B형 간염비루스 유전자를 삽입한 조환 플라스미드, 이 조환 플라스미드로 전환된 효모 및 B형 간염비루스 표면항원의 제조방법
본 내용은 요부공개 건이므로 전문내용을 수록하지 않았음
제1도는 본 발명에서 사용되는 HBV 유전자를 함유하는 HBV DNA 단편의 구조도.
제2도 (1),(2),(3)은 HBs 유전자의 아미노산 배열도.
제3도는 셔틀벡터 PAT 77 및 AM 82의 구조도.

Claims (23)

  1. 효모의 유전자와 대장균의 유전자를 함유하고 포스파티아제 촉진유전자의 조절하에 조환되는 효모의 억제성 산성 포스파티아제 유전자의 발현조절영역과 B형 간염비루스 유전자를 나르는 플라스미드 벡터로 된 조환 플라스미드.
  2. 제1항에 있어서, 전술한 효모의 억제성 산성 포스파타아제의 유전자의 발현조절영역이 포스파타아제를 구성하는 60,000달톤(P60)의 폴리펩티드의유전자이며, 포스파타아제의 구조유전자의 일부 또는 전부 및 임의의 상류의 어떤 영역이 제거되어 있는 조환 플라스미드.
  3. 제2항에 있어서, 전부의 포스파타아제 구조유전자에 대해서 포스파타아제 구제유전자의 상류에서 제거될 영역이 =1(ATG)에서 -100bp 범위 이내인 조환 플라스미드.
  4. 제3항에 있어서, 제거될 영역이 +1에서 -50bp 범위 이내인 조환 플라스미드.
  5. 제1항에 있어서, 전술한 효모의 유전자가 ars 1 및 2μ ori를 함유하는 조환 플라스미드.
  6. 제1항에 있어서, 전술한 효모의 유전자가 ars 1, 2μori 및 전환효모의 선택표식 유전자를 가지고 있는 조환 플라스미드.
  7. 제6항에 있어서, 전술한 선택표식 유전자가 로이신 생선유전자, 히스티딘 생산유전자, 트립토판 생산유전자, 우라실 생산유전자, 아대닌 생산유전자로 된 군에서 일원 및 이것의 둘 또는 그 이상의 결합인 조환 플라스미드.
  8. 제1항에 있어서, 전술한 대장균의 유전자가 대장균의 세포 내에서 유전자의 증식에 필요한 DNA 배열을 가지고 있는 조혼 플라스미드.
  9. 제1항에 있어서, 전술한 대장균의 유전자가 대장균의 세포 내에서 유전자의 증식에 필요한 DNA 배열과 전환 대장균으 선택표시 유전자를 가지고 있는 조환 플라스미드.
  10. 제9항에 있어서, 선택표식 유전자가 암피실린 내성유전, 자카나마이신 내성유전자, 테트라사이클린 내성유전자 및 클로람페니콜 내성유전자로 된 균에서 선택되는 일원 및 이것을 둘 또는 그 이상의 결합인 조환 플라스미드.
  11. 제9항에 있어서, 전술한 대장균의 유전자가 암피실린 내성유전자와 테트라사이클린 내성유전자를 함유하는 대장균 플라스미드 pBR 322인 조환 플라스미드.
  12. 제1항에 있어서, 전술한 플라스미드 벡터가 효모 플라스미드 및 대장균 플라스미드로 된 셔틀벡터이고, 이 효모플라스미드가 일부 또는 전부의 산성 포스파타아제 구조유전자와 이 포스파타아제 구조유전자의 상류의 어떤 영역이 제거되는데, ars 1, 2μori, Leu 2 및 억제성산성 포스파타아제 유전자의 발현조절영역을 이 순서대로 함유하고 있고 또 전술한 대장균 플라스미드가 암피실린 내성유전자와 테트라사이클린 내성유전자를 가진 대장균 플라스미드 pBR 322인 조환 플라스미드.
  13. 제12항에 있어서, 전술한 효모 플라스미드가 산성 포스 파타아제 구조유전자의 전부와 이 포스파타아제 구조유전자상류의 영역이 +1에서 -50bp 범위내에서 결핍되어 있는 조환 플라스미드.
  14. 제13항에 있어서 포스파타아제 구조유전자 상류의 전술한 결핍영역이 +1내지 -50bp 범위내인조환 플라스미드.
  15. 제1항에 있어서, 플라스미드 벡터와 조환될 전술한 B형 간염비루스의 유전자가 226아미노산에 대응하는 HBs 유전자의 전부를 함유하는 단편인 조환 플라스미드.
  16. 제15항에 있어서, HBs 유전자의 전부를 함유한 전술한 단편이 제한효소 Xho I로 개열되는 HBV DNA의 전부인 조환 플라스미드.
  17. 제15항에 있어서, HBs 유전자의 전부를 함유하는 전술한 단편이 HBV DNA를 제한효소 Xho I 및 BamHI로 절단시켜서 제조한 단편인 조환 플라스미드.
  18. 제1항에 있어서 설명한 조환 플라스미드로 효모를 전환시켜서 제조한 전환효모.
  19. 제18항에 있어서, 전술한 출발재료가 로이신 요구성 돌연변이 균주, 히스리딘 요구성 돌연변이 균주, 트립토판 요구성 돌연변이 균주, 우리실 요구성 돌연변이 균주 및 아데닌 요구성 돌연변이 균주로 된군에서 선택되는 일원의 전환효모.
  20. 제18항에 있어서, 출발재료가 로이신 요구성 돌연변이 균주; 시카로미세스 세레비시아에 〔a, Leu 2, his 4, can 1(Cir+)〕인 전환효모.
  21. 제18항에 있어서, 설명한 전환효모를 배지에서 배양하고 또 생성된 항원을 종래방법으로 수집하는 것으로 B형 간염비루스 표면항원의 제조방법.
  22. 제21항에 있어서, 전환효모의 배양이 산성 포스파타아제 촉진유전자가 억제되지 않는 조건하에 수행되는 방법.
