Abstract
본 발명은 라이노바이러스에 대한 항바이러스 활성을 가지는 바나바, 땅빈대, 마르타로즈마리 또는 산국의 추출물, 또는 이의 배합물을 포함하는 조성물 및 상기 조성물을 투여하여 바이러스 감염을 예방 또는 치료하는 방법에 관한 것이다.

Description
본 발명은 라이노바이러스에 대한 항바이러스 활성을 가지는 바나바, 땅빈대, 마르타로즈마리 또는 산국의 추출물, 또는 이의 배합물을 포함하는 조성물 및 상기 조성물을 투여하여 바이러스 감염을 예방 또는 치료하는 방법에 관한 것이다.
라이노바이러스(Rhinovirus)는 피코나비리데(Phcormaviridae)과에 속하는 바이러스로 부비강염, 발작적인 천식, 중이염, 낭성 섬유종, 기관지염 등을 수반한다.
바이러스 감염에 치료하기 위한 약물로 다음과 같은 항바이러스제들이 널리 사용되고 있다: 리바비린(Ribavirin)은 C형 간염(Foster, 2004, Semin Liver Dis. 24(suppl.2):97-104), SARS, 에볼라 바이러스(Ebola virus) 감염 치료에 이용되고 있으며(Gunther 등, 2004, Antiviral Res. 63:209-215), 아씨클로비르(acyclovir) 는 허피즈 바이러스(herpesvirus) 치료에 이용되고 있고 (Dewhurst 2004. Herpes 11:105A-111A), 아지도싸이미딘(azido thymidine)은 AIDS의 치료에 이용되고 있으며(Coovadia, 2004, N Eng J Med 351:289-292), 글리시리진(glycyrrhizin)은 SARS 치료에 이용되고 있는 약제이다(Chen 등, 2004, J. Clin. Virol.:69-75).
그러나, 상기 항바이러스제들은 이들 약물에 대한 바이러스의 내성 및 부작용으로 지속적인 사용에 문제가 있고 무엇보다 라이노바이러스에 특이적인 활성을 나타내지 않는다.
라이노바이러스 특이적인 항바이러스제의 개발도 이루어지고 있다. 라이노바이러스를 억제하는 약제는, 라이노바이러스의 3C 프로티아제를 억제하는 기능을 가지는 약제(Wang, Q. M. 등, 1998, Antimicrob. Agents Chemother; Webber, S. 등, 1996, J. Med. Chem.; Sheperd 등, 1996, Med. Chem. Lett.) 들이 주를 이루고 있다. 미국 특허 US 6369226은 리노바이러스의 3C 프로티아제를 억제하는 신규 치환된 벤자마이드 유도체에 대하여 기재하고 있다.
그 외에도, 한국특허 10-0005666-000는 시로데스미움 디베르슘(Sirodesmium diversum) 균주를 배양하여, 라이노바이러스 등의 바이러스에 항바이러스 활성을 갖는 시로데스민 유도체를 제공한다. 미국특허 US5580577은 N-(C1-4)-아실시스테인(N-(C1-4)-acylcysteine) 또는 그의 약학적으로 허용되는 염을 경구 또는 비경구적으로 유효량으로 투여하여 인간 라이노바이러스 감염 증상을 치료하는 방법을 제공한다. 국제 출원공개 WO0183529A2는 라이노바이러스 감염을 예방 또는 치료하는 신규 이뮤노에드헤신(Immunoadhesin)에 관한 발명으로, Ig의 중쇄에 연결된 리노바 이러스 수용체 단백질, 선택적으로 J 쇄 및 분비 용소를 포함하는 키메릭 ICAM-1분자를 포함하는 이뮤노에드헤신을 제공한다.
그러나, 독성과 부작용이 적고, 효과가 뛰어난 천연물 소재의 라이노바이러스에 특이적인 항바이러스제의 개발은 미흡한 상태이다.
