Abstract
이를 위해, 본 발명은 (a) 필렛(filleting) 작업 후 발생되는 참치 부산물 중 참치 껍질 부분을 별도로 준비하는 단계; (b) 준비된 참치 껍질을 원료로 하여 세척하여 참치 껍질에 흡착 및 잔여한 이물질을 제거하고 분쇄하는 단계; (c) 세척된 참치 껍질에 잔여한 수분을 제거하는 건조단계; (d) 참치 껍질에 효소를 처리하여 참치 껍질에 함유되어 있는 DHA(Docosa Hexaenoic Acid)를 분리하는 단계를 포함하고, (d) 분리단계는 발효조에 상기 참치 껍질과 용리액을 투입하고, DHA를 선택적으로 가수분해하는 리파아제(lipase)를 처리하는 것을 특징으로 하는 참치 껍질로부터 DHA를 분리 및 정제하는 방법을 제공할 수 있다.

Claims (3)
- (a) 필렛(filleting) 작업 후 발생되는 참치 부산물 중 참치 껍질 부분만을 별도로 준비하는 단계;
(b) 준비된 참치 껍질을 원료로 하여 산 용매 하에서 5~15℃ 온도 조건에서 1~4일간 세척 및 분쇄처리하거나 물로 세척 및 분쇄처리하여 참치 껍질에 흡착 및 잔여한 이물질을 제거하고 분쇄하는 단계;
(c) 세척된 참치 껍질에 잔여한 수분을 제거하는 건조단계;
(d) 참치 껍질에 효소를 처리하여 참치 껍질에 함유되어 있는 DHA(Docosa Hexaenoic Acid)를 분리하는 단계;
(e) (d) 분리단계에 의해 수득된 결과물에 단백질 분해효소 및 물을 처리한 후 원심분리기를 이용하여 800 내지 1000rpm으로 원심분리하여 단백질가수분해물이 함유된 수층과 DHA가 포함되어 있는 유층으로 재차 분리하고 그 중 상층액인 유층만을 수득하는 정제단계;
(f) 40℃~60℃, 120~200bar의 온도 및 압력 조건 하에서 헵탄알(heptanal)과 옥탄알(octanal)과 같은 지방취와 같은 이취를 발생시키는 물질을 분리시키는 이취발생원 제거단계
를 포함하고,
(d) 분리단계는
발효조에 상기 참치 껍질과 용리액을 투입하고, DHA를 선택적으로 가수분해하는 리파아제(lipase)를 처리하는 것
을 포함하며,
(d) 분리단계에서 참치 껍질과 함께 투입되는 용리액은 정제수 또는 메탄올(methanol), 에탄올(ethanol), 프로판올(propanol), 이소프로판올(isopropanol),부탄올(butanol)을 포함하는 알콜을 포함하는 용매 중 선택된 하나 또는 그 이상을혼합하여 사용하는 것
을 포함하고,
(e) 정제단계는
염산, 황산을 포함하는 산 원액 또는 희석액 중 선택된 어느 하나를 투입하여 참치껍질 용액의 pH를 1 내지 5 범위로 조절하여 단백질과 지질의 점착성을 저하시켜 고도불포화지방의 순도가 높은 지질을 분리할 수 있도록 하는 것을 포함하는 참치 껍질로부터 DHA를 분리 및 정제하는 방법.
