Abstract
Description
도 2는 본 발명에 따른 유잔자 회로 및 이를 적용한 대장균에서 낮은 pH-의존성 자일론산의 생산 결과를 나타낸다. 도 2a는 lacIq 프로모터의 하류에 있는 cadC ΔF159-165 로 구성된 자일로오스 산화 경로에서 조작된 발현 벡터(pH 센서)를 도식화한 것으로, T7 프로모터 하류에는 자일론산 생산 유전자인 xylB CC (xylose dehydrogenase) 및 xyl C (xylonolactonase)가 위치하고, lacI 유전자(LacI 리프레서 코딩 유전자)는 cadBA 프로모터의 하류에 위치한다. 도 2b 및 도2c는 발현 벡터(pH 센서)가 없는 BLXA 균주 및 발현 벡터(pH 센서)가 있는 BLXAPH 균주의 자일론산 생산 결과를 나타낸 것이다.
도 3은 xylose oxidative pathway(XOP)에서 발현 벡터(pH 센서)에 의한 자일론산 축적의 조절을 보여주는 것으로, 도 3a 및 3b는 자일로오스 탈수소효소의 pH 수준 및 효소 활성을 나타내는 결과이고, 도 3c 및 도3d는 균주의 자일로오스 소모, 자일론산 축적 및 바이오매스 생성 결과이다.
도 4a는 배지의 pH 레벨을 나타내는 것이고, 도 4b는 효소 활성나타내며, 도 4c 및 도 4d는 EWXA and EWXAPH에서 자일론산 소모 및 자일론산 축적을 확인한 결과이다.
도 5는 낮은 pH 조절을 통한 에틸렌 글리콜 생산 결과를 나타내는 것으로, 도 5a는 발현 벡터(pH 센서)에 의해 조절되는 에틸렌 글리콜 생합성 과정을 도식화한 것이고, 도 5b는 대조 균주 EWEG 및 EWEGPH에 따른 배지의 pH를 나타내는 결과이고, 도5c 및 도 5d는 EWEGPH 및 EWEG 균주에서의 대사 산물 농도를 나타낸다.

Claims (9)
- i) pH 센서로서 CadC 단백질의 염기서열인 서열번호 1에서 475bp 내지 495bp가 결실된 서열번호 2의 염기서열로 표시되는 유전자 및 상기 서열번호 2의 염기서열로 표시되는 유전자의 발현을 조절하는 프로모터;
ii) 락토오스 단백질을 코딩하는 lacI 유전자 및 상기 lacI 유전자의 발현을 조절하는 프로모터; 및
iii) 자일로오스 탈수소효소를 코딩하는 xylBcc 유전자 및 자일로노락토네이즈를 코딩하는 xylC 유전자; 및 락토오스 단백질의 발현에 따라 하류의 xylBcc 유전자 및 xylC 유전자의 발현을 조절하는 프로모터; 를 함유하고,
상기 i)에서 유전자의 발현을 조절하는 프로모터는 프로모터 lacIq이고, ii)에서 유전자의 발현을 조절하는 프로모터는 cadBA이며, iii)에서 유전자의 발현을 조절하는 프로모터는 프로모터 T7인 것을 특징으로 하는 D-자일로오스 대사 산물을 생산하기 위한 발현 벡터.
- 삭제
- 제1항에 있어서,
상기 D-자일로오스 대사 산물은 에틸렌글리콜, 글리콜산, 1,2,4-부탄트리올, 1,4-부탄디올, 에탄올 및 옥살로글루타르산으로 구성된 군에서 선택되는 것을 특징으로 하는 발현 벡터.
- 제1항의 발현 벡터로 형질 전환된 D-자일로오스 대사 산물을 생산하기 위한 형질전환체.
- 제4항에 있어서,
상기 형질전환체는 대장균(Escherichia coli)인 것을 특징으로 하는 형질전환체.
- 제4항에 있어서,
상기 D-자일로오스 대사 산물은 에틸렌글리콜, 글리콜산, 1,2,4-부탄트리올, 1,4-부탄디올, 에탄올 및 옥살로글루타르산으로 구성된 군에서 선택되는 것을 특징으로 하는 형질전환체.
- 제1항의 발현 벡터로 형질 전환된 형질전환체를 포함하는 D-자일로오스 대사 산물을 생산하기 위한 배지의 pH를 감지하기 위한 바이오 센서.
- 제7항에 있어서,
상기 바이오 센서는 배지의 pH가 5.5 내지 6.5 이하로 감소하는 것을 방지하고, 배지의 pH를 5.5 내지 6.5로 유지하는 것을 특징으로 하는 바이오 센서.
- 제7항에 있어서,
상기 D-자일로오스 대사 산물은 에틸렌글리콜, 글리콜산, 1,2,4-부탄트리올, 1,4-부탄디올, 에탄올 및 옥살로글루타르산으로 구성된 군에서 선택되는 것을 특징으로 하는 바이오 센서.
