Abstract
Description
상기 뇌하수체 호르몬 추출단계(102)에서 얻어진 뇌하수체 호르몬을 단백질정제장치(FPLC)에 연결된 겔 크로마토그래피 컬럼(세파크릴-100, φ2.6 x70cm)에 용출용 0.05몰 암모니움 아세테이트(pH 9.0) 버퍼로 정제하여 균질화 한 후, 유속 3㎖/200s/ tube로 용출하여 분취물(S3)을 얻는 겔 크로마토그래피법에 의한 정제단계(103)(도 2)를 수행한다.
즉, 상기 고속액체크로마토그래피 컬럼에 의한 고순도 정제단계(105)를 통해 수득한 정제물을 역상 고속액체크로마토그래피의 피크들을 전기영동법으로 확인하는 전기영동법에 의한 정제물질의 검증을 수행한 다음, 피크 9번(S3DE5W9)과 12번(S3DE5W12)을 N-말단 아미노산 서열분석기를 통해 N-말단부터 9개 정도의 아미노산 서열을 밝히는 아미노산 서열 분석기에 의한 정제물질의 검증을 표 1에 나타낸 바와 같이 수행함으로써 아미노산 서열을 밝혔으며, 이것을 기 보고된 조피볼락 각 서브 유니트인 당단백질성 알파(RF GPα) 및 황체형성호르몬 베타(RF LHβ)에 대한 뉴크레오타이드 결과에서 연역된 아미노산 결과와 비교하여 확인, 검증하였다.
즉, 토끼의 경동맥을 절단하여 전 채혈하는 채혈단계를 수행한 다음, 채혈된 혈액을 4℃에서 16시간 동안 비등화 시킨 후, 중력가속도 10,000g에서 30분간 원심 분리하여 항혈청을 분리하는 항혈청분리단계를 수행하였으며, 분리된 각각의 항혈청은 1㎖씩 분주 및 동결 건조시킨 후, 사용할 때까지 -80℃에 보관하였다.
또한 면역조직화학법으로 조피볼락 뇌하수체내 황체형성호르몬(RF LH) 분비세포의 분포 구성을 통해 항체 특이성을 재확인하였다(도 6).

- 조피볼락(rockfish:RF)의 뇌하수체를 채취 후 정제하여 조피볼락 황체형성호르몬(RF LH)을 얻고,상기 조피볼락 황체형성호르몬(RF LH)의 서브 유니트인 당단백질성 알파(RF GPα) 및 황체형성호르몬 베타(RF LHβ)를 분리 후 각각에 대한 다클론 항체을 얻고,상기 황체형성호르몬 베타(RF LHβ)에 대한 다클론 항체를 이용한 효소면역측정계로서 성숙도를 판정하도록 하는 것을 특징으로 하는 조피볼락 황체형성호르몬의 다클론 항체를 이용한 성숙도 판정방법.
- 제3항에 있어서 상기 조피볼락 황체형성호르몬(RF LH)은,성숙중인 조피볼락(RF)의 뇌하수체를 채취하는 뇌하수체 채취단계와,상기 뇌하수체 채취단계에서 채취된 뇌하수체를 0.2몰 암모니움 아세테이트(pH 6.1) 호모게나이져 후, 중력가속도 15,000g에서 30분간 원심분리하여 침전물인 뇌하수체 호르몬을 얻는 뇌하수체 호르몬 추출단계와,상기 뇌하수체 호르몬 추출단계에서 얻어진 뇌하수체 호르몬을 크로마토그래피 컬럼에 0.05몰 암모니움 아세테이트(pH 9.0) 버퍼로 정제하여 분취물(S3)을 얻는 겔 크로마토그래피법에 의한 정제단계와,상기 겔 크로마토그래피법에 의한 정제단계에서 얻어진 분취물(S3)을 음이온 크로마토그래피 컬럼에 0.05, 0.1, 0.2, 0.4, 1.0 몰의 암모니움 아세테이트(pH 9.0) 버퍼로 정제하여 분취물(S3DE5)을 얻는 이온 크로마토그래피 컬럼에 의한 정제단계와,상기 이온 크로마토그래피 컬럼에 의한 정제단계에서 얻어진 분취물(S3DE5)을 역상 고속액체크로마토그래피 컬럼에 흡착시켜 아세트나이테르의 농도구배로 정제하는 고속액체크로마토그래피 컬럼에 의한 고순도 정제단계로 추출되는 것을 특징으로 하는 조피볼락의 다클론 항체를 이용한 성숙도 판정방법.
- 제3항 또는 제4항에 있어서 상기 조피볼락 황체형성호르몬(RF LH) 서브 유니트의 황체형성호르몬 베타(RF LHβ)에 대한 다클론 항체는,상기 조피볼락 황체형성호르몬(RF LH) 서브 유니트의 당단백질성 알파(RF GPα) 및 황체형성호르몬 베타(RF LHβ) 50㎍를 동량의 프룬드 컴퍼리트 어쥬번트와 섞어 토끼의 임파선에 주사하는 제1정제품 면역단계와,상기 제1정제품 면역단계 후, 10일 간격으로 RF GPα 및 RF LHβ 50㎍와 동량의 프룬드 인컴퍼리트 어쥬번트와 섞어 피하에 4회 주사하는 제2정제품 면역단계와,상기 제2정제품 면역단계 후, 토끼의 혈액을 채혈하는 채혈단계와,상기 채혈된 혈액을 4℃에서 16시간 동안 비등화 시킨 후, 중력가속도 10,000g에서 30분간 원심 분리하여 항혈청을 분리하는 항혈청분리단계로 제조되는 것을 특징으로 하는 조피볼락의 다클론 항체를 이용한 성숙도 판정방법.