천연물 중에서 식물의 경우, 여러 질병에 대한 치료수단으로 오랫동안 사용되어 왔고(Seeff LB 등, 2001, Hepatology, 34(3) : 595-603; Langmead L 등, 2001, Aliment Pharmacol Ther., 15(9) : 1239-52) 일부 식물의 경우, 항바이러스 활성을 가지고 있음이 보고 되었으나(Kinghorn AD, 1994, Biotechnology, 26 : 81-108; Patrick L, 1999, Altern Med Rev., 4(4) : 220-38), 식물 추출물이 기존의 항바이러스제보다 효능이 뛰어나지 않아 바이러스 감염 치료에 적용되는 사례는 아직까지 없었다.
이에, 본 발명자들은 라이노바이러스에 대하여 우수한 항바이러스 활성을 갖는 생약재를 찾고자 노력한 결과, 바나바, 땅빈대, 마르타로즈마리 또는 산국의 추출물, 또는 이의 배합물을 포함하는 조성물이 라이노바이러스에 대하여 우수한 특이적 증식 억제능을 가지고 있음을 확인함으로써 본 발명을 완성하게 되었다.
본 발명의 목적은 라이노바이러스에 대하여 항바이러스 활성을 가지는 바나바, 땅빈대, 마르타로즈마리 또는 산국의 추출물, 또는 이의 배합물을 포함하는 조 성물을 제공하는 것이다.
본 발명의 또 다른 목적은 바나바, 땅빈대, 마르타로즈마리 또는 산국의 추출물, 또는 이의 배합물을 포함하는 조성물을 투여하여 라이노바이러스 감염을 예방 또는 치료하는 방법을 제공하는 것이다.
하나의 양태로서, 본 발명은 라이노바이러스에 대하여 항바이러스 활성을 가지는 바나바, 땅빈대, 마르타로즈마리 또는 산국의 추출물, 또는 이의 배합물을 포함하는 조성물을 제공한다.
본 발명의 목적상 바이러스는 감기의 주원인균인 라이노바이러스이다. 라이노바이러스는 피코나비리데과에 속하는 대표적인 바이러스로, 단쇄 양성 극성 RNA를 게놈으로 가지며 인간에게 감염하는 가장 일반적인 바이러스다. 예를 들어, 이러한 바이러스에는 인간 라이노바이러스 A(HRV-A: Human rhinovirus A) 및 B(HRV-B: Human rhinovirus B)가 있다.
본 발명에서 용어, “항바이러스(antiviral) 활성"이란 라이노바이러스 입자 생성을 억제하는 능력, 즉 숙주세포, 특히 인간을 포함하는 포유동물에서 라이노바이러스의 형성 및/또는 복제를 효과적으로 억제하는 능력을 의미한다. 본 발명에서 제공하는 조성물은 바이러스 복제(replication) 또는 증식(proliferation) 등에 필요한 기작을 방해하여 항바이러스 활성을 가진다.
본 발명에서 용어, "추출물"은 식물로부터 분리된 활성성분, 여기서는 라이 노바이러스에 대한 항바이러스 활성을 가지는 유효성분을 가지는 물질을 의미하며, 물 또는 알콜(예: 메탄올, 에탄올, 프로판올, 부탄올 등) 등의 유기 용매를 이용하는 추출 과정에 의하여 제조된다. 추출물은 수 또는 유기용매 추출액, 이의 건조 분말 또는 이를 이용하여 제형화된 모든 형태를 포함한다.
본 발명에서 용어, “바나바(Lagerstroemia speciosa Pars)”는 천연, 잡종 또는 변종 바나바의 줄기, 잎, 꽃을 모두 포함하나 바람직하게는 바나바 잎을 의미한다. 바람직하게는, 바나바의 수추출물을 라이노바이러스에 대한 항바이러스 조성물로 사용할 수 있다.
