- 7-아미노-3-[(1-메틸-1-피롤리디니오)-메틸]세프-3-엠-4-카르복실레이트의 실릴레이션된 유도체와 실질적으로 안티-이성질체가 섞이지 않은 2-(2-아미노티아졸-4-일)-2-메톡시이미노 아세틸 클로라이드 하이드로클로라이드의 신-이성질체를 불활성 유기 용매에서 반응시키는 것으로 이루어지는, 실질적으로 안티-이성질체와 △2이성질체가 섞이지 않은 세페핌 디하이드로클로라이드 수화물 항생제를 제조하는 방법.
- 제1항에 있어서, 추가로, 2-(2-아미노티아졸-4-일)-2-메톡시이미노 아세트산의 신-이성질체의 무수 산 하이드로클로라이드 염과, 적어도 1몰 당량의 옥사릴 클로라이드와 적어도 1몰 당량 내지 옥사릴 클로라이드보다 약간 과량의 디메틸포름아미드를 함유하는 혼합물을 -10℃ 이하의 불활성 유기 용매에서 반응시켜 상기 실질저으로 안티-이성질체가 섞이지 않은 2-(2-아미노티아졸-4-일)-2-메톡시이미노 아세틸 클로라이드 하이드로클로라이드의 신-이성질체를 제조하는 것을 특징으로 하는 제조방법.
- 제1항에 있어서, 추가로 7-아미노-3-[(1-메틸-1-피롤리디니오)-메틸]세프-3-엠-4-카르복실레이트 염과 실릴레이팅제(silylating agent)를 불활성 유기 용매에서 반응시켜 상기 실릴레이션된 유도체를 제조하는 것을 특징으로 하는 제조방법.
- 제1항에 있어서, 추가로, 1몰 당량 이하의 염기를 첨가하는 것을 특징으로 하는 제조방법.
- 제1항에 있어서, 추가로, 유기액-수용액층의 이층 용액을 생성하기 위해 반응 혼합물에 충분한 양의 물을 첨가하고, 분리한 수용액층에 충분한 양의 산 또는 잘녹고 비독성인 그것의 염을 첨가하는 것을 특징으로 하는 제조방법.
- 제5항에 있어서, 상기 산이 상기 항생제를 제조하기 위한 염산인 것을 특징으로 하는 제조방법.
- 제5항에 있어서, 상기 산이 황산인 것을 특징으로 하는 제조방법.
- 제7항에 있어서, 추가로, 결과물인 황산염을 염기로 중화시키고 상기 항생제를 생산하기 위한 충분한 양의 염산을 첨가하는 것으로 이루어진 제조방법.
- 제8항에 있어서, 상기 염기가 약염기성 이온 교환 수지인 것을 특징으로 하는 제조방법.
- 제2항에 있어서, 상기 옥사릴 클로라이드의 양이 1.0-2.0몰당량이고 디메틸포름아미드의 양이 상기 옥사릴 클로라이드보다 약간 과량의 몰인 것을 특징으로 하는 제조방법.
- 제10항에 있어서, 상기 옥사릴 클로라이드의 양이 1.05몰당량이고 디메틸포름아미드의 양이 상기 무수 산 하이드로클로라이드 염의 1.075당량인 것을 특징으로 하는 제조방법.
- 제2항에 있어서, 상기 온도가 -15℃--40℃인 것을 특징으로 하는 제조방법.
- 제3항에 있어서, 상기 실릴레이션제가 트리메틸클로로실란과 헥사메틸디실라잔의 혼합물인 것을 특징으로 하는 제조방법.
- 제4항에 있어서, 상기 염기가 n-메틸몰포린 또는 트리에틸아민인 것을 특징으로 하는 제조방법.
- 제13항에 있어서, 상기 실릴레이션제의 양이 약 2.0-2.5몰당량인 것을 특징으로 하는 제조방법.
- 제1-3항에 있어서, 상기 불활성 유기 용매가 아세토니트릴 또는 디클로로메탄인 것을 특징으로 하는 제조방법.
- 제1항에 있어서, 상기 항생제가 2.5-7.0무게%의 물을 함유하는 것을 특징으로 하는 제조방법.
- 제1항에 있어서, 상기 항생제가 세페핌 디하이드로클로라이드 일수화물인 것을 특징으로 하는 제조방법.
- 제1항에 있어서, 상기 항생제가 세페핌 디하이드로클로라이드 이수화물인 것을 특징으로 하는 제조방법.
