- 【特許請求の範囲】 1.ヒト免疫不全ウイルス(HIV)糖タンパク質gp120と免疫反応でき、かつHIV を中和できるヒトモノクローナル抗体であって、 そのモノクローナル抗体が配列番号66、67、68、70、72、73、74、75、78及び 97、並びにこれらの保存置換からなる群から選ばれたH鎖免疫グロブリン可変領 域アミノ酸残基配列を含むモノクローナル抗体の結合特異性を有することを特徴 とするヒトモノクローナル抗体。 2.モノクローナル抗体が配列番号66:95、67:96、72:102、66:97、73:107、74: 103、70:101、68:98、75:104、72:105、78:110、66:118、66:122、66:121、66:1 15、97:124、97:132及び66:98、並びにこれらの保存置換からなる群から選ばれ た対中にH鎖及びL鎖免疫グロブリン可変領域アミノ酸残基配列を有するモノク ローナル抗体の結合特異性を有する請求の範囲第1項に記載のヒトモノクローナ ル抗体。 3.モノクローナル抗体がATCC 69078、ATCC 69079またはATCC 69080により産生 されたモノクローナル抗体の結合特異性を有する請求の範囲第1項に記載のヒト モノクローナル抗体。 4.モノクローナル抗体がATCC 69078、ATCC 69079またはATCC 69080により産生 されたモノクローナル抗体である請求の範囲第3項に記載のヒトモノクローナル 抗体。 5.ヒト免疫不全ウイルス(HIV)糖タンパク質gp120と免疫反応でき、かつHIV を中和できるヒトモノクローナル抗体であって、 そのモノクローナル抗体が配列番号95、96、97、98、101、102、103、104、10 5、107、110、115、118、121、122、124及び132、並びにこれらの保存置換から なる群から選ばれたL鎖免疫グロブリン可変領域アミノ酸残基配列を含むモノク ローナル抗体の結合特異性を有することを特徴とするヒトモノクローナル抗体。 6.ヒト免疫不全ウイルス(HIV)糖タンパク質gp120と免疫反応でき、かつHIV を中和できるヒトモノクローナル抗体のH鎖免疫グロブリン可変領域アミノ酸残 基配列部分をコードするポリヌクレオチド配列であって、 そのモノクローナル抗体が配列番号66、67、68、70、72、73、74、75、78及び 97からなる群から選ばれた前記H鎖免疫グロブリン可変領域アミノ酸残基配列、 及びそのアミノ酸残基配列の保存置換、並びにこれらに相補性のポリヌクレオチ ド配列を含むモノクローナル抗体の結合特異性を有することを特徴とするポリヌ クレオチド配列。 7.ポリヌクレオチドがDNAである請求の範囲第6項に記載のポリヌクレオチド 配列。 8.ヒト免疫不全ウイルス(HIV)糖タンパク質gp120と免疫反応でき、かつHIV を中和できるヒトモノクローナル抗体のL鎖免疫グロブリン可変領域アミノ酸残 基配列部分をコードするポリヌクレオチド配列であって、 そのモノクローナル抗体が配列番号95、96、97、98、101、102、103、104、10 5、107、110、115、118、121、122、124及び132からなる群から選ばれた前記L 鎖免疫グロブリン可変領域アミノ酸残基配列、及びそのアミノ酸残基配列の保存 置換、並びにこれらに相補性のポリヌクレオチド配列を含むモノクローナル抗体 の結合特異性を有することを特徴とするポリヌクレオチド配列。 9.ヒト免疫不全ウイルス(HIV)糖タンパク質gp41と免疫反応でき、かつHIVを 中和できるヒトモノクローナル抗体であって、 そのモノクローナル抗体が配列番号142、143、144、145及び146、並びにこれ らの保存置換からなる群から選ばれたH鎖免疫グロブリン可変領域アミノ酸残基 配列を含むモノクローナル抗体の結合特異性を有することを特徴とするヒトモノ クローナル抗体。 10.モノクローナル抗体が配列番号142:147、143:148、144:149、145:150、及び 146:151、並びにこれらの保存置換からなる群から選ばれた対中にH鎖及びL鎖 免疫グロブリン可変領域アミノ酸残基配列を有するモノクローナル抗体の結合特 異性を有する請求の範囲第9項に記載のヒトモノクローナル抗体。 11.ヒト免疫不全ウイルス(HIV)糖タンパク質gp41と免疫反応でき、かつHIVを 中和できるヒトモノクローナル抗体であって、 そのモノクローナル抗体が配列番号147、148、149、150、及び151、並び にこれらの保存置換からなる群から選ばれたL鎖免疫グロブリン可変領域アミノ 酸残基配列を含むモノクローナル抗体の結合特異性を有することを特徴とするヒ トモノクローナル抗体。 