바나바는 혈당조절능이 있어서 당뇨병 질환에 치료효과가 있다고 보고 되었고(Judy WV 등, 2003, J Ethnopharmacol.), 대한민국 특허출원 10-2004-0070652에는 당뇨병 치료 효과를 가지는, 바나바 추출물, 호로파 추출물 및 여주 추출물을 고체 발효시킨 발효물을 유효 성분으로 함유하는 혈당강하용 약학적 조성물에 대해 기재하고 있다. 그러나, 바나바에서 항바이러스 활성, 특히 라이노바이러스에 대한 항바이러스 활성을 살펴본 예는 아직까지 없다.
본 발명의 바나바의 수추출물의 경우, 감염된 세포에서 약 155.74%의 라이노바이러스 억제능(%)을 보였다. 기존의 항바이러스제인 리바비린은 2.34% 라이노바이러스 증식 억제능(%), 아만타딘은 0.69% 라이노바이러스 증식 억제능(%), 이지도데옥시 싸이미딘은 23.57% 증식 억제능(%)을 보였으며, 이에 비하여 바이러스 증식 억제능이 월등히 높음을 알 수 있다(표 3 및 4).
본 발명에서 용어, “땅빈대(Euphorbia humifusa)”는 천연, 잡종 또는 변종 땅빈대의 줄기, 잎, 꽃을 모두 포함하나 바람직하게는 땅빈대 잎을 의미한다. 바람직하게는, 땅빈대의 알콜 추출물을 라이노바이로스에 대한 항바이러스 조성물로 사용할 수 있다.
땅빈대(Euphorbia humifusa)는 탄닌성분이 있는 것으로만 알려졌을 뿐(Yoshida 등 1994, Chem pharm.Bull), 다른 약리적 특성은 알려진 바 없다.
중국특허 CN1233497A는 치질을 치료하기 위한 분말 약물에 대한 발명으로, 진주 가루(pearl powder), 사향 카테큐(musk catechu), 알륨(alum), 계관석(realgar) 및 삼칠삼(notoginseng )을 혼합하여 100 내지 140 메쉬로 분쇄한 분말과 익힌 땅빈대를 100 내지 140 메쉬로 분쇄한 분말을 혼합하여 제조하는 약물 및 방법을 기술하고 있다. 또한, 중국특허 CN1176811A은 매독, 임질을 예방 및 치료하기 위한 유칼립투스 잎(eucalyptus leaf), 땅빈대, 무화과나무(fig), 토사자(Semen Cuscutae)로 구성되는 조성물을 기술하고 있다. 그러나, 땅빈대에서 항바이러스 활성, 특히 라이노바이러스에 대한 항바이러스 활성을 살펴본 예는 아직까지 없었다.
본 발명의 땅빈대의 메탄올 추출물의 경우, 감염된 세포에서 약 100.66%의 라이노바이러스 억제능(%)을 보였다. 기존의 항바이러스제인 리바비린은 2.34% 라이노바이러스 증식 억제능(%), 아만타딘은 0.69% 라이노바이러스 증식 억제능(%), 아지도데옥시 싸이미딘은 23.57% 증식 억제능(%)을 보였으며, 이에 비하여 바이러스 증식 억제능이 월등히 높음을 알 수 있다(표 3 및 4).
본 발명에서 용어, “마르타로즈마리(Martn Rosemary)”는 천연, 잡종 또는 변종 마르타로즈마리의 줄기, 잎, 꽃을 모두 포함하나 바람직하게는 마르타로즈마리의 잎을 의미한다. 바람직하게는, 마르타로즈마리의 알콜 추출물을 라이노바이로스에 대한 항바이러스 조성물로 사용할 수 있다.
마르타로즈마리는 화장품 조성물로 많이 사용되었으나 항바이러스 활성, 특히 라이노바이러스에 대한 항바이러스 활성을 살펴본 예는 아직까지 없었다.
