Claims (1)
- 【特許請求の範囲】 1、有効量の外膜小胞からなり、前記小胞は発現する相同性および/または異種 のまたは少なくとも1つの髄膜炎菌のクラスIの外膜タンパク質またはそのエピ トープを含有する断片またはオリゴペプチドから分離されたものである、髄膜炎 菌の病気に対して有効なワクチン。 2、髄膜炎菌のクラスIの外膜タンパク質は、クラス2/3の外膜タンパク質に 対して陰性である突然変異の髄膜炎菌株から由来する、上記第1項記載のワクチ ン。 3、クラスIの外膜タンパク質、断片またはオリゴペプチドは、前記クラスIの 外膜タンパク質、断片またはオリゴペプチドをエンコードする異種遺伝子を含有 する微生物により産生される、上記第1項記載のワクチン。 4、クラスIの外膜タンパク質、断片またはオリゴペプチドは、群A、B、C、 W−135またはYの髄膜炎菌から誘導される、上記第1項記載のワクチン。 5、クラスIの外膜タンパク質、断片またはオリゴペプチドは、群Bの髄膜炎菌 から誘導される、上記第1項記載のワクチン。 6、5〜10の種々の群の髄膜炎菌のクラスIの外膜タンパク質、断片またはオ リゴペプチドからなる、上記第1項記載のワクチン。 7、タンパク質断片はクラスIの外膜タンパク質の臭化シアン処理により得られ る、上記第1項記載のワクチン。 8、クラスIの髄膜炎菌の外膜タンパク質は、エンドLys−C、エンドArg −C、エンドGlu−Cおよびブドウ球菌属(Staphylococcus) V8−プロテアーゼから成る群より選択される酵素を使用するタンパク質分解に より得られる、上記第1項記載のワクチン。 9、オリゴペプチドは髄膜炎菌のクラスIの外膜タンパク質のエピトープと結合 する少なくとも1つの抗体からなる、上記第1項記載のワクチン。 10、オリゴペプチドは、【配列があります】、【配列があります】、【配列が あります】、【配列があります】、【配列があります】、【配列があります】、 【配列があります】、【配列があります】、および【配列があります】から成る 群より選択される少なくとも1つのアミノ酸配列からなる、上記第1項記載のワ クチン。 11、エピトープは髄膜炎菌のクラスIの外膜タンパク質の表面ループのアミノ 酸24−34および176−187の区域に位置する、上記第1項記載のワクチ ン。 12、クラスIの外膜タンパク質、断片またはオリゴペプチドは、T細胞のエピ トープに化学的カップリングを経て接合されている、上記第1項記載のワクチン 。 13、クラスIの外膜タンパク質、断片またはオリゴペプチドは、タンパク質産 生物との接合体の形態のA、B、C、W−135および/またはまたはYの多糖 類を含有する、上記第1項記載のワクチン。 14、両性イオン、カチオン、アニオンおよび/または非イオンを発生する洗浄 剤をさらに含む、上記第1項記載のワクチン。 15、洗浄剤は、ツビッタージェント(Zwittergent)Zw3−10 、ツビッタージェント(Zwittergent)Zw3−±4、ツイーン(T ween)−20、塩素酸ナトリウム、オクチルグリコシドおよびデオキシ塩素 酸ナトリウムから成る群より選択される、上記第1項記載のワクチン。 16、リン酸アルミニウム、水酸化アルミニウムおよびリン酸カルシウムから成 る群より選択される吸着剤をさらに含む、上記第1項記載のワクチン。 17、免疫刺激複合体(ISCOM)をさらに含む、上記第1項記載のワクチン 。 18、クラスIの外膜タンパク質、断片またはオリゴペプチドはリポソーム内に 含有されている、上記第1項記載のワクチン。 19、クラスIの外膜タンパク質、断片またはオリゴペプチドは脂質にカップリ ングされている、上記第1項記載のワクチン。 