Abstract
Description
도 3은 염료의 첨가시에 아밀로이드 베타 단백질에 결합하는 것과 어드미턴스가 변하는 것을 나타낸 것이다.
도 4는 혈액 플라즈마 시료를 사용하여 본 발명의 일 구현예에 따른 방법에 따라 측정한 결과를 나타낸 것이다.
도 5는 검출소자의 전극 패턴의 실시예와 패턴 사이에 항체가 고정되어 있는 전극 단면을 나타낸다.
Claims (17)
- 아밀로이드 베타 항체에 액상 시료를 가하는 단계;
액상에서 전하를 띠고, 아밀로이드 베타 단백질에 특이적으로 결합하는 염료를 상기 시료가 가해진 항체에 가하는 단계; 및
상기 염료를 가한 후 액상에서의 전기적 신호의 변화를 측정하는 단계;를 포함하는 아밀로이드 베타 올리고머의 검출 방법.
- 청구항 1에 있어서, 상기 염료는 티오플라빈 T, 티오플라빈 S, 콩고 레드, BSB((trans,trans)-1-Bromo-2,5-bis-(3-hydroxycarbonyl-4-hydroxy)styrylbenzene), Chrysamine G, 1,4-bis(3-carboxy-4-hydroxy phenylethenyl)-benzene), ISB((E,E)-1-iodo-2,5-bis(3-hydroxycarbonyl-4-hydroxy)styrylbenzene), IMSB(1-iodo-2,5-bis-(3-hydroxycarbonyl-4-methoxy)styrylbenzene), PiB(Pittsburgh Compound B), FDDNP(2-(1-{6-[(2-[fluorine-18]fluoroethyl)(methyl)amino]-2-naphthyl}-ethylidene)malononitrile) 또는 IMPY(6-iodo-2 -(4´-dimethylamino)-phenyl-imidazo[1,2-a]pyridine)인, 방법.
- 청구항 1에 있어서, 상기 염료가 아밀로이드 베타 단백질에 결합하여 발생하는 전기적 신호 변화를 측정하여, 그 변화 정도가 클수록 액상 시료에 아밀로이드 베타 올리고머가 많이 포함된 것으로 판단하는, 방법.
- 청구항 1에 있어서, 상기 염료를 가한 후부터 전기적 신호 변화 완료시점까지 전기적 신호를 실시간으로 측정하는, 방법.
- 청구항 1에 있어서, 상기 전기적 신호는 임피던스, 어드미턴스, 저항, 전도도, 위상, 리액턴스 및 서셉턴스로 이루어진 군에서 선택된 적어도 하나인, 방법.
- 청구항 1에 있어서, 상기 항체는 웰 플레이트 상에 고정된 것인, 방법.
- 청구항 1에 있어서, 상기 항체는 자성 마이크로 비드 상에 고정된 것인, 방법.
- 청구항 1에 있어서, 상기 항체는 교차전극 사이에 구비된 것인, 방법.
- 청구항 1에 있어서, 상기 항체에 결합된 아밀로이드 베타 올리고머를 해리시키는 단계; 및
상기 해리 전, 후의 전기적 신호를 측정하여 비교하는 단계;를 더 포함하는, 방법.
- 청구항 9에 있어서, 상기 해리 전 후의 전기적 신호의 차이가 클수록 시료에 아밀로이드 베타 올리고가 많이 포함된 것으로 판단하는, 방법.
- 액상에서 전하를 띠는 염료가 결합되고, 항체에 결합된 아밀로이드 베타 올리고머를 액상에서 해리시키는 단계; 및
상기 해리 전, 후의 액상에서의 전기적 신호를 측정하여 비교하는 단계;를 포함하는 아밀로이드 베타 올리고머의 검출 방법.
- 청구항 11에 있어서, 상기 염료는 티오플라빈 T, 티오플라빈 S, 콩고 레드, BSB((trans,trans)-1-Bromo-2,5-bis-(3-hydroxycarbonyl-4-hydroxy)styrylbenzene), Chrysamine G, 1,4-bis(3-carboxy-4-hydroxy phenylethenyl)-benzene), ISB((E,E)-1-iodo-2,5-bis(3-hydroxycarbonyl-4-hydroxy)styrylbenzene), IMSB(1-iodo-2,5-bis-(3-hydroxycarbonyl-4-methoxy)styrylbenzene), PiB(Pittsburgh Compound B), FDDNP(2-(1-{6-[(2-[fluorine-18]fluoroethyl)(methyl)amino]-2-naphthyl}-ethylidene)malononitrile) 또는 IMPY(6-iodo-2 -(4´-dimethylamino)-phenyl-imidazo[1,2-a]pyridine)인, 방법.
- 청구항 11에 있어서, 상기 전기적 신호는 임피던스, 어드미턴스, 저항, 전도도, 위상, 리액턴스 및 서셉턴스로 이루어진 군에서 선택된 적어도 하나인, 방법.
