Abstract

PURPOSE: To eliminate the scattering of measured values, by employing a reagent composition containing ninhydrin in the quantity within specified limits and thereby preventing the crystallization and sedimentation in the liquid-sending tube of an automatic amino acid analyzer, as well as the turbidily formation and the change in the liquid flow rate in an absorbance-measuring system.
CONSTITUTION: The quantity of ninhydrin in the ninhydrin reagent used for analysis of amino acids is set to be 0.5W1.5% (weight/volume) which is less than that in the conventional one. In this way, the crystallization and sedimentation caused by the combination of ninhydrin and hydrinduntin formed by the reduction of the former with a reducing agent present in the reagent are prevented. By setting the quantity of ninhydrin within the limits as mentioned above, the composition whose analytical sensitivity (strength of coloration) is not changed and which has excellent stability with the passage of time can be obtained.
