Abstract
PURPOSE: Penicillin acylase is fixed on a water-insoluble aminated polyacrylo-nitrile polymer, thereby producing high-purity 6-aminopenicillanic acid in high yield.
CONSTITUTION: Penicillin acylase collected from cells of a bacterium producing penicilline acylase or the culture filtrate is fixed on a water-insoluble aminated acrylonitrile polymer that is made into fiber, membrane or hollow fiber and has porous structure with 100W200 micron pore diameter by physical adsorption or covalent bonds. The fixed enzyme is filled in a column and the pH is adjusted to 6.5W9.5 and an aqeuous solution of penicillin or its water-soluble salt of 3W 12W/V% concentration whose pH is adjusted to 6.5W9.5 is passed at 15W40°C and a low rate of 100W300 bed volume/hr for 1W10hr until more than 90% of penicillin is converted into 6-aminopenicillanic acid. Then the 6-aminopenicillanic acid is isolated and purified.
