Claims (8)
- 치료용, 연구용 및 산업용 단백질을 위치선택적으로 원하는 위치에서 수식시켜서 수식된 단백질 변이체를 생성하는 방법으로서,수식해야 할 위치의 아미노산에 해당하는 코오돈을 돌연변이시켜서 앰버를 포함하는 중단 코오돈이나 희귀 코오돈으로 바꾸고,무세포 단백질 생산시 앰버를 포함하는 중단코오돈이나 희귀 코오돈에 해당하는 안티코오돈을 갖는 억제 tRNA를 첨가하여 억제 tRNA에 부착되어 있는 비천연성 아미노산을 단백질에 도입하여 단백질 변이체를 생성하고(여기서, 비천연성 아미노산에는 수식물질이 미리 부착되어 있을 수 있다),생성된 단백질 변이체의 원하는 위치에 위치선택적으로 수식물질을 부가시키는 것을 포함하는 방법.
- 제 1 항에 있어서, 수식물질은 피이지류, 단당류, 이당류, 올리고당류 및 다당류 등 변이체에 부가될 수 있는 천연, 비천연성 물질 중에서 선택되는 방법.
- 제 1 항에 있어서, 상기 단백질이 EPO인 것을 특징으로 하는 방법.
- 제 1 항 내지 제 3 항 중 어느 한 항의 방법에 따라 생성된 위치선택적으로 수식된 단백질 변이체.
- 치료용, 연구용 및 산업용 단백질을 위치 선택적으로 원하는 위치에서 수식시켜서 수식된 단백질 변이체를 생성하는 방법으로서,수식해야 할 위치의 아미노산에 해당하는 DNA상의 코오돈을 돌연변이시켜서 시스테인 아미노산에 해당하는 코오돈으로 바꾸고,상기 변이된 DNA로 숙주세포를 형질전환시켜서 자유로운 설프히드릴기를 갖는 시스테인이 도입된 변이체를 생성하고,생성된 변이체의 자유로운 설프히드릴기에 수식물질을 결합시켜서 변이체의 원하는 위치에 수식물질을 부가시키는 것을 포함하는 방법.
- 제 5 항에 있어서, 수식물질은 피이지류, 단당류, 이당류, 올리고당류 및 다당류 등 변이체에 부가될 수 있는 천연, 비천연성 물질 중에서 선택되는 방법.
- 제 5 항에 있어서, 상기 단백질이 EPO인 것을 특징으로 하는 방법.
- 제 5 항 내지 제 7 항 중 어느 한 항의 방법에 따라 생성된 위치 선택적으로 수식된 단백질 변이체.