본 발명의 마르타로즈마리의 메탄올 추출물의 경우, 감염된 세포에서 약 76.80%의 라이노바이러스 억제능(%)을 보였다. 기존의 항바이러스제인 리바비린은 2.34% 라이노바이러스 증식 억제능(%), 아만타딘은 0.69% 라이노바이러스 증식 억제능(%), 아지도데옥시 싸이미딘은 23.57% 증식 억제능(%)을 보였으며, 이에 비하여 바이러스 증식 억제능이 월등히 높음을 알 수 있다(표 3 및 4).
본 발명에서 용어, “산국(Chrysanthemum boreale Makino)”은 천연, 잡종 또는 변종 산국의 줄기, 잎, 꽃을 모두 포함하나 바람직하게는 산국의 잎을 의미한다. 바람직하게는, 산국의 알콜 추출물을 라이노바이로스에 대한 항바이러스 조성물로 사용할 수 있다. 한국특허 출원 10-2000-0006958는 산국으로부터 고혈압 치료제인 큐맘브린 A를 제조하는 방법 및 고혈압 치료제로서의 그의 용도에 대하여 기재하고 있다. 한국특허 출원 10-2002-0023335는 산국으로부터 병원성 그람양성세균, 그람음성세균 및 진균의 생장을 저해할 수 있는 휘발성 추출물과 이를 효율적으로 추출, 정제하는 방법 및 그 추출물에 유효성분을 함유하는 항균 및 항진균용 조성물을 기술하고 있다. 그러나, 항바이러스 활성, 특히 라이노바이러스에 대한 항바이러스 활성을 살펴본 예는 아직까지 없었다.
본 발명의 산국의 메탄올 추출물의 경우, 감염된 세포에서 약 52.45%의 라이노바이러스 억제능(%)을 보였다. 기존의 항바이러스제인 리바비린은 2.34% 라이노바이러스 증식 억제능(%), 아만타딘은 0.69% 라이노바이러스 증식 억제능(%), 아지도데옥시 싸이미딘은 23.57% 증식 억제능(%)을 보였으며, 이에 비하여 바이러스 증식 억제능이 월등히 높음을 알 수 있다(표 3 및 4).
본 발명의 바나바, 땅빈대, 마르타로즈마리 또는 산국의 추출물은 각각을 단독 사용하거나 하나 이상의 성분을 배합하여 사용할 수 있다. 배합 사용할 경우에는 개개 성분들을 배합하여 추출하거나 개개 성분들로부터 각각 추출한 후 혼합할 수 있으며, 바람직하게는 최적의 조건에서 추출한 후 혼합할 수 있다.
본 발명의 바나바, 땅빈대, 마르타로즈마리 또는 산국의 추출물, 또는 이의 배합물을 포함하는 조성물은 약효를 증가시키지는 않으나 약재 조성물에 통상 사용되어 냄새, 맛, 시각 등을 향상시킬 수 있는 추가성분을 포함할 수 있다. 또한, 본 발명의 조성물은 비타민B1, B2, B6, C, E, 니아신, 카르니친, 베타인, 엽산 판토텐산, 비오틴, 아연, 철, 칼슘, 크롬, 마그네슘, 이들의 혼합물 등의 무기, 유기 첨가물들을 추가로 포함할 수 있다.
본 발명의 조성물에 포함되는 바나바, 땅빈대, 마르타로즈마리 또는 산국은 물 또는 유기 용매를 사용하여 추출하여 바로 사용될 수 있으나 보다 바람직하게는 추출액을 여과하고 여과액을 건조하여 사용한다.
바람직한 본 발명의 추출물 제조 과정은 약재를 분쇄하는 단계; 용매로 추출하는 단계; 추출액을 여과하는 단계; 및 여과액을 건조시키는 단계를 포함한다.
