Abstract
Description
도 2a는 알기네이트 히드로겔 스탬핑(stamping)을 통한 세포 패터닝 방법을 나타내는 모식도이고, 도 2b는 스탬핑된 세포의 이미지이다.
도 3은 알기네이트 히드로겔의 높이 및 스탬핑 시간에 따른 세포의 이미지이다(음성 대조군: PDMS 몰드로 스탬핑한 세포).
도 4는 스탬핑된 세포를 각각 세포골격 및 항-MF20 항체로 면역염색하여 수득한 이미지이다.
도 5는 패턴이 형성된 알기네이트 히드로겔을 사용하여 세포를 스탬핑한 후 알기네이트 히드로겔을 제거하는 경우, 0일, 3일, 또는 7일 동안 배양된 세포를 면역염색하여 수득한 이미지이다.
Claims (18)
- 트랜스웰 챔버의 하단부에 부착된 알기네이트 히드로겔을 포함하고,
상기 알기네이트 히드로겔의 하부 표면은 세포의 상단부와 접촉하는 세포 패터닝용 물질. - 청구항 1에 있어서, 상기 알기네이트 히드로겔은 패턴이 형성된 것인 세포 패터닝용 물질.
- 청구항 2에 있어서, 상기 알기네이트 히드로겔의 홈의 너비는 0.1 ㎛ 내지 50 ㎛이고, 마루는 0.1 ㎛ 내지 50 ㎛이고, 및 홈과 마루 사이의 높이는 0.1 ㎛ 내지 50 ㎛인 것인 세포 패터닝용 물질.
- 청구항 3에 있어서, 홈과 마루 사이의 높이는 0.1 ㎛ 내지 20 ㎛인 것인 세포 패터닝용 물질.
- 청구항 1에 있어서, 상기 알기네이트는 N-말단으로부터 아르기닌-글리신-아스파르트산의 아미노산 서열(Arg-Gly-Asp: RGD)을 갖는 폴리펩티드를 포함하거나 포함하지 않는 것인 세포 패터닝용 물질.
- 패턴이 형성된 고분자 몰드에 알기네이트 용액을 위치시키는 단계;
상기 고분자 몰드에 위치한 알기네이트 용액에 트랜스웰 챔버를 접촉시키는 단계;
상기 트랜스웰 챔버에 칼슘 용액을 가하여 알기네이트 용액을 알기네이트 히드로겔로 겔화시키는 단계; 및

- 청구항 6에 있어서, 상기 고분자 몰드는 알킬실록산, 폴리스티렌, 폴리아크릴아미드, 폴리락트산, 폴리히드록시산, 폴리아미드, 폴리아미노산, 폴리아세탈, 폴리시아노아크릴레이트, 폴리우레탄, 폴리피롤, 폴리에스테르, 폴리메타크릴레이트, 폴리에틸렌, 폴리카르보네이트. 폴리에폭시에탄, 또는 이들의 조합인 것인 방법.
- 청구항 8에 있어서, 상기 알킬실록산은 폴리디메틸실록산인 것인 방법.
- 청구항 6에 있어서, 상기 알기네이트는 RGD 아미노산 서열을 포함하거나 포함하지 않는 알기네이트인 것인 방법.
- 청구항 6에 있어서, 상기 칼슘 용액은 CaCl2, CaSO4, CaCO3, 또는 이들의 조합 용액인 것인 방법.
- 세포와 청구항 1의 세포 패터닝용 물질을 접촉시켜 알기네이트 히드로겔의 하부 표면과 세포의 상단부를 접촉시키는 단계;
상기 세포 패터닝용 물질의 트랜스웰 챔버에 알기네이트 분해 효소 또는 칼슘 킬레이트 제제를 가하여 알기네이트 히드로겔을 제거하는 단계; 및
상기 세포와 트랜스웰 챔버를 분리하는 단계를 포함하는, 세포 패터닝 방법. - 청구항 12에 있어서, 상기 세포는 근육 세포, 신경 세포, 줄기 세포, 결합 조직 세포, 혈관 세포, 또는 상피 세포인 것인 방법.
- 청구항 12에 있어서, 세포와 청구항 1의 세포 패터닝용 물질을 접촉시키는 시간은 1 초 내지 24 시간인 것인 방법.
- 청구항 12에 있어서, 상기 트랜스웰 챔버에 알기네이트 분해 효소 또는 칼슘 킬레이트 제제를 가한 후 인큐베이션하는 단계를 더 포함하는 방법.
- 청구항 15에 있어서, 상기 칼슘 킬레이트 제제는 시트레이트(Citrate), 에틸렌디아민 테트라아세트산(ethylenediamine tetraacetic acid: EDTA), 에틸렌 글리콜 테트라아세트산(ethyleneglycol tetraacetic acid: EGTA), BAPTA-AM, 또는 이들의 조합인 것인 방법.
- 청구항 15에 있어서, 상기 인큐베이션은 1 분 내지 2 일 동안 수행되는 것인 방법.
- 청구항 12의 방법으로 패턴된 세포를 포함하는 바이오센서.
