Claims (6)
- 제 2항에 있어서,
상기 S10단계는,
Fmoc(9-플루오레닐메톡시카르보닐(9-fluorenylmethoxycarbonyl))을 Nα-아미노산의 보호기로 사용하는 고체상 합성법(Solid-Phase Peptide Synthesis; SPPS)에 의해 합성되되,
HOBt(N-히드록시벤조트리아졸)-DCC(N,N-디시클로헥실카보디이미드) 활성제를 사용하여 아미노산 잔기를 연장하여 이루어지는 것을 특징으로 하는 펩타이드 화합물의 제조방법.
- 제 2항에 있어서,
상기 S20단계는,
NH2-보호된-펩타이드(라이실-글루타밀-시스테이닐-글리신)-레진에 20% 피페리딘(Piperidine)/N-메틸피롤리돈 용액을 가하여 아미노기(-NH)에 붙어있는 Fmoc(9-플루오레닐메톡시카르보닐)기를 제거하는 S21단계와,
N-메틸피롤리돈과 디클로로메탄으로 세척 후 팔미틱산(Palmitic Acid, Aldrich 사)을 반응시키는 S22단계와,
커플링이 끝난 후 N-메틸피롤리돈과 디클로로메탄으로 여러 번 세척한 다음 건조시키는 S23단계와,
트리플루오로아세트산 : 페놀 : 티오아니솔 : 물 : 에탄디티올(82.5 : 5 : 5 : 5 : 2.5(v/v))의 혼합 용액으로 2~3시간 반응시켜 펩타이드를 구성하고 있는 아미노산의 보호기인 트리틸(트리페닐메틸)을 제거하는 S24단계와,
레진으로부터 팔미토일-트리펩타이드를 분리시킨 다음, 차가운 디에틸에테르로 펩타이드를 침전시켜 팔미토일-트리펩타이드를 수득하는 S25단계와,
팔미토일-트리펩타이드를 0.1% 트리플루오로아세트산이 포함된 물과 아세토니트릴을 용매로 하여 역상 고성능액체크로마토그래피 컬럼(reverse phase high performance liquid chromatography column, Gemini, C18 110A 250×21.2mm)을 이용하여 정제함으로써, 팔미토일-트리펩타이드를 합성하는 S26단계로 이루어지는 것을 특징으로 하는 펩타이드 화합물의 제조방법.
- 제 1항의 팔미토일-트리펩타이드 화합물을 함유하는 콜라겐 증식, 피부 주름 개선 및 상처 치유용 피부 외용제 조성물.
- 제 5항에 있어서,
상기 피부 외용제 조성물은 용액, 현탁액, 유탁액, 페이스트, 겔, 크림, 로션, 파우더, 비누, 계면활성제-함유 클렌징, 오일, 분말 파운데이션, 유탁액 파운데이션, 왁스 파운데이션 및 스프레이 등 어느 하나로 제형화되는 것을 특징으로 하는 피부 외용제 조성물.
