Description
Claims (12)
- (i) 하기 화학식 (II)로 표시되는 리바스티그민과 하기 화학식 (III)으로 표시되는 (+)-다이-O,O-p-톨루일-타르타르산 일수화물을 에틸아세테이트 및 에탄올을 포함하는 혼합용매 중에서 반응시키는 단계;(ii) 생성된 (S)-리바스티그민 (+)-다이-O,O-p-톨루일-타르타르산염을 결정화 용매에서 재결정하는 단계; 및(iii) 재결정된 (S)-리바스티그민 (+)-다이-O,O-p-톨루일-타르타르산염을 염기성 수용액과 반응시켜 (+)-다이-O,O-p-톨루일-타르타르산을 제거하는 단계를 포함하는 하기 화학식 (I)로 표시되는 (S)-리바스티그민의 제조방법:
- 제1항에 있어서, 화학식 (III)으로 표시되는 (+)-다이-O,O-p-톨루일-타르타르산 일수화물이 화학식 (II)로 표시되는 리바스티그민에 비해 0.4 당량 내지 0.7 당량으로 사용되는 것을 특징으로 하는 제조방법.
- 제2항에 있어서, 화학식 (III)으로 표시되는 (+)-다이-O,O-p-톨루일-타르타르산 일수화물이 화학식 (II)로 표시되는 리바스티그민에 비해 0.5 당량 내지 0.6 당량으로 사용되는 것을 특징으로 하는 제조방법.
- 제1항에 있어서, 혼합용매가 에틸아세테이트 및 에탄올이 4:1 내지 6:1의 부피비로 혼합된 용매인 것을 특징으로 하는 제조방법.
- 제4항에 있어서, 혼합용매가 에틸아세테이트 및 에탄올이 5:1의 부피비로 혼합된 용매인 것을 특징으로 하는 제조방법.
- 제1항에 있어서, 혼합용매가 아세톤을 추가로 포함하는 것을 특징으로 하는 제조방법.
- 제6항에 있어서, 혼합용매가 에틸아세테이트, 에탄올 및 아세톤이 3:1:1 내지 5:1:1의 부피비로 혼합된 용매인 것을 특징으로 하는 제조방법.
- 제7항에 있어서, 혼합용매가 에틸아세테이트, 에탄올 및 아세톤이 4:1:1의 부피비로 혼합된 용매인 것을 특징으로 하는 제조방법.
- 제1항에 있어서, 재결정 단계가 2회 내지 4회 반복되는 것을 특징으로 하는 제조방법.
- 제1항에 있어서, 결정화 용매가 메탄올, 에탄올, 아이소프로판올, 1-뷰탄올, 테트라하이드로퓨란, 디옥산, 아이소프로필에테르, 아세토나이트릴, 아세톤, 2-뷰탄온, 에틸아세테이트, 아이소프로필아세테이트, 다이클로로메탄, 클로로폼 및 1,2-다이클로로에탄으로 구성된 군으로부터 선택된 1 종 이상인 것을 특징으로 하는 제조방법.
- 제10항에 있어서, 결정화 용매가 에탄올인 것을 특징으로 하는 제조방법.
- 제1항에 있어서, 염기성 수용액이 수산화나트륨 수용액, 수산화칼륨 수용액, 수산화리튬 수용액 또는 암모니아수인 것을 특징으로 하는 제조방법.
