Claims (17)
- CHO 세포에서 활성이며 재조합 산물 단백질의 발현을 이끄는 프로모터를 포함하고, 상기 프로모터의 활성을 강화시키는 쥐 (murine) IgG 2A 유전자 좌위 (locus) DNA 유래의 부분을 추가로 포함하는 발현 벡터로 트랜스펙션된 CHO 세포.
- 제 1 항에 있어서, 상기 벡터는 선별가능 표지자를 인코딩하는 전사 단위체를 추가로 포함하는 것을 특징으로 하는 CHO 세포.
- 제 1 항 또는 제 2 항에 있어서, 상기 CHO 세포는 안정적으로 트랜스펙션되는 것을 특징으로 하는 CHO 세포.
- 하기 단계를 포함하는, 재조합 단백질을 발현시키는 방법:a. CHO 세포에서 활성이며 재조합 산물 단백질의 발현을 이끄는 프로모터를 포함하고 상기 프로모터의 활성을 강화시키는 쥐 IgG 2A 유전자 좌위 DNA 또는 이의 DNA 서열 변이체 또는 DNA 절편을 추가로 포함하는 발현 벡터로 트랜스펙션된 CHO 세포를 배양하는 단계, 및b. 상기 산물 단백질을 수확하는 단계.

- 제 10 항에 있어서, 상기 mCMV 프로모터 또는 이의 기능성 절편이 천연의 Xho I 제한효소 절단부위까지 확장되는 벡터.
- 제 10 항에 있어서, 상기 전사 시작 부위가 재조합 유전자 산물의 삽입을 위한 적절한 제한효소 절단부위를 포함하도록 유전자 조작되는 벡터.
- 제 9 항 또는 제 10 항에 있어서, 상기 첫번째 전사 단위체가 하나 이상의 인트론 (intron) 서열을 내포하는 벡터.
- 제 13 항에 있어서, 상기 인트론이 상기 mCMV 프로모터의 첫번째의 천연 인트론이 아닌 벡터.
- CHO 세포에서 트랜스펙션율을 강화시키기 위하여 제 9 항의 벡터를 사용하는 방법.
- 제 1 항에 있어서, 상기 벡터는 글루타민 합성효소 (GS) 표지자를 인코딩하는 전사 단위체를 추가로 포함하는 것을 특징으로 하는 CHO 세포.
- 제 4 항에 있어서, 상기 프로모터는 hCMV 프로모터인 것을 특징으로 하는 방법.
