Description
また、特許文献2の吸着方法では、補酵素Q10自体を吸着剤に吸着することを目的としており、補酵素Q10を得るためには、吸着処理後、さらに脱着用の溶媒を用いて吸着剤より補酵素Qを脱着溶出させる工程が必要となる。
加えて、特許文献3では、不純物除去を目的に吸着剤を使用しているが、その分の原料費アップや、処理後の吸着剤を廃棄した場合に廃棄物量が増加すること、あるいは吸着剤を再生させる設備やエネルギーの問題など、解決すべき課題が多い。
一方、特許文献4の方法は、補酵素Q10生産微生物が脂溶性の不純物量の少ない光合成細菌に限定されており、その場合においても、純度の高い補酵素Q10を得るためにはその収率を犠牲にしなければいけないなどの問題がある。
すなわち本発明に係る補酵素Q10の製造方法の構成は以下のとおりである。
1.重量基準で水分含量50ppm以上1%以下の補酵素Q10生産微生物の疎水性有機溶媒抽出液またはその濃縮抽出液を冷却する冷却工程、および
析出した固形分を分離除去する分離工程を有する補酵素Q10の製造方法。
2.補酵素Q10生産微生物の疎水性有機溶媒抽出液を、アルカリ水溶液と接触混合した後に、水洗し、それを濃縮した濃縮抽出液を冷却工程に供する上記1に記載の製造方法。
3.分離工程で得られた前記固形分を、前記アルカリ水溶液と接触混合させる前の抽出液に添加した後、アルカリ水溶液と接触混合し、水洗し、冷却工程に供することを繰り返すか、または
分離工程で得られた前記固形分を、前記アルカリ水溶液と接触混合した後の抽出液に添加した後、水洗し、冷却工程に供することを繰り返す上記2に記載の製造方法。
4.冷却工程における前記冷却温度が、20℃以下である上記1~3のいずれかに記載の製造方法。
5.冷却工程における前記冷却時の抽出液又は濃縮抽出液中の補酵素Q10の濃度が0.1g/L以上300g/L以下である上記1~4のいずれかに記載の製造方法。
6.前記疎水性有機溶媒が、炭化水素及び/又は脂肪酸エステルである上記1~5のいずれかに記載の製造方法。
7.分離工程における前記固形分の分離除去方法が、回転式フィルターを用いるものである上記1~6のいずれかに記載の製造方法。
補酵素Q10には、酸化型と還元型が存在する。本発明は、補酵素Q10として、酸化型補酵素Q10、還元型補酵素Q10のいずれをも対象とし、還元型補酵素Q10と酸化型補酵素Q10の混合物である補酵素Q10もその対象である。本発明で用いる補酵素Q10が還元型補酵素Q10と酸化型補酵素Q10の混合物である場合の還元型補酵素Q10含有比率も特に限定されない。なお、本明細書において、補酵素Q10とのみ記載した場合はその種類を問わず、酸化型補酵素Q10、還元型補酵素Q10、還元型補酵素Q10と酸化型補酵素Q10の混合物の全てを表すものである。
これらのうち培養の容易さや生産性の観点からは、細菌または酵母が好ましい。細菌では非光合成細菌がより好ましく、さらには、アグロバクテリウム(Agrobacterium)属、グルコノバクター(Gluconobacter)属等が特に好ましい例として挙げられる。また酵母ではシゾサッカロミセス(Schizosaccharomyces)属、サイトエラ(Saitoella)属、ファフィア(Phaffia)属等が特に好ましい例として挙げられる。
なお、補酵素Q10として、還元型補酵素Q10を製造する目的においては、生産される補酵素Q10中の還元型補酵素Q10含有比率の高い微生物を用いることが好ましく、例えば培養後の補酵素Q10に占める還元型補酵素Q10含有比率(重量%基準)として好ましくは70%以上、より好ましくは80%以上となる微生物を用いることがより好ましい。
上記補酵素Q10生産微生物から補酵素Q10を、有機溶媒を用いて抽出する。詳細には本発明では、後述する冷却工程において、冷却時に使用される溶媒(冷却時の抽出液または濃縮抽出液の溶媒)としては疎水性の有機溶媒を使用する必要があるが、補酵素Q10生産微生物から補酵素Q10の抽出(微生物由来成分の抽出)に用いる有機溶媒としては特に限定されず、疎水性、親水性いずれをも利用できる。抽出時に疎水性有機溶媒を使用することで、抽出液やその濃縮液をそのまま冷却工程に供することが出来るため好ましい。
また本発明の製造方法において、上記抽出時の温度は、特に制限されないが、通常0~60℃、好ましくは20~50℃の範囲で実施できる。
