Claims (17)
- 전극 상에 기능화된 표면을 제조하는 방법으로서:
a) 링커 및 염석 효과를 매개할 수 있는 적어도 하나의 중합체를, 상기 전극의 상기 표면 상에 상기 링커 및 상기 중합체의 공유 또는 비-공유 고정화를 허용하는 조건 하에서, 상기 전극에 적용하는 단계; 및
b) 상기 링커 및 상기 중합체의 고정화 시 적어도 하나의 검출제를, 고정된 상기 링커를 통해 적어도 하나의 검출제의 상기 전극에 대한 공유 또는 비-공유적 부착을 허용하는 조건 하에서, 상기 전극에 적용하는 단계를 포함하고,
상기 조건은 염석 효과를 매개할 수 있는 상기 적어도 하나의 중합체 및 상기 적어도 하나의 검출제를 상기 전극의 상기 표면 상에 분포시켜서, 상기 검출제가 전극 표면 전체의 표면적 당 동일한 양으로 존재하고, 염석 효과를 매개할 수 있는 상기 중합체가 상기 검출제의 주위에 배열되고 하기와 같은 양으로 존재하도록 하고:
i) 검출제에 근접한 유체의 이온 강도의 감소를 허용하는 양, 및
ii) 유체 샘플에 포함된 분석물을 상기 검출제에 결합시키는 것을 허용하는 양,
염석 효과를 매개할 수 있는 상기 적어도 하나의 중합체는 폴리(에틸렌 글리콜) (PEG), 폴리(에틸렌 옥사이드), 폴리(프로필렌 글리콜), 폴리글리세롤, 폴리아크릴아미드 (PAM), 폴리에틸렌이민 (PEI), 폴리메타크릴레이트 또는 또다른 아크릴 중합체, 폴리(비닐 알코올) (PVA), 폴리(비닐피롤리돈) (PVP), 다당류, 폴리실록산 및 상기 언급된 중합체의 공중합체로 이루어진 군으로부터 선택되는, 전극 상에 기능화된 표면을 제조하는 방법. - 제 1 항에 있어서, 상기 링커가 0.01 내지 5, 0.01 내지 1.0, 0.01 내지 0.5 또는 0.1 내지 0.5 kDa 의 MW 를 갖는 저분자량 (MW) 중합체이고, 상기 링커 중합체가 폴리(에틸렌 글리콜) (PEG), 폴리(에틸렌 옥사이드), 폴리(프로필렌 글리콜), 폴리글리세롤, 폴리아크릴아미드 (PAM), 폴리에틸렌이민 (PEI), 폴리메타크릴레이트 또는 또다른 아크릴 중합체, 폴리(비닐 알코올) (PVA), 폴리(비닐피롤리돈) (PVP), 다당류, 폴리실록산 및 언급된 중합체의 공중합체로 이루어진 군으로부터 선택되는, 전극 상에 기능화된 표면을 제조하는 방법.
- 제 1 항에 있어서, 염석 효과를 매개할 수 있는 상기 적어도 하나의 중합체가 폴리(에틸렌 글리콜) (PEG), 폴리(에틸렌 옥사이드), 폴리(프로필렌 글리콜), 폴리글리세롤, 폴리아크릴아미드 (PAM), 폴리에틸렌이민 (PEI), 폴리메타크릴레이트 또는 또다른 아크릴 중합체, 폴리(비닐 알코올) (PVA), 폴리(비닐피롤리돈) (PVP) 및 언급된 중합체의 공중합체로 이루어진 군으로부터 선택되는, 전극 상에 기능화된 표면을 제조하는 방법.
- 제 2 항에 있어서, 염석 효과를 매개할 수 있는 중합체가 링커 중합체와 동일한 부류의 중합체인, 전극 상에 기능화된 표면을 제조하는 방법.
- 제 4 항의 방법에 의해 얻어지는 전극으로서, 유체 샘플에 노출될 수 있는 기능화된 표면을 포함하고, 상기 기능화된 표면은 염석 효과를 매개할 수 있는 적어도 하나의 중합체 및 유체 샘플에 포함된 분석물에 결합하는 적어도 하나의 검출제를 포함하며, 염석 효과를 매개할 수 있는 상기 적어도 하나의 중합체 및 상기 적어도 하나의 검출제는 상기 전극의 표면에 분포되어, 상기 검출제가 전극 표면 전체의 표면적 당 동일한 양으로 존재하고, 상기 염석 효과를 매개할 수 있는 중합체가 상기 검출제 주위에 배열되고 하기와 같은 양으로 존재하도록 하고:
i) 검출제에 근접한 유체의 이온 강도의 감소를 허용하는 양, 및
ii) 유체 샘플에 포함된 분석물을 상기 검출제에 결합시키는 것을 허용하는 양,
염석 효과를 매개할 수 있는 상기 적어도 하나의 중합체는 폴리(에틸렌 글리콜) (PEG), 폴리(에틸렌 옥사이드), 폴리(프로필렌 글리콜), 폴리글리세롤, 폴리아크릴아미드 (PAM), 폴리에틸렌이민 (PEI), 폴리메타크릴레이트 또는 또다른 아크릴 중합체, 폴리(비닐 알코올) (PVA), 폴리(비닐피롤리돈) (PVP), 다당류, 폴리실록산 및 상기 언급된 중합체의 공중합체로 이루어진 군으로부터 선택되고,
상기 적어도 하나의 검출제가 링커를 통해 전극의 표면에 고정되며, 상기 링커가 0.01 내지 5, 0.01 내지 1.0, 0.01 내지 0.5 또는 0.1 내지 0.5 kDa 의 MW 를 갖는 저분자량 (MW) 중합체로 이루어진 군으로부터 선택되고, 상기 링커 중합체가 폴리(에틸렌 글리콜) (PEG), 폴리(에틸렌 옥사이드), 폴리(프로필렌 글리콜), 폴리글리세롤, 폴리아크릴아미드 (PAM), 폴리에틸렌이민 (PEI), 폴리메타크릴레이트 또는 또다른 아크릴 중합체, 폴리(비닐 알코올) (PVA), 폴리(비닐피롤리돈) (PVP), 다당류, 폴리실록산 및 언급된 중합체의 공중합체로 이루어진 군으로부터 선택되고,
염석 효과를 매개할 수 있는 중합체가 링커 중합체와 동일한 부류의 중합체인, 전극. - 제 5 항에 있어서, 염석 효과를 매개할 수 있는 상기 적어도 하나의 중합체의 크기는 Debye 길이가 증가되고 적어도 하나의 검출제 주위의 이온 강도가 감소되도록 하는 것인, 전극.
