Claims (1)
- 【特許請求の範囲】 1.真核細胞で通常発現する蛋白質をコードする合成遺伝子であって、該蛋白 質をコードする天然の遺伝子中の好ましくないコドンまたは比較的好ましくない コドンの少なくとも1個が、同じアミノ酸をコードする好ましいコドンで置換さ れた合成遺伝子。 2.同一条件下のインビトロ哺乳動物細胞培養系において天然遺伝子によって 発現されるレベルの少なくとも110%のレベルで真核生物蛋白質を発現させるこ とができる、請求項1記載の合成遺伝子。 3.同一条件下のインビトロ細胞培養系において天然遺伝子によって発現され るレベルの少なくとも150%のレベルで真核生物蛋白質を発現させることができ る、請求項1記載の合成遺伝子。 4.同一条件下のインビトロ細胞培養系において天然遺伝子によって発現され るレベルの少なくとも200%のレベルで真核生物蛋白質を発現させることができ る、請求項1記載の合成遺伝子。 5.同一条件下のインビトロ細胞培養系において天然遺伝子によって発現され るレベルの少なくとも500%のレベルで真核生物蛋白質を発現させることができ る、請求項1記載の合成遺伝子。 6.同一条件下のインビトロ細胞培養系において天然遺伝子によって発現され るレベルの少なくとも10倍のレベルで真核生物蛋白質を発現させることができる 、請求項1記載の合成遺伝子。 7.天然遺伝子におけるコドンの少なくとも10%が好ましくないコドンである 、請求項1記載の合成遺伝子。 8.天然遺伝子におけるコドンの少なくとも50%が好ましくないコドンである 、請求項1記載の合成遺伝子。 9.天然遺伝子に存在する好ましくないコドンおよび比較的好ましくないコド ンの少なくとも50%が好ましいコドンで置換されている、請求項1記載の合成遺 伝子。 10.天然遺伝子に存在する好ましくないコドンおよび比較的好ましくないコド ンの少なくとも90%が好ましいコドンで置換されている、請求項1記載の合成遺 伝子。 11.蛋白質が緑色蛍光蛋白質である、請求項1記載の合成遺伝子。 12.通常は真核生物細胞によって発現される蛋白質をコードする合成遺伝子を 調製する方法であって、該蛋白質をコードする天然の遺伝子における好ましくな いコドンおよび比較的好ましくないコドンを同定し、1つ又は複数の該好ましく ないコドンおよび比較的好ましくないコドンを置換コドンとして同じアミノ酸を コードする好ましいコドンで置換することを含む、方法。
