Abstract
Description
본 발명의 한가지 바람직한 구체예에서는 HPMC 캡슐을 충전 후, 공지 기술, 예컨대 밴드 접착 기술 또는 또는 캡슐 본체(body)와 캡(cap) 사이의 틈새(gap)에 밀봉 액체를 도포하고/도포하거나 상기 틈새에 열을 가함으로써, 캡슐 본체와 캡 사이의 겹쳐지는 부분을 밀봉시킨다. 바람직한 밀봉 기술은, 용매를 함유할 수도 있는 밀봉 액체를, 밀봉하고자 하는 캡슐의 틈새의 외부 가장자리에 개별적으로 그리고 균일하게 도포하여 캡슐 외주에 액상 고리를 형성한 다음, 과량의 밀봉 액체를 캡슐 외부로부터 제거한 후 외부로부터 열에너지를 가하여 캡슐을 건조시키는 방법이다. 이러한 피복 전의 밀봉은 위장으로의 캡슐 내용물의 원치 않는 조기 방출을 야기시킬 수 있는, 예컨대 스트레싱 조건 하에서의 보관시 균열이 발생하거나 또는 틈새에서의 피막의 불균일성과 관련한 문제점들을 방지해준다.
본 발명의 HPMC 캡슐에서 피막은 비어 있는 HPMC 캡슐의 본체와 캡에 별도로 적용할 수 있다. 따라서, HPMC 캡슐의 본체에는 비수용성 폴리머를 피복하고, 캡에는 전술한 장용성 폴리머나 또는 결장용성 폴리머로 피복시킬 수 있다. 비수용성 폴리머로는 에틸 셀룰로스를 들 수 있다. 또한, 본 발명의 HPMC 캡슐에서 2개의 균등한 HPMC 캡슐 절반들은 캐플렛으로 충전될 수 있으며, 여기서 비어있는 HPMC 캡슐의 절반씩에 별도로 피복을 실시할 수 있다. 예컨대, 절반은 전술한 장용성 폴리머로, 나머지 절반은 전술한 결장용성 폴리머로 피복할 수 있다. 또는 절반은 비수용성 폴리머로 피복하고 나머지 절반은 전술한 장용성 폴리머 또는 결장용성 폴리머로 피복할 수 있다. 본 발명의 일례에서 본 발명의 HPMC 캡슐은 pH < 3에서는 불용성이나 pH > 5.5에서는 용해성인 필름으로 피복되어 있다. 또 다른 구체예로 본 발명의 HPMC 캡슐의 캡슐 껍데기의 HPMC 함량은 10 내지 90 중량% 범위이다.

Claims (31)
- 약물을 함유하는 피복된 HPMC 캡슐로 구성된 약물 전달용 조성물로서 여기서 상기 HPMC 캡슐은 약물이 위장에서 캡슐로부터 방출되지 않도록, 하나의 수성 피막을 갖는 것인 약물 전달용 조성물.
- 제1항에 있어서, 약물이 주로 소장에서 캡슐로부터 방출되도록 HPMC 캡슐에 피막이 제공되어 있는 약물 전달용 조성물.
- 제1항에 있어서, 약물이 주로 결장 및/또는 말단 회장에서 캡슐로부터 방출되도록 HPMC 캡슐에 피막이 제공되어 있는 약물 전달용 조성물.
- 제2항에 있어서, 피막은 셀룰로스 아세테이트 트리멜리테이트 (CAT), 히드록시프로필메틸 셀룰로스 프탈레이트 (HPMCP), 폴리비닐 아세테이트 프탈레이트 (PVAP), 셀룰로스 아세테이트 프탈레이트 (CAP) 및 쉘락 중에서 선택된, pH 5.5 이상에서 용해되는 폴리머를 함유하는 것인 약물 전달용 조성물.
- 제3항에 있어서, 피막은 셀룰로스 아세테이트 트리멜리시에이트 (CAT), 히드록시프로필메틸 셀룰로스 프탈레이트 (HPMCP), 폴리비닐 아세테이트 프탈레이트 (PVAP), 셀룰로스 아세테이트 프탈레이트 (CAP), 쉘락, 메타크릴산과 에틸 아크릴레이트와의 코폴리머, 및 메타크릴산과 에틸 아크릴레이트와의 코폴리머에 중합화 중 메타크릴레이트 모노머가 부가된 코폴리머 중에서 선택된, pH 7 이상에서 용해되는 폴리머를 함유하는 것인 약물 전달용 조성물.