20、約25kD以下の分子量を有し、そして髄膜炎菌(Neisseria meningitidis)に対するバクテリアの抗体と反応性の連続または不 連続のエピトープを含有する、髄膜炎菌(Neisseria meningi tidis)のクラスIの外膜タンパク質の実質的な精製された断片。 21、クラスIの外膜タンパク質はサブタイプP1.7.16である、上記第2 0項記載の断片。 22、髄膜炎菌(N.meningitidis)のクラスIの外膜タンパク質 の臭化シアンの切断により産生され、ほぼ25kDの分子量を有する、上記第2 1項記載の断片。 23、異なる髄膜炎菌のポリン(porin)タンパク質の間のアミノ酸配列か 変化する、クラスIの外膜タンパク質のB細胞のエピトープを含有するオリゴペ プチド。 24、【配列があります】、【配列があります】、【配列があります】、【配列 があります】、【配列があります】、【配列があります】、【配列があります】 、【配列があります】および/または【配列があります】から成る群より選択さ れる少なくとも1つのアミノ酸配列を含有する、上記第23項記載のオリゴペプ チド。 25、異なる髄膜炎菌のポリン調製の間のアミノ酸配列の中に保存された、クラ スIの外膜タンパク質のエピトープを含有するオリゴペプチド。 26、髄膜炎菌のクラスIの外膜タンパク質のT細胞のエピトープを含有するオ リゴペプチド。 27、髄膜炎菌のクラスIの外膜タンパク質、そのエピトープを含有する断片ま たはオリゴヌクレオチドをエンコードする、分離された核酸。 28、製剤学的に許容されうる賦形剤中の1または2以上の髄膜炎菌のクラスI の外膜タンパク質またはその断片および、必要に応じて、アジュバントからなる ワクチン組成物を投与することからなる、髄膜炎菌(Neisseria me Hingitidis)に対する保護の免疫応答を引き出す方法。 29、断片は、T細胞のエピトープ、B細胞のエピトープまたはキャリヤーのペ プチドまたはタンパク質に接合されているか、あるいはそれに遺伝子的に融合さ れている、上記第28項記載の方法。 30、接合は断片の末端にシステインまたはリジンを通してカップリングされて いる、上記第29項記載の方法。 31、キャリヤーはバクテリアの毒素、CRMまたはトキソイドである、上記第 29項記載の方法。 32、クラスIの外膜タンパク質はサブタイプのP1.7.16である、上記第 28項記載の方法。 33、製剤学的に許容されうる賦形剤中の髄膜炎菌(Neisseria me ningitidis)に対するバクテリアの抗体と反応性のクラス1の外膜タ ンパク質の連続または不連続のエピトープを含有する少なくとも1つのオリゴペ プチドおよび、必要に応じて、アジュバントからなるワクチン組成物を投与する ことからなる、髄膜炎菌(Neisseria meningitidis)に 対する保護の免疫応答を引き出す方法。 34、オリゴペプチドは【配列があります】、【配列があります】、【配列があ ります】、【配列があります】、【配列があります】、【配列があります】、【 配列があります】、【配列があります】および/または【配列があります】から 成る群より選択されるアミノ酸配列を含有する、上記第33項記載の方法。 35、タンパク質分解の断片は、T細胞のエピトープ、B細胞のエピトープまた はキャリヤーのペプチドまたはタンパク質に接合されているか、あるいはそれに 遺伝子的に融合されている、上記第33項記載の方法。 36、接合は断片の末端にシステインまたはリジンを通してカップリングされて いる、上記第33項記載の方法。 37、キャリヤーはバクテリアの毒素、CRMまたはトキソイドである、上記第 35項記載の方法。 38、キャリヤーのタンパク質またはそのエピトープに接合された髄膜炎菌のク ラスIの外膜タンパク質、そのエピトープを含有する断片またはオリゴペプチド からなる、抗原の接合体。 