Abstract
Description
도 2 는 본 발명에 따른 화합물의 제조예에서 제조된 화합물 2의 LC-MS 스펙트럼이다.
도 3a 는 본 발명에 따른 화합물의 제조예에서 제조된 화합물 3의 NMR 스펙트럼이고, 도 3b 는 상기 화합물 3의 HR-FAB-MS 스펙트럼이다.
도 4 는 본 발명에 따른 화합물의 제조예에서 제조된 화합물 4의 HR-FAB-MS 스펙트럼이다.
도 5a 는 본 발명에 따른 화합물의 제조예에서 제조된 화합물 Gd-RB 의 HR-ESI-MS 스펙트럼이고, 도 5b 는 상기 화합물 Gd-RB 의 HPLC 크로마토그램이다.
도 6a 는 대조군(control), 본 발명에 따른 화합물 (Gd-RB8), 및 상용 조영제인 Gadovist를 사용했을 때 얻어진 MR 팬텀(Phantom) 이미지이고, 도 6b는 본 발명에 따른 화합물Gd-RB 및 상용 조영제인 Gadovist 각각의 사용 농도에 따른, 팬텀 신호 강도를 보여주는 그래프이다.
도 7a는 면역형광 염색법에 의한 in vitro 결합 시험에서의 형광 이미지를 나타낸 도면이고, 도 7b는 화합물 Gd-RB의 농도에 따른 형광 강도를 보여주는 그래프이다.
도 8a는 in vivo 동물 실험을 통해 얻은 뇌 MR 영상을 “후-전”처리 이후에 컬러맵핑하여 나타낸 도면이고, 도 8b는 in vivo 결합 시험 후 동물모델에서 적출한 뇌를 면역형광염색하여 얻은 이미지이다.
도 9는 화합물 Gd-RB와 Gadovist 각각의 농도에 따른 세포 생존도를 나타낸 그래프이다.

Claims (10)
- 하기 화학식 1로 표시되는 것을 특징으로 하는 가돌리늄계 화합물;

상기 화학식 1에서,
A는 *-(CH2)n-A1-*를 나타내고,
n은 0 내지 5 중 임의 정수를 나타내고,
A1 은 *-COO-*, *-CO-*, *-NR1-*, *-CH2-*, *-CONH-*, 또는 *-O-*를 나타내고,
Linker는 *-L1-NHCO-L2-*, *-L1-O-R2-O-L2-*, *-L1-CH2-L2-*, *-L1-NR3-L2-*, 또는 *-L1-COO-L2-*를 나타내고,
L1 및 L2 는 각각 독립적으로, 선형 또는 분지형의 (C1-C30)알킬을 나타내고,
R1 및 R3은 각각 독립적으로, 수소 또는 선형 또는 분지형의 (C1-C10)알킬을 나타내고,
R2는 선형 또는 분지형의 (C1-C20)알킬을 나타내고,
RB는 하기 화학식 2를 나타내고:

- 제1항에 있어서, n은 1 내지 5 중 임의 정수를 나타내고, A1 은 *-CONH-*을 나타내는 것을 특징으로 하는 가돌리늄계 화합물.
- 제1항에 있어서, L1 는 선형 또는 분지형의 (C1-C10)알킬을 나타내고, L2 는 선형 또는 분지형의 (C2-C20)알킬을 나타내는 것을 특징으로 하는 가돌리늄계 화합물.
- 제1항에 있어서, 상기 가돌리늄이 적어도 1 이상의 물 분자와 배위하는 것을 특징으로 하는 가돌리늄계 화합물.
- 제1항에 있어서, 포유동물의 소포성 글루타메이트 수송체(vesicular glutamate transporter)(VGLUT)에 특이적으로 결합하는 것을 특징으로 하는 가돌리늄계 화합물.
- 제1항에 따른 가돌리늄계 화합물을 함유하는 것을 특징으로 하는 MRI 조영제.
- 제6항에 있어서, 퇴행성 뇌질환의 진단에 사용되는 것을 특징으로 하는 MRI 조영제.
- 제7항에 있어서, 알츠하이머병의 진단에 사용되는 것을 특징으로 하는 MRI 조영제.
- (a) 로즈 벵갈의 염을 할로겐-치환된 알카노산(halogen-substituted alkanoic acid)와 함께 반응시켜, 하기 화학식 1-1의 화합물을 수득하는 단계;

L2 는 제1항에서 정의한 바와 같다.
(b) 상기 화학식 1-1의 화합물을 하기 화학식 1-2의 화합물과 반응시켜, 하기 화학식 1-3의 화합물을 수득하는 단계:

상기 화학식 1-2 및 1-3에서,
PT는 보호기를 나타내고,
L1, A, Linker, 및 RB는 제1항에서 정의한 바와 같다.
(c) 상기 화학식 1-3의 화합물에서 보호기(PT)를 제거하는 단계, 및
(d) 상기 (c) 단계에서 수득한 화합물을 가돌리늄 수화물과 반응시켜 제1항에 기재된 화학식 1의 화합물을 수득하는 단계를 포함하는, 제1항에 따른 가돌리늄계 화합물의 제조 방법.
- 제9항에 있어서, 상기 (a) 단계에서, 상기 로즈 벵갈의 염은 로즈 벵갈의 알칼리 금속 염이고, 상기 할로겐-치환된 알카노산은 선형 또는 분지형의 (C1-C30)알칸에서 유래되고 1개의 할로겐에 의해 치환된 알카노산인, 제조 방법.
