Abstract
Description
도 2는 본 발명의 시스템을 이용하여 이중-배럴형 피펫을 사용한 세포 침투의 도식도이다.
도 3은 시간 경과에 따른 나노피펫 팁으로부터 방출된 액체의 부피를 나타낸 그래프이다. 그래프에 나타낸 바와 같이, 방출 속도는 시간에 따라 선형이다.
도 4a는 패턴화된 스팟 어레이를 나타낸 상면(top view) 사진이다.
도 4b는 술포로다민을 사용한 패턴화된 스팟 어레이의 형광 영상의 모의-3-D 그래프이다.
도 5는 본 발명의 나노주입 기기를 나타낸 일련의 사진이다. 도 5a는 카르복시플루오레세인의 HeLa 세포로의 주입을 나타낸 사진이다. 세포 및 나노피펫의 명시 야상(brightfield image)이다. 화살표는 나노피펫의 팁을 나타낸다. 도 5b는 또한 도 5a에서와 같은 세포 주입의 사진이다. 도 5b는 주입 후의 HeLa 세포 및 나노피펫의 형광 영상을 나타낸다.
도 6은 세포 체(sifter) 제작을 나타낸 일련의 도면이다. 도 6a는 규소 웨이퍼 상에 침착된 질화규소 층을 나타낸 도면이다. 도 6b는 질화물 층을 통과하는 심도 반응성 이온 에칭을 나타낸 도면이다. 도 6c는 후방 KOH 에칭을 나타낸 도면이다. 도 6d는 유리 웨이퍼에 결합된 규소 웨이퍼를 나타낸 도면이다. 도 6e는 최종 기기에서 가소성 고리가 표면에 부착된 것을 나타낸 도면이다.
도 7은 배양 챔버 및 진공을 적용하기 위한 기구를 나타낸 최종 기기의 사진이다. 페니는 기기의 상대적 크기가 페니보다 작다는 것을 설명하기 위해 도시된다. 상기 기기를 이용하여 세포를 배양 고리에서 배양하고 음의 압력을 인가하여 관통 구멍 상에 고정시킬 수 있다.
도 8은 세포 체 상에 고정된 10 mm 폴리스티렌 비드의 형광 영상을 나타낸 사진이다.
도 9a는 주사 이온 전도 현미경 (SICM)을 이용한 지형 맵핑을 나타낸 카툰(cartoon)이다. 도 9b는 세포 상 위치에 대한 나노피펫 팁의 높이 변화를 나타낸 지형 맵핑의 그래프이다.
도 10은 나노피펫 센서에 의한 HPV18E6 항원의 선택적 검출을 나타낸 선 추적이다. 인가된 전압: -400 mV. 항원은 0 시간에 첨가하였다.
도 11은 HeLa 세포 용해물에 대한 나노피펫 센서의 반응을 나타낸 막대 그래프이다.

Claims (32)
- (a) (i) 제1 배럴 중의 액체와 접촉하도록 배열된 제1 전극을 함유하는 제1 배럴;
(ii) 제2 배럴 중의 액체와 접촉하도록 배열된 제2 참고 전극을 함유하는, 제1 배럴에 인접한 제2 배럴; 및
(iii) 상기 제1 전극과 상기 제2 참고 전극 사이의 바이어스 전압을 제공하기 위해 상기 제1 전극에 연결된 증폭기
를 갖는 다중-배럴형 나노피펫;
(b) 다중-배럴형 나노피펫의 기계적 움직임을 서브마이크론 x 및 y 단계로 실행하고 상기 다중-배럴형 나노피펫의 움직임을 기판을 향하거나 기판으로부터 멀어지는 z 방향으로 실행하기 위해 상기 다중-배럴형 나노피펫에 부착되고, 사용자 정의된 제어에 따라 상기 기계적 움직임을 제어하기 위한 전자 제어를 추가로 갖는 xyz 제어기; 및
(c) 상기 제1 전극과 상기 나노피펫 중의 액체가 방출될 세포를 포함하는 목적하는 위치에서 제1 배럴로부터 액체가 방출되도록 상기 제1 전극에 방출 전압을 인가하기 위해 배열된 고전압원에 연결된 회로로서, 상기 회로는 xyz 제어기가 목적하는 위치로부터 멀어지는 나노피펫의 기계적 움직임을 실행할 때 상기 방출 전압을 제거하기 위한 제어기를 추가로 포함하며, 상기 회로는 상기 바이어스 전압과 상기 방출 전압 사이의 변환을 위한 계전기를 추가로 포함하여, 개별 세포가 방출된 액체와 접촉할 수 있도록 하기 위한 회로
를 포함하는, 기판 상에서 개별 세포를 조작하기 위한 장치. - 제1항에 있어서, 상기 증폭기가 상기 바이어스 전압의 제공 및 추가적으로 제1 배럴을 통한 이온 전류의 전류 측정을 위해 저 노이즈 증폭기를 포함하는 것인 장치.
- 제1항에 있어서, xyz 제어기의 서브마이크론 제어를 위해 압전 작동기를 추가로 포함하는 장치.
- 제1항에 있어서, 상기 목적하는 위치를 결정하도록 사용자 입력을 위한 필드-프로그래밍가능한 게이트 어레이 (FPGA)를 추가로 포함하는 장치.
- 제4항에 있어서, 상기 FPGA가 상기 기판 상에 스팟을 패턴으로 침착시키기 위해 프로그래밍된 것인 장치.
- 제4항에 있어서, 상기 FPGA가 상기 기판 상의 기정 위치에 있는 세포를 주입하도록 프로그래밍된 것인 장치.
- 제6항에 있어서, 상기 FPGA가 상기 세포 내의 소기관을 주입하도록 프로그래밍된 것인 장치.
- 제1항 내지 제7항 중 어느 한 항에 있어서, 개별 위치들에서 다수개의 개별 세포들을 각 위치마다 한 세포씩 함유하도록 적용된 기판에 작동가능하게 연결된 장치.
- 제8항에 있어서, 상기 기판이 그 안에 단일 공동 내로 개별 세포를 수용하기 위한 크기를 갖도록 정의된 공동을 갖는 것인 장치.
- 제9항에 있어서, 상기 기판이 세포를 공동에 보유시키도록 음의 압력을 인가하기 위한 관통 구멍을 포함하는 것인 장치.
- 제10항에 있어서, 상기 공동이 세포를 공동으로 유인하기 위한 전극을 포함하는 것인 장치.
