Abstract
Description
Claims (13)
- OB 단백질 성분의 N-말단에 면역글로불린의 Fc 부위를 결합시킴으로써 유도한, 약 6.0 내지 약 8.0 의 pH 및 적어도 0.5 mg/ml 의 농도를 갖는 인체 OB 단백질 현탁액.
- 제 1 항에 있어서, 상기 인체 Fc-OB 단백질의 Fc 부분은 다음 중에서 선택함을 특징으로 하는 인체 OB 단백질 현탁액 :(a) 서열 번호 : 2 및 4 에 나타나 있는 Fc 아미노산 서열 ;(b) 다음 위치 중 하나 또는 그 이상을 결실시키거나 다른 아미노산으로 치환시킨 (a) 항의 아미노산 서열(서열 번호 : 2 의 번호 체계 사용) :(ⅰ) 하나 또는 그 이상의 시스테인 잔기를 알라닌 또는 세린 잔기로 치환 ;(ⅱ) 하나 또는 그 이상의 티로신 잔기를 페닐알라닌 잔기로 치환 ;(ⅲ) 위치 5 의 아미노산을 알라닌으로 치환 ;(ⅳ) 위치 20 의 아미노산을 글루타민산으로 치환 ;(ⅴ) 위치 103 의 아미노산을 알라닌으로 치환 ;(ⅵ) 위치 105 의 아미노산을 알라닌으로 치환 :(ⅶ) 위치 107 의 아미노산을 알라닌으로 치환 ;(ⅷ) 위치 1, 2, 3, 4 및 5 의 아미노산을 결실 ;(ⅸ) Fc 수용체 결합 부위를 제거하기 위해 하나 또는 그 이상의 잔기를 치환하거나 결실 ;(ⅹ) 보체(C1q) 결합 부위를 제거하기 위해 하나 또는 그 이상의 잔기를 치환하거나 결실 : 및(xi) ⅰ - ⅹ 항의 조합 ; 및(c) N-말단에 메티오닌 잔기를 갖는 (a)항 또는 (b)항의 아미노산 서열.
- 제 1 항에 있어서, 상기 농도는 적어도 5 mg/ml 임을 특징으로 하는 인체 OB 단백질 현탁액.
- 제 1 항에 있어서, 상기 농도는 5 mg/ml 내지 50 mg/ml 임을 특징으로 하는 인체 OB 단백질 현탁액.
- 제 1 항에 있어서, 상기 pH 는 7.0 임을 특징으로 하는 인체 OB 단백질 현탁액.
- 제 2 항에 있어서, 상기 인체 OB 단백질은 rmetHu-Fc-렙틴임을 특징으로 하는 인체 OB 단백질 현탁액.
- 제 1 항에 있어서, 현탁액은 제약학적으로 용인가능한 침전제를 함유함을 특징으로 하는 인체 OB 단백질 현탁액.
- 제 1 항에 있어서, 현탁액은 염 및 수용성 중합체 중에서 선택한 침전제를 함유함을 특징으로 하는 인체 OB 단백질 현탁액.
- 제 1 항에 있어서, 현탁액은 칼슘, 마그네슘, 아연, 나트륨, 철, 코발트, 망간, 칼륨 및 니켈 중에서 선택한 침전제를 함유함을 특징으로 하는 인체 OB 단백질 현탁액.
- 제 1 항의 인체 OB 현탁액의 유효량을 투여함으로써 다음 중에서 선택한 증상을 갖는 개체를 치료하는 방법 :(a) 체중 조절,(b) 비만 조절,(c) 당뇨병,(d) 혈액내 지질 수준 조절,(e) 무지방 질량 증가 및(f) 인슐린 감수성 증가.
- 다음 단계를 포함하는 인체 OB 단백질 현탁액의 제조방법 :(ⅰ) OB 단백질 성분의 N-말단에 면역글로불린의 Fc 부위를 결합시킴으로써 유도한 용액내 인체 OB 단백질 및 침전제를 적당한 조건하에서 화합시키는 단계 ;(ⅱ) 상기 인체 OB 단백질을 침전시키는 단계 ;(ⅲ) 침전된 인체 OB 단백질을 모으는 단계 : 및(ⅳ) 선택적으로, 상기 인체 OB 단백질을 희석액 내로 재현탁시키는 단계.
- 제 11 항에 있어서, 상기 인체 OB 단백질을 pH 가 6.0 내지 8.0 인 희석액 내로 재현탁시킴을 특징으로 하는 제조방법.
- 제 11 항에 있어서, 상기 인체 OB 단백질은 rmetHu-Fc-렙틴임을 특징으로 하는 제조방법.
