Description
Claims (1)
- 【特許請求の範囲】 1.赤芽系前駆体の集団を含む細胞を赤芽系細胞の増殖に必要な通常の成分を含 む培地において、エストロゲンレセプターの少なくとも一つのリガンドおよびグ ルココルチコイドレセプターの少なくとも一つのリガンドおよびチロシンキナー ゼレセプターの少なくとも一つの、好ましくは少なくとも二つのリガンドを含む 増殖因子の組合せに対し、少なくとも細胞がそれ自身で複製を始めるまで晒し、 必要ならば、細胞をさらに持続した自己−複製に必要な因子を含む培地で培養す ることを特徴とする赤芽系統の非−永久化造血性前駆細胞のインビトロ産生方法 。 2.細胞がヒトの細胞である請求項1の方法。 3.使用される出発細胞が、CD34−陽性細胞の濃縮物を有する細胞集団である請 求項2の方法。 4.骨髄、末梢血管または臍帯血液からの細胞集団を使用する請求項3の方法。 5.組合せ因子がチロシンキナーゼレセプターの少なくとも二つのリガンドを含 む請求項1−4のいずれか1の方法。 6.組合せ因子が様々なクラスのチロシンキナーゼレセプターからのレセプター と結合する少なくとも二つのリガンドを含む請求項5の方法。 7.リガンドが異なった構造のキナーゼ領域を有するレセプターと結合する群か ら選択される請求項6の方法。 8.リガンドが、以下の群: i) 連続するキナーゼ領域を有するチロシンキナーゼレセプターと結合 するリガンド;および ii) 挿入部により中断されたキナーゼ領域を有するチロシンキナーゼレ セプターと結合するリガンド から選択される請求項7の方法。 9.組合せ因子が、以下のもの: i) EGF レセプターおよび/またはHGF レセプターの類からのレセプタ ーのリガンド、および ii) c-キットのリガンド を含む請求項8の方法。 10. 組合せ因子が、以下のもの: i) TGF αおよび/またはEGF および/またはHGF、 ii )SCF、 iii )デキサメタゾンおよび/またはヒドロコーチゾンおよびエストラ ジオール を含む請求項9の方法。 11.組合せ因子がまた、自己-複製能力の獲得を促進する他の因子一つ以上を含 む請求項1−10のいずれか1の方法。 12.そのまたはそれぞれの追加の因子がサイトキンおよび/またはチロシンキナ ーゼのリガンドおよび/またはセリンキナーゼレセプターの群から選択される請 求項11の方法。 13.サイトキンがエリトロポエチンである請求項12の方法。 14.チロシンキナーゼレセプターリガンドがIGF-1 である請求項12の方法。 15.組合せ因子が、以下のもの: i) TGF αおよび/またはEGF および/またはHGF、 ii )SCF、 iii )デキサメタゾンおよびエストラジオール iv )エリトロポエチンおよびIGF-1 を含む請求項10−14のいずれか1の方法。 16.自己−複製能力を細胞の持続した自己- 複製に必要な因子の存在下に獲得し た後、細胞をさらに培養する請求項1−15のいずれか1の方法。 17. EGFレセプターまたはHGF レセプター、SCF 、エリトロポエチンおよびIGF- 1の類のリガンド少なくとも一つの存在下にヒト細胞をさらに培養する請求項16 の方法。 18.リガンドがEGF および/またはTGF αおよび/またはHGF である請求項17の 方法。