- 제 6 항에 있어서, 염석 효과를 매개할 수 있는 상기 적어도 하나의 중합체의 분자량 (MW) 이 1 내지 100 kDa, 10 내지 100 kDa, 10 내지 50 kDa, 10 내지 25 kDa, 10 내지 20 kDa 또는 10 내지 15 kDa 인, 전극.
- 제 5 항에 있어서, 염석 효과를 매개할 수 있는 상기 적어도 하나의 중합체가 폴리(에틸렌 글리콜) (PEG), 폴리(에틸렌 옥사이드), 폴리(프로필렌 글리콜), 폴리글리세롤, 폴리아크릴아미드 (PAM), 폴리에틸렌이민 (PEI), 폴리메타크릴레이트 또는 또다른 아크릴 중합체, 폴리(비닐 알코올) (PVA), 폴리(비닐피롤리돈) (PVP) 및 언급된 중합체의 공중합체로 이루어진 군으로부터 선택되는, 전극.
- 제 5 항에 있어서, 염석 효과를 매개할 수 있는 상기 적어도 하나의 중합체가 링커를 통해 전극의 표면에 고정되거나 추가의 링커없이 작용기를 통해 직접 부착되는, 전극.
- 제 9 항에 있어서, 염석 효과를 매개할 수 있는 적어도 하나의 중합체를 고정하는 상기 링커가 하기 구조를 갖는 링커인, 전극:
여기서 A 는 하기 부류: 티올, 실란, 포스폰산, 방향족 분자, 카르복실, 아민, NHS 에스테르, 말레이미드로부터의 제 1 작용기이고,
B 는 하기 부류: 상기 기재된 부류로부터의 저분자량 중합체, 탄화수소, 산소 함유 기, 할로알칸, 질소 함유 기, 황 함유 기, 인 함유 기, 붕소 함유 기, 또는 짧은 무기 중합체 사슬로부터의 짧은 유기 사슬이고,
C 는 하기 부류: 카르복실, 아민, NHS 에스테르, 말레이미드로부터의 제 2 작용기임. - 제 5 항에 있어서, 상기 적어도 하나의 검출제가 유체 샘플에 포함된 분석물에 특이적으로 결합하는, 전극.
- 제 5 항에 있어서, 상기 적어도 하나의 검출제가, 항체 및 그의 단편, 핵산, 앱타머, 펩티드 핵산 (PNA) 수용체 또는 리간드 단백질 또는 펩티드, 및 효소로 이루어진 군으로부터 선택되는, 전극.
- 제 5 항에 있어서, 적어도 하나의 검출제의 크기는 검출제가 염석 효과를 매개할 수 있는 적어도 하나의 중합체에 의해 둘러싸이는 정도의 것인, 전극.
- 제 5 항 내지 제 13 항 중 어느 한 항의 전극을 포함하는 적어도 하나의 분석물을 측정하기 위한 분석물 검출기로서, 상기 전극이 변환기와 전기적으로 접촉하거나 또는 변환기의 일부인 분석물 검출기.
- 제 14 항에 있어서, 유체 샘플에서 적어도 하나의 분석물을 측정하기 위해 사용되는, 분석물 검출기.
- 하기 단계를 포함하는, 적어도 하나의 분석물을 측정하는 방법:
(a) 적어도 하나의 분석물을 포함하는 것으로 의심되는 유체 샘플을, 제 5 항 내지 제 13 항 중 어느 한 항의 전극, 또는 제 5 항 내지 제 13 항 중 어느 한 항의 전극을 포함하는 적어도 하나의 분석물을 측정하기 위한 분석물 검출기로서, 상기 전극이 변환기와 전기적으로 접촉하거나 또는 변환기의 일부인 상기 분석물 검출기와 접촉시키는 단계; 및
(b) 상기 전극 또는 검출기로 전기화학적 측정을 수행함으로써, 적어도 하나의 분석물이 결정될 것인 단계. - 제 16 항에 있어서, 상기 전기화학적 측정이 30℃ 내지 40℃ 의 온도에서 수행되는, 적어도 하나의 분석물을 측정하는 방법.
