Abstract

PURPOSE: To measure the concentration with high precision using a spectrophotometer by separating and making azidomethoglycocylhemoglobin and azidomethohemoglobin effuse when glycocylhemoglobin is measured by chromatographic method.
CONSTITUTION: Specimens containing azidomethoglycocylhemoglobin and azidomethohemoglobin obtained by methoxylating with sodium azide, glycocylhemoglobin and hemoglobin contained in a hemolytic solution of erythrocytes, are obtained. Next, these specimens are supplied to a column containing weak acidic cation exchange resin as a filler or a liquid chromatographic device within a temperature range of 25°CW45°C (selective measurement temperature to be maintained within precison range of ±1°C). Then azidomethohemoglobin is caused to effuse with a developer (buffersolution) containing sodium axide. Thus it is possible to measure the concentration with high precision using the spectrophotometer.
