Claims (13)
- 다음 단계로 이루어진 2',3'-디데히드로-3'-데옥시티미딘(d4T)의 개선된 제조방법 :a) N-메틸-모르폴린, 피콜린, 및 루티딘중에서 선택된 유기 염기의 존재하에 티미딘(V)을 메실 클로라이드와 반응시켜 3',5'-디-0-메실티미딘(Ⅳ)을 제공하고;b) 비스-메실레이트(Ⅳ)를 수성 히드록시드로서 처리하여 3',5'-안히드로티미딘(Ⅲ)은 형성하고,c) 화합물(Ⅲ)을 알코올릭 히드록시드에서 가열함으로서 화합물(Ⅲ)을 d4T· NMPO 용매화물(Ⅱ)로 전환하고 산중화후에, 반응 혼합물을 N-메틸피롤리디논과 비양성자성 에스테르, 아미드 또는 케톤 용매로서 처리하여 화합물(Ⅱ)을 침전시키고;d) 알코올릭 용액에서 분리된 d4T·NMPO 용매화물(Ⅱ)을 가열하여 고순도의 d4T(I)를 얻는다.
- 제 1항에 있어서, 단계 (a)에서 유기 염기가 N-메틸모르폴린인 제조방법.
- 제 1항에 있어서, 단계 (c)의 d4T·NMPO 용매화물(Ⅱ)을 아세톤 슬러리로부터 여과에 의해 분리하는 제조방법.
- 제 1항에 있어서, d4T·NMPO 용매화물(Ⅱ)을 단계 (d)에서 이소프로판올내 d4T(I)로 전환하는 제조방법.
- 공정 반웅 혼합물로부터 d4T(I)의 N-메틸피롤리디논 용매화물의 형성 및 제거에 이어서 알코올릭 매질에서 분리된 d4T·NMPO 용매화물(Ⅱ)을 가열함으로서 정제된 d4T(I)의 생성으로 이루어진 원 d4T의 분리 및 정제방법.
- 제 5항에 있어서, 알코올릭 매질이 이소프로판올인 분리 및 정제방법.
- 다음 단계로 이루어진 3',5'-안히드로티미딘으로부터 2',3'-디데히드로-3'-데옥시티미딘(d4T)의 개선된 제조방법:(a) 알코올릭 히드록시드에서 3',5'-안히드로티미딘(Ⅲ)을 가열하고;(b) 산으로 중화한다음 불용성 염과 알코올을 제거하고;(c) N-메틸피롤리디논과 아세톤으로 처리하여 공정 반웅 혼합물로부터 분리되는 d4T·NMPO 용매화물(Ⅱ)을 침전시키며;(d) 알코올에서 d4T·NMPO 용매화물(Ⅱ)을 가열하여 d4T(I)을 생성한다.
- 제 7항에 있어서, 단계 (a)의 알코올이 이소프로판올인 제조방법.
- 제 7항에 있어서, d4T·NMPO 용매화물(Ⅱ)을 단계 (c)에서 아세톤 슬러리로부터 여과에 의해 분리하는 제조방법.
- 제 7항에 있어서, 단계 (d)의 알코올이 이소프로판올인 제조방법.
- 다음식의 d4T·NMPO 용매화물(Ⅱ)
- 반웅 혼합물로부터 제 11항의 d4T·NMPO 용매화물(Ⅱ)의 분리, 이어서 d4T로 그의 전환을 이용하는 d4T의 분리 및 정제방법.
- 알코올에서 제 11항의 분리된 d4T·NMPO 용매화물(Ⅱ)을 가열하는 것으로 구성된 d4T(I)의 제조방법.