추출 용매로는, 물 또는 알콜 등의 유기 용매를 사용하며 생약의 유효성분이 파괴되지 않거나 최소화된 조건에서 진행하고 열추출 또는 냉침 추출한다. 여과는 추출액으로부터 부유하는 고체 입자를 제거하는 과정으로, 여과지, 면, 나일론 등을 이용하여 입자를 걸러내거나 냉동여과법, 원심분리법 등을 사용할 수 있으나 이에 제한되지 않는다. 여액을 건조하는 단계는 동결건조, 진공건조, 열풍건조, 분무건조, 감압건조, 분무건조, 포말건조, 고주파건조, 적외선건조 등을 포함하나 이에 제한되지 않는다. 건조공정을 통하여 수분함량을 10% 이하로 줄일 수 있으며, 또한 건조 전에 여과액을 농축기에 넣고 수분함량을 줄이는 공정을 추과할 수 있다. 경우에 따라, 최종 건조된 추출물을 분쇄하는 공정을 추가할 수도 있다.
수 추출하는 경우, 본 발명의 조성물의 성분들을 물에 가하여 50℃ 내지 70℃ 에서, 30분 내지 2시간 정도 가열 하는 것이 바람직하다. 높은 온도에서 가열하는 경우 시간은 줄어들지만 유효성분이 파괴되어 손실될 수 있고, 상기 온도 보다 낮은 온도에서 가열하게 되면 혼합물의 유효성분이 완전하게 추출되지 않으므로 온도 조절과 시간을 적절하게 조절하는 것이 중요하다. 본 발명에서는 바나바를 60℃에서 1시간 가열하여 추출하였고, 불용성 물질을 원심분리하여 여과하였으며 상등액을 동결건조시켜 수추출물을 제조하였다
유기 용매를 사용하여 추출하는 경우, 메탄올, 에탄올, 이소프로탄올, 부탄올, 에틸렌, 아세톤, 에테르, 헥산, 클로로포름, 에틸아세테이트, DMF(N,N-디메틸 포름아미드), DMSO(디메틸설폭사이드) 등을 이용하여 실온에서 또는 가온하여 유기 용매로 추출할 수 있다. 추출하는 유기 용매에 따라 약제의 유효성분의 추출 정도와 손실 정도가 차이가 날 수 있으므로, 알맞은 유기 용매를 선택하여 사용하도록 한다. 바람직하게는 20℃ 내지 30℃에서, 12시간 내지 48시간 정도 방치하여 추출한다. 본 발명에서는 땅빈대, 마르타로즈마리, 산국을 알콜을 사용하여 실온에서 24시간 방치하여 추출하였다. 이어서, whatman 여과지를 사용하여 불용성 물질을 여과하였으며, 여과액을 감압 건조시켜 유기용매 추출물을 제조하였다.
본 발명의 조성물은 약학적으로 허용 가능한 담체를 포함할 수 있다. 약제학적으로 허용되는 담체는 경구 투여시에는 결합제, 활탁제, 붕해제, 부형제, 가용화제, 분산제, 안정화제, 현탁화제, 색소, 향료 등을 사용할 수 있으며, 주사제의 경우에는 완충제, 보존제, 무통화제, 가용화제, 등장제, 안정화제 등을 혼합하여 사용할 수 있으며, 국소 투여용의 경우에는 기제, 부형제, 윤활제, 보존제 등을 사용할 수 있다. 본 발명의 약제학적 조성물의 제형은 상술한 바와 같은 약제학적으로 허용되는 담체와 혼합하여 다양하게 제조될 수 있다. 예를 들어, 경구 투여시에는 정제, 트로키, 캡슐, 엘릴시르, 서스펜션, 시럽, 웨이퍼 등의 형태로 제조할 수 있으며, 주사제의 경우에는 단위 투약 앰플 또는 다수회 투약 형태로 제조할 수 있다.
또 다른 양태로서, 본 발명은 상기 조성물을 라이노바이러스의 감염이 의심되는 환자에게 투여하여 라이노바이러스 감염 질환을 예방 또는 치료하는 방법을 제공한다.
