소실을 일으키는 암호하는 어댑터(adaptor)를 사용하여 결합하고, 여기에서 상기 제1, 제2DNA 분체는 동일한공 리딩 프레임내에 존재하게 하며, 제1분체의 입접 및 하부의 종결코돈을 제공하고, "전구"-IGF 유전자를 효모내 발현벡터내로 클로닝시키며, 여기서 상기 "전구"-IGF가 분비되어 처리되는 처리되는 것을 특징으로 하는 효모분비 및 처리시그날에 의해 촉진 분비되는 인체인슐린 유사생장인자(IGF)를 효모내에서 발현시키는 방법.
- 제7항에 있어서, 상기 IGF가 IGF I인 방법.
- 제7항에 있어서, 상기 IGF가 IGF II인 방법.
- 제7항에 있어서, 상기 발현벡터가 박테리아에 의해 인지되는 복제시스템을 포함하는 방법.
- 제7항에 있어서, 상기 효모복제 체계가 2㎛플라스미드 또는 그 일부를 포함하는 방법.