39、オリゴペプチドは、【配列があります】、【配列があります】、【配列が あります】、【配列があります】、【配列があります】、【配列があります】、 【配列があります】、【配列があります】および/または【配列があります】か ら成る群より選択される、上記第8項記載の抗原の接合体。 40、抗原のキャリヤーはバクテリアの毒素、CRMまたはそのエピトープであ る、上記第39項記載の抗原の接合体。 41、キャリヤーのタンパク質はCRM197である、上記第40項記載の抗原 の接合体。 42、キャリヤーのタンパク質、そのペプチドまたはエピトープに融合した髄膜 炎菌のクラスIの外膜タンパク質のエピトープからなる、遺伝子の融合ペプチド またはタンパク質。 43、エピトープは【配列があります】、【配列があります】、【配列がありま す】、【配列があります】、【配列があります】、【配列があります】、【配列 があります】、【配列があります】および/または【配列があります】から成る 群より選択される、上記第42項記載の遺伝子の融合ペプチドまたはタンパク質 。 44、その内部に挿入された髄膜炎菌のクラスIの外膜タンパク質のエピトープ のためのアミノ酸配列を有するフラジェリンタンパク質からなる、融合タンパク 質。 45、アミノ酸配列は、フラジェリンタンパク質内に、フラジェリンタンパク質 の機能に対して非必須である領域において挿入されている、上記第44項記載の 融合タンパク質。 46、アミノ酸配列はフラジェリンタンパク質の超可変領域の中に挿入されてい る、上記第45項記載の融合タンパク質。 47、それに接合した髄膜炎菌の莢膜のオリゴサッカリドまたは多糖類をさらに 含む、上記第44項記載の融合タンパク質。 48、上記第44項記載の融合タンパク質をエンコードする、組み換え遺伝子。 49、髄膜炎菌(Neisseria meningitidis)のクラスI の外膜タンパク質、または髄膜炎菌(NeisseriamenlngItid is)に対するバクテリアの抗体と反応性の連続または不連続のエピトープを含 有する断片またはオリゴペプチドを発現することができる、感染性組み換え微生 物。 50、オリゴペプチドは【配列があります】、【配列があります】、【配列があ ります】、【配列があります】、【配列があります】、【配列があります】、【 配列があります】、【配列があります】および/または【配列があります】から 成る群より選択される、上記第49項記載の微生物。 51、ワクシニアのウィルス、アデノウィルス、またはサイトメガロウィルスで ある、上記第49項記載の微生物。 52、サルモネラ層(Salmonella)のバクテリアである、上記第49 項記載の微生物。 53、髄膜炎菌のクラスIの外膜タンパク質のエピトープは組み換えフラジェリ ンとして発現される、上記第49項記載の微生物。 54、上記第49項記載の微生物を個体に接種することからなる、髄膜炎菌(N eisseria meningitidis)に対して免疫化する方法。 55、クラス2および3の外膜タンパク質を産生することができない突然変異の 髄膜炎菌株。 56、突然変異の髄膜炎菌株HII15、CBS636.89。 57、非天然のクラスIの外膜タンパク質をエンコードする異種遺伝子で形質転 換された、クラス2および3の外膜タンパク質を産生することができない、突然 変異の髄膜炎菌株。 58、a、ポリンタンパク質のアミノ酸配列を獲得し、b、ポリンタンパク質の アミノ酸配列を髄膜炎菌のクラスIの外膜タンパク質(OMP)のアミノ酸配列 と比較して、クラスIのOMP中の既知の対応する構造に基づいて、ポリンタン パク質の露出した表面ループの構造の位置、大きさおよび配列を予測し、そして c、予測された構造の位置、大きさおよび配列に基づいて表面ループ構造中の潜 在的ワクチンのエピトープを同定する、からなる、グラム陰性のポリンタンパク 質のワクチンのエピトープを同定する方法。
