Abstract

PURPOSE:To measure human chorionic gonadotropin (hCG) accurately with high sensitivity by a sandwich method by producing the antibody reacting specifically with the hCG out of the antibodies held on a carrier and marked antibodies by a specific method and using the same. CONSTITUTION:Reptide consisting of about 30 members of amino acids at the C terminal of the hCG-beta expressed by the formula is synthesized by a conventional method. The antibody which is obtd. by inoculating said peptide and protein for a carrir (bovine thyroglobulin, etc.), which are condensated in the presence of glutaraldehyde, in a warm-blooded animal (a rabbit or the like) and reacts specifically with the hCG is obtained. Such antibody is deposited on polyestyrene particles. On the other hand, the antibody consisting of marking the antibody obtd. by immunizing the hCG obtd. from human urine in an eel with beta-D-galactosidase, etc. is obtd. The deposited antibody, the marked antibody, and the hCG in the specimen (bodily fluid such as serum) are caused to react, and the activity of the mark bound with the carrier or the activity of the mark in the unreacted marked antibody is measured. It is measured accurately without disturbance of other protein and the like causing cross reaction with the hCG, and the diagnosis of pregnancy and the like are accomplished accurately.