본 발명에서 용어, “라이노바이러스 감염 질환”이란 라이노바이러스의 감염으로 유발되는 질환을 일컫으며, 부비강염, 발작적 천식, 중이염, 낭성 섬유종, 기관지염 등을 예시할 수 있다(Pitkaranta와 Hayden, 1998, Ann. Med.).
본 발명에서 용어, "예방"이란 조성물의 투여에 의해 바이러스 감염을 억제시키거나 발병을 지연시키는 모든 행위를 의미한다. 본 발명에서 용어, "치료"란 조성물의 투여에 의해 바이러스 감염에 의한 증세가 호전되거나 이롭게 변경하는 모든 행위를 의미한다.
상기 조성물의 투여 경로는 목적 조직에 도달할 수 있는 한 어떠한 일반적인 경로를 통하여 투여될 수 있다. 복강내 투여, 정맥내 투여, 근육내 투여, 피하 투여, 피내 투여, 경구 투여, 국소 투여, 비내 투여, 폐내 투여, 직장내 투여 될 수 있으나, 이에 제한되지는 않는다. 또한 제약 조성물은 활성 물질이 표적 세포로 이동할 수 있는 임의의 장치에 의해 투여될 수 있다.
본 발명의 조성물은 약제학적으로 유효한 양으로 투여한다.
용어 "약제학적으로 유효한 양"은 의학적 치료에 적용 가능한 합리적인 수혜/위험 비율로 질환을 치료하기에 충분한 양을 의미하며, 유효 용량 수준은 환자의 질병 종류 및 중증도, 연령, 성별, 약물의 활성, 약물에 대한 민감도, 투여 시간, 투여 경로 및 배출 비율, 치료 기간, 동시 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 결정될 수 있다. 본 발명의 조성물은 개별 치료제로 투여하거나 다른 치료제와 병용하여 투여될 수 있고 종래의 치료제와는 순 차적 또는 동시에 투여될 수 있다. 그리고 단일 또는 다중 투여될 수 있다. 상기 요소를 모두 고려하여 부작용 없이 최소한의 양으로 최대 효과를 얻을 수 있는 양을 투여하는 것이 중요하며, 당업자에 의해 용이하게 결정될 수 있다.
일반적으로, 1일 당 20 내지 100 ㎎/kg, 바람직하게는 40 내지 60 ㎎/kg, 보다 바람직하게는 50 ㎎/kg의 양을 투여되도록 하며, 1회 내지 4회 분할 투여할 수 있다.
이하, 본 발명의 이해를 돕기 위하여 바람직한 실시예를 제공한다. 그러나 하기의 실시예는 본 발명을 보다 쉽게 이해하기 위하여 제공되는 것일 뿐, 본 발명이 하기의 실시예에 의하여 제한되는 것은 아니다.
참조예 1: 수용성 추출물의 제조
경남농업기술원 약용식물 재배포에서 채취한 재료(마황의 잎, 삼백초의 잎, 조각자의 열매, 사철쑥의 잎, 감초의 뿌리, 박하의 잎, 사자발쑥의 잎, 싸주아리쑥의 잎)와 허브랜드(주)에서 수확한 재료(루의 잎, 레이디스 맨틀의 잎, 마르타 로즈마리의 잎, 슈퍼민트의 잎)와 외국에서 직수입한 바나바의 잎을 생초 중량의 25%가 될 때까지 건조시킨 다음 믹서로 갈아서 분말로 만들었다. 각 식물 분말은 각각의 중량에 대해 증류수 10배량을 넣어 60 ℃에서 1 시간동안 중탕하고 12000 rpm으로 20 분간 원심분리하여 불용성 물질을 제거하고 상등액을 동결건조시켜 수용성 추출물을 제조하였다(표 1).
