Claims (16)
- 프레그난-3,20-디온의 3-케토 위치에서 화학 선택적 반응 및 디아스테레오머 선택적 반응을 통해 3(R)-프레그난-3-스피로-2'옥시란-20-온을 형성하고, 이 3(R)-프레그난-3-스피로-2'옥시란-20-온을 20-케토 위치를 보호하지 않고 부분 선택적 방식으로 개환시키는 것을 특징으로 하는, 3α-히드록시, 3β-치환 프레그난의 제조 방법.
- A) 트리메틸 술폭소늄 요오다이드를 DMSO 중에서 NaH와 반응시키거나 또는 THF 중에서 포타슘 t-부톡시드와 반응시켜 일리드를 형성하는 단계,B) 상기 일리드를 프레그난-3,20-디온과 반응시켜 (3R)-프레그난-3-스피로-2'옥시란-20-온을 형성하는 단계, 및C) 상기 3(R)-프레그난-3-스피로-2'옥시란-20-온을 1,2-디메톡시에탄 중에서 요오드화나트륨 및 트리부틸 틴 하이드라이드와 반응시키는 단계로 이루어지는 것을 특징으로 하는, 3α-히드록시, 3β-메틸-프레그난-20-온의 제조 방법.
- A) 트리메틸 술폭소늄 요오다이드를 DMSO 중에서 NaH와 반응시키거나 또는 THF 중에서 포타슘 t-부톡시드와 반응시켜 일리드를 형성하는 단계,B) 상기 일리드를 프레그난-3,20-디온과 반응시켜 (3R)-프레그난-3-스피로-2'옥시란-20-온을 형성하는 단계, 및C) 상기 3(R)-프레그난-3-스피로-2'옥시란-20-온을 THF와 메탄올의 혼합물 중에서 요오드화나트륨, 수소 가스 및 탄소 기재 팔라듐과 반응시키는 단계로 이루어지는 것을 특징으로 하는, 3α-히드록시, 3β-메틸-프레그난-20-온의 제조 방법.
- A) 트리메틸 술폭소늄 요오다이드를 적절한 용매 중에서 적절한 염기와 반응시켜 일리드를 형성하는 단계,B) 상기 일리드를 프레그난-3,20-디온과 반응시켜 (3R)-프레그난-3-스피로-2'옥시란-20-온을 형성하는 단계, 및C) 상기 3(R)-프레그난-3-스피로-2'옥시란-20-온을 적절한 용매 중에서 상기 옥시란의 3'위치에서 반응시켜 상기 옥시란 고리를 개환시킬 수 있는 친핵체와 반응시키는 단계로 이루어지는 것을 특징으로 하는, 3α-히드록시, 3β-치환-프레그난-20-온의 제조 방법.
- 제4항에 있어서, 단계(A)의 적절한 염기가 NaH, 포타슘 t-부톡시드 및 NaNH2로 이루어진 군으로부터 선택되는 것인 방법.
- 제4항에 있어서, 단계(C)의 친핵체가 알콕시드, 티오알콕시드, 아지드, 시아나이드, 이소시아나이드, 아민 및 할라이드 음이온으로 이루어진 군으로부터 선택되는 것인 방법.
- 제6항에 있어서, 3α-히드록시, 3β-치환-프레그난-20-온이 3β-에틸기를 함유하고 단계(C)의 친핵체가 디메틸 리튬 쿠프레이트인 방법.
- 제6항에 있어서, 3α-히드록시, 3β-치환-프레그난-20-온이 3β-브로모메틸기를 함유하고 단계(C)의 친핵체가 브롬화나트륨인 방법.
- 제6항에 있어서, 3α-히드록시, 3β-치환-프레그난-20-온이 3β-아지도메틸기를 함유하고 단계(C)의 친핵체가 소듐 아지드 또는 트리메틸실릴 아지드인 방법.
- 제6항에 있어서, 3α-히드록시, 3β-치환-프레그난-20-온이 3β-프로필기를 함유하고 단계(C)의 친핵체가 디에틸 리튬 쿠프레이트인 방법.
- 제6항에 있어서, 3α-히드록시, 3β-치환-프레그난-20-온이 3β-트리플루오로에틸옥시메틸기를 함유하고 단계(C)의 친핵체가 소듐 트리플루오로에톡시드인 방법.
- 제6항에 있어서, 3α-히드록시, 3β-치환-프레그난-20-온이 3β-요오도메틸기를 함유하고 단계(C)의 친핵체가 요오드화 나트륨인 방법.
- 제12항에 있어서, 3β-요오도메틸기를 추가로 소듐 벤질옥시드와 반응시켜 3β-벤질옥시기를 형성하는 것인 방법.
- 제13항에 있어서, 3β-벤질옥시기를 가수소분해시켜 3β-히드록시메틸기를 형성하는 것인 방법.
- 제12항에 있어서, 3β-요오도메틸기를 추가로 소듐 메톡시드와 반응시켜 3β-메톡시메틸기를 형성하는 것인 방법.
- 제4항에 있어서, 3α-히드록시, 3β-치환-프레그난-20-온의 20-케토기를 상기 옥시란 고리의 형성 후에 추가로 환원, 축합, 산화, 치환 또는 제거시키는 것인 방법.
