Abstract
Description
도2는 3종의 HER2 발현세포를 대상으로 HER2에 결합하는 어피바디들의 세포 결합 능력을 확인하고, MFI 값을 그래프화 한 도이다(A. NCI-N87, B. SK-OV-3, C. MDA-MB-231).
도3은 HER2에 결합하는 5종의 어피바디를 Fc 결합 형태로 생산하여, HER2 단백질의 결합 정도를 정량적으로 비교 확인한 도이다.
도4는 HER2에 결합하는 5종의 어피바디를 Fc 결합 형태로 생산하여, HER2 발현세포 4종의 세포에 대한 결합 정도를 정량적으로 분석한 도이다(A. NCI-N87, B. SK-OV-3, C. MDA-MB-453, D. MDA-MB-231).
도 5는 코티닌이 결합된 어피바디 5종의 HER2 단백질 결합 정도를 정량적으로 확인한 도이다.
도 6은 HER2 양성 세포인 SK-OV-3와 HER2 음성 세포인 Raji 세포에 대해 코티닌이 결합된 어피바디와 항 코티닌 항체를 이용한 키메릭항원수용체 T세포(Cot-sCART)의 세포독성효과를 측정한 도이다.
도 7은 면역결핍 마우스(NSG)에 SKOV3-Luc세포주로 질환모델을 형성한 후, 해당 모델에 대한 코티닌이 결합된 어피바디와 Cot-sCART의 효능을 측정한 도이다(A. Luminescence 측정 이미지, B. Luminescence signal 정량 자료).
도 8는 SKOV3-Luc 세포주로 형성된 질환모델에 대한 코티닌이 결합된 어피바디와 Cot-sCART의 효능 평가에서 코티닌이 결합된 어피바디의 농도에 따른 효능을 확인한 도이다(A. Luminescence 측정 이미지, B. Luminescence signal 정량 자료).
도9은 친화도가 향상된 클론들을 Fc결합 형태로 생산하여, HER2 단백질의 결합 정도를 정량적으로 비교 확인한 도이다.
도10은 코티닌이 결합된 최적화된 어피바디들의 HER2 단백질 결합 정도를 정량적으로 확인한 도이다.
도 11는 코티닌이 결합된 최적화된 어피바디들의 HER2 발현세포인 SK-OV-3에 대한 결합능을 측정한 도이다.
도 12은 HER2 양성 세포인 SK-OV-3에 대해 코티닌이 결합된 최적화된 어피바디와 Cot-sCART의 세포독성효과를 측정한 도이다.

Claims (16)
- 코티닌과 결합된, 서열번호 1, 3, 및 6 내지 8 중 어느 하나의 아미노산 서열로 표시되는 항-HER2 어피바디를 포함하는, 키메라 항원 수용체 효과기 세포(chimeric antigen receptor-effector cell)를 활성화하기 위한 스위치 분자(switch molecule).

- 제1항에 있어서, 상기 효과기 세포의 활성화는 표적세포에 대한 세포독성, 사이토카인 분비, 및 이들의 조합인 것을 특징으로 하는 스위치 분자.
- 제1항에 있어서, 상기 효과기 세포는 수지상 세포, 킬러 수지상 세포, 비만세포, 자연살해 세포, B 림프구, T 림프구, 대식세포 및 이들의 전구세포로 이루어진 군으로부터 선택되는 것을 특징으로 하는 스위치 분자.
- 제1항, 제8항 및 제9항 중 어느 한 항의 스위치 분자; 및 약제학적으로 허용되는 담체를 포함하는 암세포 표면에 HER2를 발현하는 암의 치료용 약제학적 조성물.
- 다음을 포함하는 스위처블 키메라 항원 수용체:
(a) 제1항, 제8항 및 제9항 중 어느 한 항의 스위치 분자; 및
(b) 다음을 포함하는 키메라 항원 수용체:
i) 제1항, 제8항 및 제9항 중 어느 한 항의 스위치 분자를 표적화하는 항체 또는 그의 항원 결합 단편을 포함하는 세포외 도메인(extracellular domain);
ii) 막관통 도메인(transmembrane domain); 및
iii) 세포내 신호전달 도메인.
- 제11항에 있어서, 상기 막관통 도메인은 T-세포 수용체, CD27, CD28, CD3, 엡실론, CD45, CD4, CD5, CD8(CD8α), CD9, CD16, CD22, CD33, CD37, CD64, CD80, CD86, CD134, CD137 및 CD154의 알파, 베타 또는 제타 사슬로 이루어진 군으로부터 선택된 단백질의 막관통 도메인인, 스위처블 키메라 항원 수용체.
- 제11항에 있어서, 상기 세포내 신호전달 도메인은 CD3ζ(CD3 제타) 사슬로부터 유래된 도메인인, 스위처블 키메라 항원 수용체.
- 제11항에 있어서, 상기 세포내 신호전달 도메인은 OX40 (CD134), CD2, CD27, CD28, CDS, ICAM-1, LFA-1(CD11a/CD18) 및 4-1BB (CD137)로 이루어진 군으로부터 선택된 공동자극 분자(costimulatory molecule)를 추가적으로 포함하는 것인, 스위처블 키메라 항원 수용체.
- 제11항에 있어서, 상기 i)의 항체 또는 그의 항원 결합 단편은 항-코티닌 항체 또는 그의 항원 결합 단편인, 스위처블 키메라 항원 수용체.
- 제11항에 있어서, 상기 i)의 항체 또는 그의 항원 결합 단편은 서열번호 9의 아미노산 서열을 포함하는 HCDR1, 서열번호 10의 아미노산 서열을 포함하는 HCDR2, 서열번호 11의 아미노산 서열을 포함하는 HCDR3, 서열번호 12의 아미노산 서열을 포함하는 LCDR1, 서열번호 13의 아미노산 서열을 포함하는 LCDR2, 및 서열번호 14의 아미노산 서열을 포함하는 LCDR3를 포함하는 것인, 스위처블 키메라 항원 수용체.
