Description
膜は、液体中の構成成分を濾過または分画するための分離プロセスまたはシステムで使用される。様々な技術において、食品および飲料産業で使用されるこれらの膜を含む膜が利用されている。
内生胞子は、フィルミクテス門内の特定の種の細菌によって生成される休眠中の丈夫な非生殖構造である。内生胞子、または胞子は、生育状態の細菌細胞がストレスまたは栄養素の欠如にさらされるときに生成される。内生胞子の代謝率は非常に低く、したがって、生育細菌細胞を迅速に検出するために通常採用されている方法では検出することができない。さらに、胞子は、紫外線、熱、殺菌剤、乾燥、飢餓などの過酷な条件で生き延びるように設計されているため、死滅させることは非常に困難である。好ましい条件および栄養素にさらされると、胞子は発芽して栄養細胞を生成する。
本開示は、膜の性能または完全性に重大な悪影響を与えることなく、細菌胞子およびバイオフィルムを膜から相乗的に除去および低減する二部式洗浄システムの使用について記載する。いくつかの態様では、洗浄システムはまた、膜のスケーリングに役立つ。2部式洗浄システムは、酸洗浄溶液および酸化剤消毒液を利用する。システムの2つの部分は、同時にまたは逐次的に膜に適用することができる。
いくつかの態様では、陰イオン界面活性剤は、酸洗浄溶液中に含まれている。界面活性剤は、膜表面上の洗浄液の表面活性を改善する。いくつかの実施形態では、陰イオン界面活性剤はまた、膜システム上の酸洗浄システムの腐食を防止または低減するのを助けることができる。陰イオン界面活性剤は、疎水性物質に負電荷を有する表面活性物質であるか、または、pHが中性以上に上昇しない限り電荷を運ばない疎水性区域を有する(例えばカルボン酸)。カルボキシレート、スルホネート、サルフェートおよびホスフェートは、陰イオン界面活性剤中に見出される極性(親水性)可溶性基である。酸洗浄溶液中で有用な陰イオン界面活性剤としては、アルキルサルフェート、アルキルスルホネート、アルカンスルホネート、直鎖および分岐鎖の第一級および第二級アルキルスルホネート、置換基有りまたは無しの芳香族スルホネート、直鎖アルキルベンゼンまたはナフタレンスルホネート、第二級アルカンスルホネート、アルキルエーテルサルフェートまたはスルホネート、アルキルホスフェートまたはホスホネート、およびそれらの混合物が挙げられる。具体例としては、アルカンスルホン酸ナトリウム、αオレフィンスルホン酸、リグノスルホン酸ナトリウム、ラウリルエーテル硫酸ナトリウム、ラウリル硫酸ナトリウム、ジオクチルスルホコハク酸ナトリウム、ドデシルベンゼンスルホン酸ナトリウム、キシレンスルホン酸ナトリウム、DOWFAX C6L、3B2、8390、および2A1を含むDOWFAXの商標名にて販売されているスルホン化ジフェニルオキシド界面活性剤、カプリレス-9カルボン酸/ヘキサス-4カルボン酸(AKYPO LF4など)、メチル2-スルホラウリン酸ナトリウム(ALPHASTEP PC48など)、サルコシネート、およびそれらの混合物が挙げられる。
酸洗浄溶液は、少なくとも1種の有機酸を含む。例示的な実施形態では、酸洗浄溶液は、少なくとも2種の有機酸を含む。好適な有機酸としては、メタンスルホン酸、エタンスルホン酸、プロパンスルホン酸、ブタンスルホン酸、キシレンスルホン酸、ベンゼンスルホン酸、ギ酸、酢酸、ピコリン酸、ジピコリン酸、クエン酸、乳酸、ギ酸、酢酸、プロピオン酸、酪酸、吉草酸、カプロン酸、グルコン酸、イタコン酸、トリクロロ酢酸、安息香酸、およびそれらの混合物が挙げられる。好ましくは、有機酸は、クエン酸、乳酸、ギ酸、およびメチルスルホン酸から選択される。いくつかの実施形態では、有機酸は、クエン酸と乳酸との組み合わせを含む。いくつかの実施形態では、有機酸は、ギ酸とメチルスルホン酸との組み合わせを含む。
酸化剤消毒液は、酸化剤を含む。いくつかの実施形態では、酸化剤は、過酸(ペルオキシカルボン酸)である。いくつかの実施形態では、酸化剤は、酸化剤と、カルボン酸と、ペルオキシカルボン酸を生成する安定剤との組み合わせである。
安定剤
一態様では、酸洗浄溶液と酸化剤消毒液の組み合わせを使用して、バイオフィルム形成および細菌胞子による汚染を受けやすい膜を相乗的に洗浄および消毒する。いくつかの実施形態では、酸洗浄溶液は、最初に膜に適用されて、膜を洗浄する。次いで、酸化剤消毒液を酸洗浄溶液と組み合わせて、強化抗菌溶液を生成する。次いで、この強化溶液は、膜内で循環させて、膜を消毒する。膜システムの透過側に洗浄の問題がある場合、膜システムの圧力を修正して、この組み合わせの洗浄液の透過率を増減することができる。
最初に、バチルス種の野生分離株の生存を低減するための抗菌溶液の性能を比較するためにイン-ビトロ試験(in-vitro test)を行った。
次に、実験配合物を試験し、膜からの胞子除去の有効性について比較した。最初に、胞子分離株を滅菌蒸留水で希釈して、バチルス胞子野生分離株の混合物の最終濃度が104CFU/mLである溶液を生成した。6枚の限外濾過(UF)ポリエーテルスルホン(PES)膜(Spirapro,KOCH,Wilmington,MA)を104CFU/mLの胞子溶液に24時間液浸した。膜を浸漬法を使用してすすぎ、個々の滅菌遠心管の中に位置付けた。AC-AとO-Aとの組み合わせ処理を、2500ppmで管内の膜のうちの2つに5分間適用した。比較として、CS-Aを2600ppmで膜のうちの2つに5分間適用した。脱イオン水(DI)を2つの膜の対照として使用した。いくつかの膜を防腐剤溶液中で湿潤状態で保存し、他の膜を乾燥状態で試験した。
