Claims (16)
- (A) 화학식 1의 연결기로 수식된 수지를 고상으로서 사용하여 고상 합성법으로 펩티드를 형성시키고,(B) 묽은 염산, 묽은 황산, 포름산 및 초산으로 이루어진 그룹으로부터 1종 이상 선택된 산으로 고상과 펩티드의 결합을 절단하여, C 말단에 카르복실기를 갖는 펩티드를 제조하고,(C) 용매 중, 1-하이드록시벤조트리아졸/디이소프로필카보디이미드 또는 벤조트리아졸-1-일옥시-트리스피롤리디노포스포늄헥사플루오로포스페이트/디이소프로필에틸아민의 존재 하에서 티올 화합물을 -20 내지 0℃에서 반응시킴을 포함함을 특징으로 하는, 펩티드의 티오에스테르 화합물의 제조 방법.[화학식 1]상기 식에서, R1은 C1-4 알킬기를 나타내고, R2는 수소 원자 또는 C1-4 알콕시기를 나타내며, n는 1 내지 4의 정수를 나타낸다.

- C 말단에 카르복실기를 갖는 펩티드를, -20℃ 내지 0℃에서 용매 중, 1-하이드록시벤조트리아졸/디이소프로필카보디이미드 또는 벤조트리아졸-1-일옥시-트리스피롤리디노포스포늄헥사플루오로포스페이트/디이소프로필에틸아민의 존재 하에서 티올 화합물과 반응시키는 것을 포함함을 특징으로 하는, 펩티드의 티오에스테르 화합물의 제조 방법.
- 제1항 또는 제3항에 있어서, 상기 펩티드의 N 말단 아미노산이 시스테인인, 제조 방법.
- 제4항에 있어서, 상기 시스테인의 티올기가 지용성 보호기로 보호된 펩티드인, 제조 방법.
- 제1항에 있어서, 상기 화학식 1의 연결기로 수식된 수지가, 아미노 PEGA 수지의 아미노기에 상기 화학식 1의 연결기가 결합한 수지인, 제조 방법.
- 제1항에 있어서, 상기 (B)의 공정을 알코올의 존재 하에서 수행함을 특징으로 하는, 제조 방법.
- 제7항에 있어서, 상기 알코올이 할로게노알코올인, 제조 방법.
- 제1항 또는 제3항에 있어서, 상기 용매가, N,N-디메틸포름아미드, N-메틸피롤리돈으로 이루어진 그룹에서 1종 이상 선택되는, 제조 방법.
