Description
도 2는 본 발명의 일 실시예에 따른 외상성 뇌손상 바이오마커 검출용 시분해 형광분석 측방유동 분석장치를 이용한 외상성 뇌손상 바이오마커 측정방법을 설명하기 위한 플로우차트이다.
도 3은 다양한 GFAP 음성 혈장 샘플에 대하여 마우스 유래 항체를 사용하는 경우와 마우스와 토끼 유래 항체를 같이 사용하는 경우의 비특이 반응 감소를 확인하는 그래프이다.
도 4는 GFAP 음성 혈장 투입시, 마우스 및 토끼 유래 항체 쌍의 조합에 따른 비특이 현상을 나타내는 측방 유동 분석 센서 사진이다.
도 5는 마우스 및 토끼 유래 항체 쌍의 조합에 따른 GFAP 농도별 형광 신호 차이를 나타내는 그래프이다.

Claims (10)
- 외상성 뇌손상 바이오마커를 검출할 수 있는 시분해 형광분석 측방유동 분석장치에 있어서,
외상성 뇌손상 바이오마커를 함유한 혈액 샘플이 주입되는 샘플패드와,
상기 샘플패드로부터 외상성 뇌손상마커가 이동할 때 상기 마커에 혼합되어 외상성 뇌손상 바이오마커 복합체를 형성하는 프로브를 포함하는 흡착패드와,
상기 흡착패드에 유체 연통하며 상기 흡착패드로부터 외상성 뇌손상 바이오마커 복합체를 검출라인까지 모세관 이동시키는 다공성막을 포함하며,
상기 프로브에는 상기 외상성 뇌손상 바이오마커와 특이적으로 결합하는 특이적 결합물질이 표지된 항체로 이루어진 포획항체와, 1 마이크로초 이상의 비교적 긴 방출 수명을 갖는 형광물질이 표지된 항체로 이루어진 검출항체를 포함하며,
상기 외상성 뇌손상 바이오마커는 신경교 섬유질 산성 단백질(GFAP)이며, 상기 특이적 결합물질은 바이오틴이 사용되며, 상기 형광물질로 유로퓸이 사용되고, 상기 검출항체용 항체로 마우스 유래 항체와 토끼 유래 항체의 혼합물이 사용되는
외상성 뇌손상 바이오마커 검출용 시분해 형광분석 측방유동 분석장치.
- 제 1 항에 있어서,
상기 흡착패드는 상기 프로브가 순차적으로 제공되는 제 1 및 제 2 흡착패드를 포함하며, 상기 제 1 흡착패드는 상기 포획항체를 형성하는 특이적 결합물질을 제공하며, 상기 제 2 흡착패드는 상기 검출항체를 형성하는 형광물질을 제공하는 외상성 뇌손상 바이오마커 검출용 시분해 형광분석 측방유동 분석장치.
- 제 1 항에 있어서,
상기 특이적 결합물질 또는 형광물질이 표지된 항체에 대하여 적어도 2 이상의 다른 종 유래 항체의 혼합물을 사용하는
외상성 뇌손상 바이오마커 검출용 시분해 형광분석 측방유동 분석장치.

- 제 7 항에 있어서,
상기 포획항체로 토끼 유래 항체를 사용하여 외상성 뇌손상 바이오마커 중 하나인 GFAP 에 대하여 100 pg/mL 미만의 민감도로 검출하는 외상성 뇌손상 바이오마커 검출용 시분해 형광분석 측방유동 분석장치.
- 제 1 항, 제 2 항, 제 3 항, 제 6 항 내지 제 8 항중 어느 한 항에 따른 외상성 뇌손상 바이오마커 검출용 시분해 형광분석 측방유동 분석장치를 이용한 외상성 뇌손상 바이오마커 측정방법에 있어서,
외상성 뇌손상 바이오마커를 함유하는 혈액 시료를 준비하는 단계와,
외상성 뇌손상 바이오마커를 함유하는 혈액 시료를 샘플패드에 주입하는 단계와,
상기 샘플패드에 인접한 흡착패드를 따라 외상성 뇌손상 바이오마커를 함유하는 혈액 시료를 모세관 현상으로 이동시키면서 특이적 결합물질과 형광물질이 각각 표지된 포획항체와 검출항체가 결합된 외상성 뇌손상 바이오마커 복합체를 형성하는 단계와
상기 외상성 뇌손상 바이오마커 복합체를 상기 흡착패드에 유체 연통하는 다공성 막을 따라 상기 다공성 막상의 검출라인상의 포획물질과 결합시키는 단계와,
상기 외상성 뇌손상 바이오마커에 결합하지 않는 임의의 프로브를 상기 검출라인을 통과시켜 교정라인의 포획물질과 결합시키는 단계와,
