Claims (2)
- 돼지 성장 호르몬의 아미노산 배열 순서에 따라 효모에서 선택적으로 사용되는 코돈 및 Sac I, Xba I 및 Sal I의 인지부위가 존재하는 올리고 뉴클리오티드를 합성한 후 이를 접합전략에 따라 접합시켜 합성올리고 뉴클리오티드의 카복시쪽 Xba I/Sal I 절편을 Xba I/Sac I으로 처리한 대장균 운반체(pUC18)에 클로닝한 다음 이를 Sac I/Xba I으로 처리하여 합성 올리고 뉴클리오티드의 아미노쪽 Sac I/Xba I 절편을 클로닝시켜 아미노 말단의 일부가 없는 돼지 성장 호르몬 유전자를 제조한 다음 이 유전자를 아미노말단 합성유전자와 함께 프로모터와 터미네이터가 있는 대장균 운반체에 클로닝하여 프로모터-전체 돼지 성장 호르몬 유전자-터미네이터의 카세트를 제조한 다음 이를 효모발현 운반체에 재클로닝시켜 최종 효모발현 운반체를 만들어 이를 효모내에서 발현시킴을 특징으로 하는 돼지 성장 호르몬의 제조방법.