참조예 2 : 메탄올 추출물의 제조
경남농업기술원 약용식물 재배포에서 채취한 재료(땅빈대의 잎, 사자발쑥의 잎, 산국의 잎, 싸주아리쑥)를 생초 중량의 25%가 될 때까지 건조시킨 다음 믹서로 갈아서 분말로 만들었다. 각 식물 분말은 각각의 중량에 대해 메탄올 10배량을 넣어 실온에서 24시간 방치한 후에 whatman 여과지로 여과하여 불용성 물질을 제거하고 여과된 액은 감압추출기로 건조하여 메탄올 추출물액을 제조하였다(표 1).
참조예 3: 대조군으로 사용된 항바이러스제
각 식물 추출물의 바이러스 증식 억제능을 비교측정하기 위하여 현재 항바이러스제로 사용되는 있는 약제(Ribavirin, amantadine, azidodeoxy thymidine)를 Sigma사로부터 구입하여 세포 배양용액에 희석하여 사용하였다(표 3)
참조예 4: 바이러스의 배양
상기 참조예 1, 2에서 얻은 18종의 식물 추출물에 대한 항바이러스능을 측정하기 위해 사용된 바이러스는 라이노바이러스를 대상으로 하였으며, 라이노바이러스는 HeLa세포에 감염시켜서 증식배양하였다. HeLa세포는 소아태아혈청이 10% 포함된 MEM medium 배지를 사용하여 37 ℃ 배양기에서 배양하였으며 바이러스 증식배양 및 항바이러스능 측정에 사용된 배지는 2% 혈청에 30mM MgCl2가 포함된 배양배지를 사용하였다.
실시예 1: 바이러스 억제능 분석
상기 참조예 1, 2에서 얻은 각 식물 추출물의 라이노바이러스에 대한 바이러스 증식억제능을 측정하기 위해서, HeLa세포를 2x104 개의 세포를 96 웰 플레이트(well plates) 의 각 웰에 넣고 24시간 배양하였다. 24시간 후에 각 웰의 배양 상등액을 제거하고 라이노바이러스 감염용액을 각 웰에 넣은 다음, 각 식물 추출물을 50 ㎍/㎖ media) 이 되도록 각 웰에 투여하였다. 각 추출물의 바이러스 증식 억제능 측정은 72시간 후에 살아있는 세포를 SRB 분석법[Martin A 등, 1997, Cytotechnology, 11 : 49-54]으로 측정하였다. 각 웰에 10% TCA(trichloroacetic acid)를 100 ㎕씩 첨가한 후 1시간 동안 4 ℃에 방치하고 증류수로 수회 세척하였다. 실온에서 건조시킨 후 1%(v/v) 아세트산에 녹인 0.4% (w/v) SRB(sulforhodamine B) 용액 100 ㎕를 첨가해 30분 동안 염색시켰다. 세포와 결합하지 않은 SRB 염색액은 1%(v/v) 아세트산으로 수회 세척한 다음 다시 건조시켰다. 각 웰에 10 mM 트리스 용액(pH 10.5) 100 ㎕를 각 웰에 가하여 세포와 결합되어 있는 염색제를 충분히 녹인 후 560 nm에서 흡광도를 측정하였다. 각 처리군은 바이러스를 처리하지 않은 군(A), 추출물만을 처리한 군(B), 바이러스만을 처리한 군(C), 바이러스와 추출물을 같이 처리한 군(D)으로 표기하였고 각각 식물추출물의 비교세포성장률(%)[수학식 1]과 식물추출물의 바이러스에 대한 증식억제능(%)[수학식 2]으로 계산하였다. 결과에 대한 통계는 동일한 조건에서 수행한 3번의 실험결과 수치에 대한 평균값과 표준편차로 계산한 값을 제시하였다.
