Abstract
본 발명의 EM17의 만난분해효소는 대장균에서 발현되어 높은 유기용매 농도, 높은 금속이온 및 폭넓은 산, 염기 농도에서도 분해 능력을 발휘하므로 헤미셀룰로오스 바이오 매스 재활용 공정에서 불필요한 정제단계를 줄일 수 있다. 또한, 다양한 가수분해 기능을 가지므로 식품, 동물사료, 커피추출, 제지. 펄프 산업, 올리고당 합성 등에 이용될 수 있다.
Description
도 2는 신규한 만난 분해효소 EM17단백질 아미노산 서열의 비교 계통분석도에 관한 것이다. 각 branch에 있는 숫자는 Bootstrap 분석치(1,000 resampling)이다(Bar; 0.1 substitutions per amino acid position).
도 3은 신규한 만난 분해효소 EM17의 대장균 BL21(DE3)에서의 이종 대량발현에 관한 것이다(Lane 1, Induction 하기 전의 대장균 조단백질; Lane 2, EM17을 과대 발현한 대장균 조단백질; Lane 3, Soluble fraction; Lane 4, 정제된 EM17 단백질; M은 단백질 size marker).
도 4는 정제된 만난 분해효소 EM17단백질을 이용한 생화학적 최적 활성온도(A)와 최적 활성 pH(C)를 나타내는 것이다. 또한 EM17 단백질의 주어진 온도에서의 시간별 잔류활성(B)과 주어진 pH에서의 4℃, 15시간 후의 잔류활성(D)에 관한 것이다.
도 5는 정제된 만난 분해효소 EM17의 유기용매(A), 계면활성제(B), 금속이온(C)에 의한 기능억제(inhibition)에 관한 것이다.
도 6은 정제된 만난 분해효소 EM17 단백질을 이용한 만난분해 기능을 기질별로 확인한 결과를 나타낸다.
Claims (7)
- 서열번호 1의 핵산서열로 표시되는 만난분해효소 EM17 유전자.
- 서열번호 2의 아미노산서열로 표시되는 만난분해효소 EM17 단백질.
- 제 1항의 유전자를 포함하는 재조합 백터.
- 제 3항의 재조합 백터로 형질전환된 형질전환체.
- 제 4항에 있어서, 상기 형질전환체는 형질전환 되는 숙주세포를 대장균으로 하는 것을 특징으로 하는 형질전환체.
- 제5항에 있어서,
상기 형질전환체는 KACC 95124P균주인 것을 특징으로 하는 형질전환체. - (a) 제4항 내지 제6항 중 어느 한 항에 따른 형질전환체를 배양하는 단계;
(b) 상기 배양된 형질전환체로부터 만난분해효소를 생산하는 단계; 및
(c) 상기 생산된 만난분해효소를 회수하는 단계를 포함하는 만난분해 효소의 제조방법.