例示的な配合物の抗菌性能を、バチルス種の胞子に対して試験した。サンプルを、122°F(50℃)で2、5、または10分間、例示的な配合物にさらした。実施例1では、上記の懸濁法を利用した。サンプルを観察して、生物のlog低減を達成するのに必要な時間を決定した。
また、実験式を、図4に示す疾病管理センター(Centers for Disease Control、CDC)のバイオフィルムリアクタ(BioSurface Technologies Corporation,Bozeman,MT)を使用して、バイオフィルムをPES UF膜表面から除去する際の有効性について試験した。この試験方法は、US EPA Standard Operating Procedure for Single Tube Method for Measuring Disinfectant Efficacy Against Biofilm Grown in the CDC Biofilm Reactorに基づいていた。
材料適合性試験を実施して、膜表面の劣化の指標である膜阻止率への影響を観察した。試験は、PES UF膜材料およびRO膜の両方で実行された。サンプルを5.7日間液浸した。1日あたり15分のCIP接触に基づいて、これは化学物質へ1.5年毎日さらしたことに相当する。
化学組成物の抗菌性および適合性の利点に加えて、試験は、DDBSA、クエン酸、および乳酸(AC-A)からなる化学組成物が、バチルス種の胞子分離株がPES UF膜表面上に付着または増殖し、RO膜表面の程度を低くする能力を妨害することを示した。この研究の目的は、膜および胞子をDI水対AC-A1%溶液中に液浸したとき、ROおよびUF(PES)膜表面とのバチルス種の中温性胞子野生分離株の付着特性を決定することであった。各膜ディスクについての液浸条件の記載を、表9に提供する。膜9~12は、対照として働く。
有効性試験を、バチルス種胞子の野外分離株に対して行った。試験方法としてSOP#MS009;Germicidal and Detergent Sanitizing Action of Disinfectantsを使用して、いくつかの異なる温度(70°F(約21℃)、110°F(約43℃)、および122°F(50℃))で5分間の接触時間で試験を行った。実施例1では、上記の懸濁法を利用した。酸洗浄剤B(AC-B)とO-Aとの新しい組み合わせ処理を、AC-A、AC-B、およびO-Aの個々に、ならびにAC-AとO-Aとの組み合わせに対して比較した。
AC-Bを含む例示的な配合を、膜から細菌胞子を除去する際の性能について試験した。O-Aの有効性を、O-AとAC-AおよびO-AとAC-Bの組み合わせで比較した。
AC-AとO-AおよびAC-BとO-Aの例示的な溶液の組み合わせを、カルシウムを分解するこれらの能力について比較した。結果を図15に示す。

Claims (18)
- 膜システム内の膜部材を洗浄および消毒する方法であって、前記方法は:
洗浄液を、前記膜システムを通して、約70°F~約125°F(約21℃~約52℃)の温度で約2分~約30分間循環させることであって、前記洗浄液が、約0.1重量%~約1重量%の有機酸および約0.01重量%~約0.1重量%の陰イオン界面活性剤を含み、前記有機酸は、メチルスルホン酸、ギ酸、クエン酸及び乳酸から選択される、少なくとも2種の有機酸の組み合わせを含む、ことと、
消毒液を前記洗浄液に添加して強化抗菌溶液を生成することであって、前記消毒液が、約0.02重量%~約0.15重量%の酸化剤としての過酸化水素と、約0.001重量%~約0.03重量%のカルボン酸と、約0.0005重量%~約0.01重量%の安定剤としてのホスホネートキレート剤と、約0.0001重量%~約0.05重量%の過カルボン酸と、を含む、ことと、
前記強化抗菌溶液を、前記膜システムを通して、約70°F~約125°F(約21℃~約52℃)の温度でさらに約1~約20分間循環させることと、を含む方法。 - 前記有機酸がメチルスルホン酸とギ酸との組み合わせを含む、請求項1に記載の方法。
- 前記有機酸がクエン酸と乳酸との組み合わせを含む、請求項1又は2に記載の方法。

- 前記方法が、バイオフィルム、生物付着物および/または粘液形成細菌の少なくとも3logの低減をもたらす、請求項1~11のいずれか一項に記載の方法。
- 有機酸、陰イオン界面活性剤および過カルボン酸の組み合わせが、過カルボン酸のみと比較して、前記膜との改善された化学的適合性をもたらし、改善された化学的適合性が、UF膜のタンパク質阻止および/またはRO膜の塩阻止によって示される、請求項1~12のいずれか一項に記載の方法。
- 前記膜システムが、乳製品工場、醸造所、ワイナリー、水工場または食品工場における膜濾過システムである、請求項1~13のいずれか一項に記載の方法。
- 前記方法が定置洗浄法である、請求項1~14のいずれか一項に記載の方法。
