Description
도 2는 재조합 Ⅳ형 인간화 콜라겐 C4P7C6의 발현 및 정제 과정에서의 전기 영동 검출 결과이다.
도 3은 재조합 Ⅳ형 인간화 콜라겐 C4P7E5의 발현 및 정제 과정에서의 전기 영동 검출 결과이다.
도 4는 재조합 Ⅳ형 인간화 콜라겐 C4P7C6과 재조합 Ⅳ형 인간화 콜라겐 C007의 발현 정제에 있어서 안정성을 비교한 결과이다.
도 5는 각 재조합 Ⅳ형 인간화 콜라겐의 세포 접착 활성을 비교한 결과이다.

Claims (10)
- 아미노산 서열이 n(n은 1 이상의 정수)개의 반복 서열을 포함하는 재조합 IV형 인간화 콜라겐으로서,
상기 반복 서열이 인간 천연 IV형 콜라겐의 기능영역 서열이고,
바람직하게는, 상기 n이 1, 2, 3, 4, 5, 6, 7 또는 8이고, 여기서 n이 2 이상의 정수인 경우, 각 반복 서열끼리 직접 연결되어 있고,
보다 바람직하게는, 상기 재조합 IV형 인간화 콜라겐의 아미노산 서열이 하기 (i) 내지 (iii) 중 어느 하나를 포함하는 것을 특징으로 하는, 재조합 IV형 인간화 콜라겐:
(i) SEQ ID NO. 1, SEQ ID NO. 2, SEQ ID NO. 3에 표시된 아미노산 서열,
(ii) SEQ ID NO. 1, SEQ ID NO. 2, SEQ ID NO. 3에 표시된 아미노산 서열에서 1 또는 복수 개의 아미노산 잔기가 부가, 치환, 결실 또는 변형된 아미노산 서열이면서, 인간 천연 IV형 콜라겐의 세포 접착 효과를 유지하고 있는 아미노산 서열,
(iii) 하기 뉴클레오티드 서열에 의해 코딩되는, 인간 천연 IV형 콜라겐의 세포 접착 효과를 유지하고 있는 아미노산 서열로서, 상기 뉴클레오티드 서열이 SEQ ID NO. 1, SEQ ID NO. 2, SEQ ID NO. 3에 표시된 서열을 코딩하는 폴리뉴클레오티드 서열과 엄격성 조건하에 혼성화하고, 상기 엄격성 조건이 중(中) 엄격성 조건, 중-고 엄격성 조건, 고(高) 엄격성 조건 또는 초고(超高) 엄격성 조건인, 아미노산 서열. - 청구항 1에 있어서,
아미노산 서열이 추가로 SEQ ID NO. 7에 표시된 TEV 프로테아제에 의해 절단 가능한 아미노산 서열을 포함하는 재조합 IV형 인간화 콜라겐으로서,
바람직하게는, 상기 TEV 프로테아제에 의해 절단 가능한 아미노산 서열과 상기 반복 서열이 직접 연결되어 있고,
바람직하게는, 상기 TEV 프로테아제에 의해 절단 가능한 아미노산 서열이 상기 재조합 IV형 인간화 콜라겐의 N 말단에 위치하는 것을 특징으로 하는, 재조합 IV형 인간화 콜라겐. - 청구항 2에 기재된 재조합 IV형 인간화 콜라겐을 코딩하는 폴리뉴클레오티드로서,
바람직하게는, SEQ ID NO. 4, SEQ ID NO. 5 또는 SEQ ID NO. 6에 표시된 서열을 포함하는, 폴리뉴클레오티드. - 청구항 3에 기재된 폴리뉴클레오티드를 포함하는 재조합 발현 벡터로서,
바람직하게는, 상기 재조합 발현 벡터가 pET 시리즈 벡터, 셔틀 벡터, 파지 또는 바이러스 벡터를 포함하고,
보다 바람직하게는, 상기 재조합 발현 벡터가 pET-28a-Trx-His인 것을 특징으로 하는, 재조합 발현 벡터. - 청구항 4에 기재된 재조합 발현 벡터를 포함하는 재조합 숙주 세포로서,
바람직하게는, 상기 재조합 숙주 세포가 원핵 세포, 효모 또는 진핵 세포이고,
보다 바람직하게는, 상기 재조합 숙주 세포가 대장균 BL21(DE3)인 것을 특징으로 하는, 재조합 숙주 세포. - 청구항 1 또는 2에 기재된 재조합 IV형 인간화 콜라겐의 제조방법으로서,
S1: 청구항 5에 기재된 재조합 숙주 세포를 발효 배양하여 단백질 발현을 유도하는 공정;
S2: 상기 단백질을 채집하는 공정; 및
S3: 상기 단백질을 정제하는 공정을 포함하고,
Ni 친화 크로마토그래피 칼럼 및/또는 이온 교환 크로마토그래피 칼럼을 이용해서 상기 단백질을 정제하는 것이 바람직하고,
임의로 상기 단백질을 효소 소화하는 것을 포함하며, TEV 프로테아제를 이용해서 상기 단백질을 효소 소화하는 것이 바람직하고, 상기 단백질과 상기 TEV 프로테아제의 질량비가 (15 내지 25):1인 것이 보다 바람직한, 제조방법. - 청구항 6에 있어서,
상기 S1 공정에서, 상기 발효 배양은 회전 속도 200 내지 240rpm, 온도 35 내지 38℃의 쉐이커 내에서 이루어지는 것을 특징으로 하는, 제조방법. - 청구항 6 또는 7에 있어서,
상기 S1 공정에서, 상기 단백질 발현을 유도하는 구체적인 절차는 16 내지 30℃까지 온도를 낮추어 IPTG를 첨가하고, 바람직하게는 상기 IPTG의 사용 농도가 0.3 내지 0.7mM인 것을 특징으로 하는, 제조방법. - 청구항 6 내지 8 중 어느 한 항에 있어서,
상기 S2 공정에서, 상기 단백질을 채집하는 구체적인 절차는 S1 공정에서 얻어진 혼합물을 4℃ 환경하에 5,000 내지 7,000rpm으로 10 내지 15분간 원심 분리하고, 20 내지 30mM Tris, 150 내지 250mM 염화나트륨 및 15 내지 25mM 이미다졸을 포함하는 수용액을 이용해서 침전물을 재현탁하고, 고압으로 균질화하거나 초음파로 세포를 파쇄한 후, 4℃ 환경하에 15,000 내지 18,000rpm으로 20 내지 40분간 원심 분리하는, 제조방법. - 청구항 1 또는 2에 기재된 재조합 IV형 인간화 콜라겐의 의료기기, 화장품, 건강식품 또는 의약품을 포함하는 제품 제조에서의 사용.