また、本発明の製造方法においては、必要に応じて上記補酵素Q10生産微生物の抽出液を濃縮した濃縮抽出液を用いてもよい。例えば、抽出時には有機溶媒を多く使用して抽出操作の安定性や抽出率を高め、冷却工程前に適宜濃縮することで、固形分析出に適した濃度に調整することも好ましい方法の一つである。この場合の濃縮方法は特に限定されず、蒸発濃縮、膜濃縮、凍結濃縮、減圧濃縮、超音波霧化分離などが挙げられ、これらを組み合わせて濃縮してもよい。また濃縮の程度も特に限定されないが、不純物を固形分として必要量析出させる観点から、前述した抽出液と同様、濃縮後の濃縮抽出液中の補酵素Q10濃度は、好ましくは0.1g/L以上、より好ましくは1g/L以上、さらに好ましくは10g/L、更により好ましくは20g/Lである。上限も特に限定されないが、補酵素Q10の損失を抑える観点から、300g/L程度が好ましく、150g/L以下がより好ましく、100g/L以下がより好ましい。もちろん、得られた補酵素Q10生産微生物の疎水性有機溶媒抽出液が上記好ましい範囲を満たす場合は,濃縮は必ずしも必要ではない。
さらに本発明の製造方法においては、上記(2)により得られた抽出液を冷却工程に供する前に、アルカリ処理を濃縮と組み合わせて行うことが好ましい。すなわち、補酵素Q10生産微生物を疎水性有機溶媒で抽出して得られた抽出液を、アルカリ水溶液と接触混合した後に、水洗し、それを濃縮した濃縮抽出液を、冷却工程に供するのが、本発明の好ましい態様である。上記抽出液をアルカリ処理することで微生物由来の脂肪酸、脂肪酸エステル、リン脂質などの脂溶性成分がけん化されて水相に移行するため、不純物の除去効率がさらに向上し、後段の工程への負荷を低減させることができる。
上記のようにして得られる補酵素Q10生産微生物の抽出液または濃縮抽出液は、特に微生物の培養液、湿微生物細胞、微生物細胞の水性懸濁液やその破砕物から抽出した場合、それらに由来する水分を含んでいる。本発明の製造方法においては、抽出溶媒として疎水性有機溶媒を利用した場合や、若干の親水性有機溶媒を疎水性有機溶媒と併用して用いた場合、疎水性有機溶媒抽出液または濃縮抽出液中の水分含量が重量基準で50ppm以上1%以下の場合は、当該疎水性有機溶媒抽出液または濃縮抽出液をそのまま、次の冷却工程に供することが出来る。また、水分含量が上記範囲を満たさない場合は、適宜加水あるいは脱水して冷却工程に供すればよい。一方、抽出溶媒として親水性の有機溶媒を使用した場合には、冷却工程の前に、疎水性有機溶媒に置換し、その後必要に応じて水分量を調整し、冷却を実施すれば良い。
なお、本明細書においては、補酵素Q10生産微生物の任意の抽出液を別の疎水性有機溶媒に溶媒置換したものも、便宜上「補酵素Q10生産微生物の疎水性有機溶媒抽出液」と表現する。また、本発明においては、上記補酵素Q10生産微生物の疎水性有機溶媒抽出液から、別の方法で不純物をある程度除去したものを、必要に応じて水分含量を調整して用いることも出来る。
本発明の製造方法においては、上記のようにして得られた補酵素Q10生産微生物の疎水性有機溶媒抽出液あるいはその濃縮抽出液を、冷却することで補酵素Q10以外の微生物由来の不純物を固形分として析出させる。この冷却工程において、冷却の対象となる抽出液または濃縮抽出液は前述したとおり、疎水性有機溶媒を溶媒とするものであり、その水分含量は重量%として50ppm以上1%以下の範囲内に制御されている必要がある。上記水分含量が50ppm未満の場合、冷却工程における補酵素Q10以外の成分の除去率が低くなるなどの問題がある。一方、上記水分含量が1%超の場合、冷却工程後も溶液中に水分が多く含まれるため、その後のカラムクロマトグラフィーなどの後段の工程に悪影響を及ぼす可能性がある。上記水分含量の上限は0.4%以下が好ましく、0.3%以下がより好ましく、0.2%以下がさらに好ましい。
本発明の製造方法においては、冷却したときに固形分をより多く析出させることで、ろ液中の補酵素Q10の純度を向上させることができる。析出させる固形分濃度としては特に限定されないが、例えば1g/L以上、好ましくは1.5g/L以上、より好ましくは2g/L以上である。
上記のようにして冷却した後、析出した固形分を分離除去する。本発明の製造方法において、析出した固形分を分離除去するために用いられる方法は、固形分の分離除去が可能であれば特に限定されず、例えば、濾紙、濾布、円筒状フィルターなどを用いた一般的な濾過方法の他、自然沈降分離、遠心分離、膜分離、振動式膜分離、液体サイクロン、回転式フィルター、吸着分離などの方法あるいはこれらの分離方法の組み合わせを利用することが出来る。
