Abstract
PURPOSE: To obtain the titled compound useful as an intermediae for cephalosporin antibiotic substances in high yield, by reacting phenylacetamide with paraformaldehyde in the presence of relatively reduced amounts of polyphosphoric acid and an alkylbenzene.
CONSTITUTION: Phenylacetamide is reacted with paraformaldehyde or the phenylacetamide is previously reacted with a formaldehyde to form an N-hydroxymethylphenylacetamide of formula II, which is then reacted in the presence of polyphosphoric acid and an alkylbenzene, e.g. toluene, ethylbenzene or mixed xylene, at 95W150°C for 1W40hr and dehydrated to give the aimed compound of formula III. The polyphosphoric acid to be used is a viscous syrupy substance obtained by dehydrating and concentrating orthophosphoric acid under heating, and preferably contains 77W86wt% P2O5. The amount thereof to be used is 150W270g based on one mole compund of formulaIor II.