12.ヒト免疫不全ウイルス(HIV)糖タンパク質gp41と免疫反応でき、かつHIVを 中和できるヒトモノクローナル抗体のH鎖免疫グロブリン可変領域アミノ酸残基 配列部分をコードするポリヌクレオチド配列であって、 そのモノクローナル抗体が配列番号142、143、144、145、及び146からなる群 から選ばれた前記H鎖免疫グロブリン可変領域アミノ酸残基配列、及びそのアミ ノ酸残基配列の保存置換、並びにこれらに相補性のポリヌクレオチド配列を含む モノクローナル抗体の結合特異性を有することを特徴とするポリヌクレオチド配 列。 13.ポリヌクレオチドがDNAである請求の範囲第12項に記載のポリヌクレオチド 配列。 14.ヒト免疫不全ウイルス(HIV)糖タンパク質gp41と免疫反応でき、かつHIVを 中和できるヒトモノクローナル抗体のL鎖免疫グロブリン可変領域アミノ酸残基 配列部分をコードするポリヌクレオチド配列であって、 そのモノクローナル抗体が配列番号147、148、149、150、及び151からなる群 から選ばれた前記L鎖免疫グロブリン可変領域アミノ酸残基配列、及びそのアミ ノ酸残基配列の保存置換、並びにこれらに相補性のポリヌクレオチド配列を含む モノクローナル抗体の結合特異性を有することを特徴とするポリヌクレオチド配 列。 15.請求の範囲第6項、第8項、第12項または第14項に記載のポリヌクレオチド 配列を含むことを特徴とする宿主細胞。 16.請求の範囲第6項、第8項、第12項または第14項に記載のポリヌクレオチド 配列を含むことを特徴とするDNA発現ベクター。 17.ヒト免疫不全ウイルス(HIV)の検出方法であって、 HIVを含むと推測される試料を診断有効量の請求の範囲第1項、第5項、第9 項または第11項に記載のモノクローナル抗体と接触させ、モノクローナル抗体が 試料と免疫反応するか否かを測定することを特徴とするヒト免疫不全ウイルスの 検出方法。 18.検出が生体内である請求の範囲第17項に記載の方法。 19.モノクローナル抗体を放射性同位元素及び常磁性標識からなる群から選ばれ た標識で検出可能に標識する請求の範囲第18項に記載の方法。 20.検出が試験管内である請求の範囲第17項に記載の方法。 21.モノクローナル抗体を放射性同位元素、蛍光化合物、コロイド金属、ケミル ミネセンス化合物、バイオルミネセンス化合物、及び酵素からなる群から選ばれ た標識で検出可能に標識する請求の範囲第20項に記載の方法。 22.モノクローナル抗体を固相に結合する請求の範囲第20項に記載の方法。 23.ヒトのヒト免疫不全ウイルス(HIV)疾患の受動免疫療法であって、 免疫治療有効量の請求の範囲第1項、第5項、第9項または第11項に記載のモ ノクローナル抗体をヒトに投与することを特徴とするヒト免疫不全ウイルス疾患 の受動免疫療法。 24.受動免疫療法を予防のために施す請求の範囲第23項に記載の方法。 25.投与が非経口投与である請求の範囲第23項に記載の方法。 26.非経口投与が皮下、筋肉内、腹腔内、洞内、経皮、または静脈内の注射によ るものである請求の範囲第25項に記載の方法。 27.非経口投与が徐々の注入によるものである請求の範囲第25項に記載の方法。 28.徐々の注入が静脈内またはぜん動性の装置によるものである請求の範囲第27 項に記載の方法。 29.免疫治療有効量が約0.1mg/kgから約300mg/kgまでである請求の範囲第25項に 記載の方法。 30.ヒトのヒト免疫不全ウイルス(HIV)疾患に活性な免疫治療を誘導する方法 であって、 免疫原有効量の請求の範囲第1項、第5項、第9項または第11項に記載のモノ クローナル抗体に対する抗イディオタイプ抗体をヒトに投与することを特徴とす るヒト免疫不全ウイルス疾患の免疫治療の誘導方法。 31.薬理学的担体中に免疫治療有効量の請求の範囲第1項、第5項、第9項また は第11項に記載のモノクローナル抗体の少なくとも1回の投薬量を含むことを特 徴とする医薬組成物。 32.前記組成物が2種以上の異なるモノクローナル抗体を含む請求の範囲第31項 に記載の医薬組成物。 33.ヒト免疫不全ウイルス(HIV)を含むと推測される源中のHIVの検出に有益な キットであって、 そのキットが請求の範囲第1項、第5項、第9項または第11項に記載のモノク ローナル抗体を含む容器を含む一つ以上の容器を密閉してその中に収容するよう に区画されているキャリヤー装置を含むことを特徴とするHIVの検出に有益なキ ット。