固形分の分離除去性能および所要動力、分離除去設備のコンパクト化、並びにオペレーションの負荷を加味して効果的に補酵素Q10を工業的に製造するためには、回転式フィルターを用いることが好ましい。回転式フィルターの材質としては特に限定されないが、例えば、ポリエチレン、ポリプロピレン、ポリメチルメタアクリレート、ポリスチレン、フッ素樹脂などの合成樹脂やその複合物;アルミナ、ジルコニア、チタン酸バリウム、酸化チタンやこれらの複合物などの酸化物系、ハイドロキシアパタイトなどの水酸化物系、炭化ケイ素などの炭化物系、窒化ケイ素などの窒化物系、蛍石などのハロゲン化物系、リン酸塩系などのセラミック;鉄、銅、亜鉛、スズ、水銀、鉛、アルミニウム、ステンレスやこれらの複合物などの金属が挙げられる。また、回転式フィルターの孔径も特に限定されないが、目的とする固形分と分離するためには1nm~2μmが好ましく、処理量や洗浄のしやすさを考慮すれば60nm~1μmが好ましい。上記回転式フィルターの具体例としては、広島メタル&マシナリー社製やユーロテック社製の「セラミックロータリーフィルター」や三菱化工機社製の「三菱ダイナフィルター」が挙げられる。
前段工程に戻す際には、完全な固液分離を実施せず、析出した固形分をスラリー状態として抽出液又は濃縮抽出液の一部と一緒に戻しても良い。

水分含量は、カールフィッシャー(AQUACOUNTER AQ-2100 HIRANUMA社製)を用いて測定した。
実施例1と同じ補酵素Q10生産微生物の濃縮抽出液(水分含量663.4ppm)を、40℃の環境に2時間設置し、実施例1と同じようにして吸引ろ過にて固形分を分離除去した。ろ液を分析した結果、補酵素Q10の純度は3.2%pt.向上したものの、ろ液中の水分含量は334.7ppm、エルゴステロール除去率は2.9%であることが確認された。
実施例5と同じ補酵素Q10生産微生物の濃縮抽出液(水分含量585ppm)を、冷却することなく30℃の条件下で、実施例5と同様の遠心分離機を用いて9000gで5分間遠心分離し、上清を回収した。回収した上清中の水分含量は309.5ppmと低減していることが確認された。
実施例1と同様の方法で補酵素Q10生産微生物の濃縮抽出液(550.5ppm)を調整し、それに、該溶液に対して3.5重量%の活性白土、4重量%のろ過助剤(ロカヘルプ:三井金属鉱業社製)を添加し、40℃にて撹拌し、吸引ろ過にて固形分を分離除去した。ろ過操作には桐山ロートおよび桐山ロート用のろ紙No.5-Cを用いた。その結果、濃縮抽出液中の水分含量が550.5ppmだったのに対し、ろ液中の水分含量は402.5ppm、補酵素Q10の収率は97.8%、エルゴステロール除去率は8.8%、補酵素Q10の純度向上パーセントは0.5%pt.であることが確認された。
比較例3と同じ補酵素Q10生産微生物の濃縮抽出液(水分含量550.5ppm)に、該溶液に対して3.5重量%の活性白土、4重量%のろ過助剤(ロカヘルプ:三井金属鉱業社製)を添加し、40℃にて撹拌後、18℃まで冷却した後で、比較例3と同様の方法で固形分を分離除去した。その結果、ろ液中の水分含量は220.5ppm、エルゴステロール除去率は62.4%、補酵素Q10の純度は3.5%pt.向上していることが確認されたが、補酵素Q10の収率は98.4%だった。
Claims (6)
- 補酵素Q10生産微生物の疎水性有機溶媒抽出液を、アルカリ水溶液と接触混合した後に、水洗し、それを濃縮した、重量基準で水分含量50ppm以上1%以下の濃縮抽出液を冷却する冷却工程、および
析出したステロール誘導体を含む不純物の固形分を分離除去し、補酵素Q10を含むろ液を得る分離工程を有する補酵素Q10の製造方法。 - 分離工程で得られた前記固形分を、前記アルカリ水溶液と接触混合させる前の抽出液に添加した後、アルカリ水溶液と接触混合し、水洗し、冷却工程に供することを繰り返すか、または
分離工程で得られた前記固形分を、前記アルカリ水溶液と接触混合した後の抽出液に添加した後、水洗し、冷却工程に供することを繰り返す請求項1に記載の製造方法。 - 冷却工程における前記冷却温度が、20℃以下である請求項1または2に記載の製造方法。
- 冷却工程における濃縮抽出液中の補酵素Q10の濃度が0.1g/L以上300g/L以下である請求項1~3のいずれか1項に記載の製造方法。
- 前記疎水性有機溶媒が、炭化水素及び/又は脂肪酸エステルである請求項1~4のいずれか1項に記載の製造方法。
- 分離工程における前記固形分の分離除去方法が、回転式フィルターを用いるものである請求項1~5のいずれか1項に記載の製造方法。
