Abstract
Description
Claims (6)
- (i) 항균 펩타이드의 양전하를 실질적으로 중화시킬 수 있으며 하나 이상의 S-S 결합을 가지는 산성 펩타이드의 유전자와 양전하를 띠는 항균 펩타이드의 유전자를 연결하여 융합유전자를 제조하는 공정; (ii) 융합유전자를 벡터에 클로닝하고, 그것을 미생물에 도입하여 발현시키는 공정; 및, (iii) 발현된 융합 펩타이드를 분리하여 상기 산성 펩타이드를 제거하고, 염기성의 항균 펩타이드를 수득하는 공정으로 이루어지는 항균 펩타이드의 제조방법.
- 제1항에 있어서, 상기 산성 펩타이드의 유전자는, 상기 융합 펩타이드로부터 항균 펩타이드를 분리할 수 있는 효소 또는 화합물에 의해 절단되는 잔기를 암호화하는 유전자가 부가된 것을 특징으로 하는 항균 펩타이드 제조방법.
- 제1항에 있어서, 상기 항균 펩타이드의 유전자는 N말단에, 상기 융합 펩타이드로부터 항균 펩타이드를 분리할 수 있는 효소 또는 화합물에 의해 절단되는 잔기를 암호화하는 유전자가 부가된 것을 특징으로 하는 항균 펩타이드 제조방법.
- (i) 항균 펩타이드의 양전하를 실질적으로 중화시킬 수 있으며 하나 이상의 S-S 결합을 가지는 산성 펩타이드의 유전자와 양전하를 띠는 항균 펩타이드의 유전자를 연결하여 융합유전자를 제조하는 공정; (ii) 상기 융합유전자의 중합체를 제조하는 공정; (iii) 상기 융합유전자 중합체를 벡터에 클로닝하고, 그것을 미생물에 도입하여 발현시키는 공정; 및, (iv) 발현된 융합 펩타이드를 분리하여 산성 펩타이드를 제거하고, 염기성의 항균 펩타이드를 수득하는 공정으로 이루어지는 항균 펩타이드의 제조방법.
- 유전자에 작동가능하도록 연결된 프로모터와 유전자로 이루어지며, 상기 유전자가 항균 펩타이드 유전자의 양전하를 중화시킬 수 있으며 하나 이상의 S-S결합을 가지는 산성 펩타이드 유전자와 항균 펩타이드 유전자로 이루어지는 DNA 플라즈미드.
- 제5항에 있어서, 상기 유전자가 항균 펩타이드 유전자의 양전하를 중화시킬수 있는 산성 펩타이드의 유전자와 항균 펩타이드 유전자로 이루어진 유전자의 중합체 형태인 DNA 플라즈미드.