각 생약 추출물들의 세포독성
상기 참조예 1, 2에서 분리한 각 식물의 수용성 추출물, 메탄올 추출물, 항바이러스제에 대한 세포독성능은 50 ㎍/㎖의 농도에서 추출물을 가하지 않은 웰의 세포와 비교하였다. 상기 식물 추출물과 항바이러스 약제들은 시험한 농도에서 바이러스 배양세포로 사용되는 HeLa세포에 대해 세포독성능을 나타내지 않았거나 또는 낮은 세포독성능을 나타내거나 또는 높은 세포독성능을 나타내었다(표 2). HeLa세포에 대하여 실험한 상기 농도에서 약제나 추출물을 가하지 않은 대조군에 비하여 세포의 성장률이 억제되지 않거나 오히려 증가한 경우는 마황, 초피나무, 사철쑥, 바나바, 인동덩굴, 오가피 였으며, 산국, 마르타로즈마리, 슈퍼민트는 부분적인 성장 제한성을 보였다.
기존 항바이러스제의 라이노바이러스에 대한 항바이러스능 분석
기존 항바이러스제로 사용되는 3종의 약제를 라이노바이러스 증식억제능 시험에 사용한 결과에서 일반적으로 호흡기질환 바이러스 억제제로 사용되는 리바비린 (2.34±3.69%) 과 아만타딘 (0.69±6.12%)는 전혀 효능을 나타내지 못했으며 아지도데옥시 싸이미딘 만이 부분적으로 라이노바이러스의 증식을 억제하였다 (23.57±8.56%)(표 3).
식물 추출물의 라이노바이러스에 대한 항바이러스능 분석
참조예 1, 2에서 얻은 추출물의 라이노바이러스에 대한 항바이러스능을 각각 표 4에 나타내었다. 증식억제능 시험은 50ug/ml의 농도에서 실시하였다. 22종의 식물군에서 기존 항바이러스제인 아지도데옥시 싸이미딘의 효능(23.57%)과 비교할 때 보다 같은 농도에서 보다 높은 바이러스 증식억제능을 보인 식물은 마황, 바나바, 감초, 땅빈대, 산국, 마르타로즈마리였다. 이중 마황과 감초는 일부 성분이 감기에 대하여 진정작용이 있음이 보고되고 있어서 전통적인 감기 치료에 이용되는 경우도 있으나 바나바, 땅빈대, 산국, 마르타로즈마리는 감기 등 호흡기질환에 대한 전통적인 치료법에서도 사용된 바 없었다.
따라서, 본 발명에서는 바나바, 땅빈대물 산국, 마르타로즈마리 추출물들이 라이노바이러스에 대하여 새로운 억제효능을 갖고 있음을 밝혀냈으며 항바이러스제로 사용되는 아지도데옥시 싸이미딘의 효능과 비교하여 2배 이상의 라이노바이러스 억제효능을 보임으로써 이들 4종의 식물 추출물들이 산업적으로 유용한 가치를 가지고 있음을 확인하였다(표 4).
본 발명의 바나바, 땅빈대, 마르타로즈마리 또는 산국의 추출물 또는 이의 배합물을 포함하는 조성물은 감기 등의 호흡기질환을 유발하는 주원인 바이러스인 라이노바이러스 감염을 효과적으로 예방 및 치료할 수 있으며, 독성 및 부작용이 적다는 장점이 있다.
Claims (10)
- 라이노바이러스에 대하여 항바이러스 활성을 가지는 바나바, 땅빈대, 마르타로즈마리 또는 산국의 잎 추출물, 또는 이의 배합물을 포함하는 호흡기 질환의 치료 또는 예방용 약제학적 조성물.
- 제1항에 있어서, 바나바의 추출물이 수추출물인 조성물.
- 제1항에 있어서, 땅빈대의 추출물이 알콜 추출물인 조성물.
- 제1항에 있어서, 마르타로즈마리의 추출물이 알콜 추출물인 조성물.
- 제1항에 있어서, 산국의 추출물이 알콜 추출물인 조성물.
- 제1항에 있어서, 라이노바이러스가 HRV-A(Human rhinovirus A) 또는 HRV-B(Human rhinovirus B)인 조성물.