- 제2항에 있어서, 피막은 셀룰로스 아세테이트 트리멜리시에이트 (CAT)를 함유하는 것인 약물 전달용 조성물.
- 제2항에 있어서, 피막은 히드록시프로필메틸 셀룰로스 프탈레이트 (HPMCP)를 함유하는 것인 약물 전달용 조성물.
- 제2항에 있어서, 피막은 폴리비닐 아세테이트 프탈레이트 (PVAP)를 함유하는 것인 약물 전달용 조성물.
- 제2항에 있어서, 피막은 쉘락을 함유하는 것인 약물 전달용 조성물.
- 제2항에 있어서, 피막은 메타크릴산과 메틸메타크릴레이트와의 코폴리머를 함유하는 것인 약물 전달용 조성물.
- 삭제
- 제3항에 있어서, 피막 조성물은 아조폴리머 또는 디설파이드 폴리머를 함유하는 것인 약물 전달용 조성물.
- 제3항에 있어서, 피막 조성물은 폴리사카라이드를 함유하는 약물 전달용 조성물.
- 제3항에 있어서, 피막 조성물은, 메타크릴산과 메틸메타크릴레이트와의 코폴리머에 중합 중 메틸 아크릴레이트 모노머가 부가된 코폴리머를 함유하는 것인 약물 전달용 조성물.
- 제3항에 있어서, 피막 조성물은 셀룰로스 에스테르를 함유하는 것인 약물 전달용 조성물.
- 제3항에 있어서, 피막 조성물은 폴리비닐 아세테이트 프탈레이트를 함유하는 것인 약물 전달용 조성물.
- 제2항에 있어서, 피막은 캡슐 표면 1cm2 당 5-15mg 범위의 양으로 적용된 것인 약물 전달용 조성물.
- 제3항에 있어서, 피막은 캡슐 표면 1cm2 당 5-20mg 범위의 양으로 적용된 것인 약물 전달용 조성물.
- 제2항에 있어서, 상기 약물은 단백질 분해 효소, 디클로페낙, 나프록센, 아스피린, 인도메타신, 오메프라졸, 강심 배당체, 나트륨, 칼륨, 및 마그네슘염의 전해질 제제, 칼슘 및 철 제제, 비사코딜 제제 및 발프로산 중에서 선택된 소장에서 효과적인 약물인 것인 약물 전달용 조성물.
- 제1항에 있어서, 상기 약물은 5-ASA; 스테로이드류; 하제; 옥트레오타이드 (octreotide); 시사프리드; 항콜린제; 칼슘 채널 차단제, 5HT3-길항제, 및 인슐린 중에서 선택된, 결장에서 국소적으로 작용하는 약물인 것인 약물 전달용 조성물.
- 제1항에 있어서, 상기 피막은 비어 있는 HPMC 캡슐 본체와 캡에 별도로 적용되는 것인 약물 전달용 조성물.
- 제21항에 있어서, 상기 HPMC 캡슐 본체는 비수용성 폴리머로 피복되고 캡은 제4항에 기재된 폴리머에 의해 장용성 피복되거나 또는 제5항에 기재된 폴리머에 의해 결장용성 피복된 것인 약물 전달용 조성물.
- 제22항에 있어서, 비수용성 폴리머는 에틸 셀룰로스인 약물 전달용 조성물.
- 제 1항에 있어서, 2개의 동등한 HPMC 캡슐 절반씩이 캐플렛으로 충전되어 있는 약물 전달용 조성물.
- 제24항에 있어서, 피막은 비어있는 HPMC 캡슐 절반씩에 별도로 적용된 약물 전달용 조성물.
- 제24항에 있어서, 절반은 제4항에 기재된 폴리머에 의해 장용성 피복되고 나머지 절반은 제5항에 기재된 폴리머에 의해 결장용성 피복된 약물 전달용 조성물.
- 제24항에 있어서, 캡슐의 절반은 비수용성 폴리머로 피복되고, 나머지 절반은 제4항에 기재된 폴리머에 의해 장용성 피복되거나 제5항에 기재된 폴리머에 의해 결장용성 피복된 약물 전달용 조성물.
- 삭제
- 제1항에 있어서, HPMC 캡슐이 pH < 3에서는 불용성이고 pH > 5.5에서는 용해성인 필름으로 피복되어 있는 약물 전달용 조성물.
