Abstract
Description
도 2 내지 4는 본 발명의 세포 투과성 펩티드 이량체의 개선된 세포 투과 효과를 시각적으로 설명하는 도식이고;
도 5는 본 발명의 세포 투과성 펩티드 이량체를 제조한 결과를 나타낸 사진이고;
도 6은 발명의 카고 컨쥬게이션된 세포 투과성 펩티드 이량체의 세포 내 세포투과능을 면역세포화학으로 확인한 사진이고;
도 7은 다양한 링커를 사용한 본 발명의 카고 컨쥬게이션된 세포 투과성 펩티드 이량체의 세포투과능을 비교한 그래프를 나타낸다.
Claims (17)
- 서열번호 1의 아미노산 서열로 구성되는 제1 펩티드 도메인; 서열번호 1의 아미노산 서열로 구성되는 제2 펩티드 도메인; 및 상기 제1 및 제2 펩티드 도메인을 연결하는 펩티드 링커를 포함하는 것을 특징으로 하는 융합 단백질인 세포 투과성 펩티드 이량체로서, 상기 펩티드 링커는 상기 제1 펩티드 도메인의 카복시-말단과 상기 제2 펩티드 도메인의 아미노-말단을 서로 연결하고, 100개 이하의 아미노산으로 구성되는 신축성(flexible) 링커 또는 경직성(rigid) 링커인, 세포 투과성 펩티드 이량체.
- 삭제
- 제 1 항에 있어서, 펩티드 링커는 글리신(G) 및 세린(S)을 포함하는 것을 특징으로 하는 신축성 링커인 세포 투과성 펩티드 이량체.
- 제 3 항에 있어서, 펩티드 링커는 글리신(G) 및 세린(S)으로 구성된 아미노산 서열 반복 단위를 포함하는 신축성 링커인 세포 투과성 펩티드 이량체.
- 제 1 항에 있어서, 펩티드 링커는 EAAAK의 아미노산 서열 반복 단위를 포함하는 경직성 링커인 세포 투과성 펩티드 이량체.
- 제 1 항에 있어서, 펩티드 링커는 서열번호 2 내지 8 중 어느 하나의 아미노산 서열로 구성되는 것인 세포 투과성 펩티드 이량체.
- 제 1 항에 있어서, 카고가 컨쥬게이션될 수 있는 세포 투과성 펩티드 이량체.
- 제 1 항 및 제 3 항 내지 제 7 항 중 어느 한 항에 따른 세포 투과성 펩티드 이량체를 코딩하는 핵산.
- 제 8 항에 있어서, 펩티드 링커를 코딩하는 핵산은 제1 펩티드 도메인을 코딩하는 핵산의 3'-말단과 상기 제2 펩티드 도메인을 코딩하는 핵산의 5'-말단을 서로 연결하는 것인 핵산.
- 제 9 항에 있어서, 펩티드 또는 단백질인 카고를 코딩하는 핵산을 더 포함하고, 여기서 상기 카고를 코딩하는 핵산은 제1 펩티드 도메인을 코딩하는 핵산의 5'-말단 또는 제2 펩티드 도메인을 코딩하는 핵산의 3'-말단에 연결되는 것인 핵산.
- 제 8 항에 따른 핵산을 포함하는 재조합 벡터.
- 제 11 항에 따른 재조합 벡터가 형질전환된 숙주 세포.
- 제 12 항에 따른 숙주 세포를 배양하는 단계; 상기 배양된 숙주 세포로부터 세포 투과성 펩티드 이량체의 발현을 유도하는 단계; 및 발현된 세포 투과성 펩티드를 회수하는 단계를 포함하는, 세포 투과성 펩티드 이량체의 제조 방법.
- 제 1 항 및 제 3 항 내지 제 7 항 중 어느 한 항에 따른 세포 투과성 펩티드 이량체 및 상기 이량체에 컨쥬게이션된 카고를 포함하는 카고 전달체.
- 제 14 항에 있어서 카고는 펩티드, 단백질, 핵산, 지질, 당지질, 탄수화물, 미네랄, 나노 입자, 바이러스, 조영 물질, 화학 물질 또는 이들의 조합인 카고 전달체.
- 제 15 항에 있어서 카고는 세포 투과성 펩티드 이량체에 공유결합되는 것인 카고 전달체.
- 제 16 항에 있어서, 카고는 세포 투과성 펩티드 이량체의 제1 펩티드 도메인의 아미노-말단 또는 제2 펩티드 도메인의 카복시-말단에 공유결합되는 것인 카고 전달체.
