Abstract
Description
도 2a 내지 도 2c 는 NIS 프로모터 활성을 효과적으로 나타내는 TKI 농도 및 처리 시간을 나타낸다. 구체적으로 도 2a는 TKI 농도 및 처리 시간에 따른 Fluc 활성의 패턴을 나타내고, 도 2b는 TKI 농도 및 처리 시간에 따른 Rluc 활성의 패턴을 나타내며 도 2c는 TKI 농도 및 처리 시간에 따른 Rluc 활성으로 정규화된 Fluc 활성을 나타낸다.
도 3은 TKI의 투여에 의한 갑상선 특이적 유전자(NIS, TPO, Tg 및 TSHR) 및 갑상선 전사인자(TTF-1 및 Pax-8)의 상대적인 mRNA 발현양을 나타낸다.
도 4a 내지 4c는 8505C 세포에서 NIS 단백질의 발현양 및 발현 위치를 나타낸다. 구체적으로, 도 4a는 총 단백질 중에서 세포 내 NIS 단백질에 대한 웨스턴 블롯 결과이고, 도 4b 및 도 4c는 각각 세포막 NIS 단백질 및 세포질 NIS 단백질에 대한 웨스턴 블롯 결과를 나타낸다.
도 5a 및 도 5b는 유방암 세포에서 NIS 단백질의 발현양을 나타낸다. 도 5a 및 도 5b는 각각 MDA-MB-231 세포 및 MCF-7 세포에서 NIS 단백질에 대한 웨스턴 블롯 결과를 나타낸다.
도 6은 면역형광 이미징을 통해 세포 내 NIS 발현을 관찰한 결과를 나타낸다.
도 7a 및 도 7b는 TKI 투여시 갑상선 요오드 대사 단백질의 변화를 관찰한 결과를 나타낸다. 구체적으로 도 7a는 TKI 투여시 갑상선 특이적 단백질(Tg, TPO, NIS 및 TSHR)에 대한 웨스턴 블롯 결과를 나타내고, 도 7b는 TKI 투여시 갑상선 전사인자(Pax-8 및 TTF-1)에 대한 웨스턴 블롯 결과를 나타낸다.
도 8은 MAPK 및 PI3K-AKT 신호전달 경로와 관련된 유전자의 웨스턴 블롯 결과를 나타낸다.
도 9a 내지 도 9c는 8505C 세포에서 TKI 투여에 의한 방사성 요오드의 흡수를 평가한 결과를 나타낸다. 구체적으로, 도 9a는 TKI 투여에 의해 농도 의존적으로 방사성 요오드의 흡수능력이 현저하게 증가된 결과를 나타내며, 도 9b 및 도 9c 는 각각 12.5μM 및 25μM 의 TKI와 KClO4의 투여에 의한 방사성 요오드의 흡수능력을 나타낸다.
도 10a 및 도 10b는 131I 및 TKI 단독 또는 조합으로 투여시 8505C 세포의 생존율을 나타낸다, 구체적으로 도 10a는 131I 및 12.5μM의 TKI 단독 또는 조합으로 투여시 8505C 세포의 생존율을 나타내고, 도 10b는 131I 및 25μM의 TKI 단독 또는 조합으로 투여시 8505C 세포의 생존율을 나타낸다.
도 11a 및 도 11b는 131I 및 TKI 단독 또는 조합으로 투여시 DNA 손상정도를 나타낸다. 구체적으로 도 11a는 131I 및 TKI 단독 또는 조합으로 투여시 γH2A.X foci 형성 분석을 수행한 결과를 나타내고, 도 11b는 131I 및 TKI 단독 또는 조합으로 투여시 γH2A.X에 대한 웨스턴 블롯 결과를 나타낸다.
도 12a 내지 도 12d는 생체 내 실험에 의해 TKI 투여시 NIS 프로모터 활성이 증가한 결과를 나타낸다. 구체적으로, 도 12a는 종양 이종 이식 마우스 모델에서 비히클과 TKI 투여 그룹에서의 Fluc 활성을 나타낸다. 도 12b는 종양 이종 이식 마우스 모델에서 비히클과 TKI 투여 그룹에서 Fluc 활성을 정량분석한 결과를 나타낸다. 도 12c는 종양 이종 이식 마우스 모델에서 비히클과 TKI 투여 그룹에서의 Rluc 활성을 나타낸다. 도 12d는 종양 이종 이식 마우스 모델에서 비히클과 TKI 투여 그룹에서 Rluc 활성으로 정규화한 NIS 프로모터 활성을 정량분석한 결과를 나타낸다.
도 13a 및 도 13b는 종양 조직 내 NIS 발현양이 증가한 결과를 나타낸다. 구체적으로, 도 13a는 종양 이종 이식 마우스의 종양 조직에서 NIS 단백질 대한 웨스턴 블롯 결과를 나타내고, 도 13b는 종양 이종 이식 마우스의 종양 조직에서 NIS의 면역조직화학 염색을 수행한 결과를 나타낸다.
도 14a 내지 도 14c, 도 15a, 도 15b 및 도 16은 생체 내 실험에 의해 TKI 투여시 131I에 의해 매개된 암 치료 효과를 나타낸다.
도 14a는 131I 및 TKI를 단독 또는 조합으로 투여한 종양 이종 이식 마우스 모델에서 Rluc 활성을 나타내고, 도 14b는 131I 및 TKI를 단독 또는 조합으로 투여한 종양 이종 이식 마우스의 종양 부피를 나타내며. 도 14c는 생체 내 실험 동안 종양 이종 이식 마우스의 체중변화를 나타낸다.
도 15a는 131I 및 TKI를 단독 또는 조합으로 투여한 종양 이종 이식 마우스로부터 분리된 종양 조직을 나타내고, 도 15b는 131I 및 TKI를 단독 또는 조합으로 투여한 종양 이종 이식 마우스로부터 분리된 종양 조직에 대한 평균 무게를 나타낸다.
도 16은 종양 이종 이식 마우스로부터 분리된 종양 조직에서 γH2A.X의 면역조직화학 염색을 수행한 결과를 나타낸다.
