Abstract
Description
도 2는 본 발명의 발현 벡터를 이용하여 형질전환 된 대장균에서 재조합 단백질의 발현 결과 및 정제 후 SDS-PAGE 결과를 나타낸 것으로, (A)는 MHR 표출 단백질 나노입자 발현 결과 및 정제 결과, (B)는 도 1(A) 및 도 1(B)에 명시된 두 개의 발현 벡터를 동시에 형질전환 시킨 대장균에서 재조합 단백질 생산 후 정제한 결과이다. (C)는 도 1(E) 및 도 1(F)에 명시된 두 개의 발현 벡터를 동시에 형질전환 시킨 대장균에서 재조합 단백질 생산 후 정제한 결과이다.
도 3는 ELISA 를 이용한 항체가 분석에 사용되는 표준곡선을 나타낸 것이다.
도 4는 SpaB의 면역증강효과 검증을 위한 1차 동물 실험 결과를 나타낸 것으로, (A)는 동물 실험에 적용한 백신군, (B)는 백신군 투여 후 쥐 혈청 내 B형 간염 바이러스 항체 농도 측정을 통한 항체 양전률 비교 결과 [Seroconverted rate: 각 백신군 별 접종 후 20마리 쥐에서 추출한 혈청 내 B형 간염 바이러스 항체 농도가 4mlU/mL 이상인 쥐 마리수의 백분율 (%), GMT: 항체 농도의 기하평균 (mlU/mL]와 혈청 내 B형 간염 바이러스 항체 중 IgG1 농도/IgG2a 값의 기하평균(IgG/IgG2a), IgG1/IgG2a의 비율이 1이상인 개체수의 백분율(IgG1/IgG2a>1)]이다.
도 5는 본 발명의 발현 벡터를 이용하여 형질전환 된 대장균에서 재조합 단백질의 발현 결과 및 정제 후 SDS-PAGE 결과로, (A)는 MHR 표출 단백질 나노입자의 아미노 말단에 SpaB 가 융합된 단백질 나노입자 발현 및 정제 결과, (B)는 MHR 표출 단백질 나노입자의 아미노 말단에 SpaB 가 두 번 중복되게 융합된 단백질 나노입자 발현 및 정제 결과이다.
도 6은 SpaB의 융합 위치에 따른 면역원성 비교를 위한 2차 동물 실험 결과로, (A)는 동물 실험에 적용한 백신군, (B)는 백신군 투여 후 쥐 혈청 내 B형 간염 바이러스 항체 농도 측정을 통한 항체 양전률 비교 결과 [Seroconverted rate: 각 백신 군 별 접종 후 20마리 쥐에서 추출한 혈청 내 B형 간염 바이러스 항체 농도가 4mlU/mL 이상인 쥐 마리 수의 백분율 (%), GMT: 항체 농도의 기하평균 (mlU/mL], (C)는 혈청 내 B형 간염 바이러스 항체 중 IgG1 농도/IgG2a 값의 기하평균(IgG/IgG2a)]이다.
Claims (15)
- B형 간염 바이러스 캡시드 단백질의 SEQ ID NO: 4에 기재된 1-78번째 아미노산 서열, 면역증강제의 아미노산 서열 및 B형 간염 바이러스 캡시드 단백질의 SEQ ID NO: 5에 기재된 81-149번째 아미노산 서열이 순차적으로 연결된 키메릭 단백질로부터 제조된 단백질 나노입자로 이루어진 복수의 제1소단위체(subunit); 및
B형 간염 바이러스 캡시드 단백질의 SEQ ID NO: 4에 기재된 1-78번째 아미노산 서열, 항원의 아미노산 서열 및 B형 간염 바이러스 캡시드 단백질의 SEQ ID NO: 5에 기재된 81-149번째 아미노산 서열이 순차적으로 연결된 키메릭 단백질로부터 제조된 단백질 나노입자로 이루어진 복수의 제2소단위체를 포함하고,
상기 제1소단위체 및 제2소단위체는 1 : 1 내지 4의 중량비로 혼합되어 있고, 상기 복수의 제1소단위체 및 복수의 제2소단위체의 자가조립에 의해 단일 단백질 나노입자로 제조되는, 복합 단백질 나노입자.
